    目的 基于 JAK2 / STAT3 信号通路研究黄芩联合 5-氟尿嘧啶(5-fluorouracil, 5-Fu)对 Lewis 荷瘤模型小鼠肺癌的抑制作用。 方法 检测黄芩(20 mg / kg)联合 5-Fu 干预 Lewis 荷瘤小鼠后的体质量、进食量和瘤体积变化,通过苏木素-伊红(HE)染色观察瘤组织病理变化,通过免疫组织化学染色观察增殖相关蛋白Ki67 表达水平变化,通过 TUNEL 染色观察瘤组织中细胞凋亡水平变化,通过免疫组织化学染色观察瘤组织中对 JAK2、p-JAK2、STAT3 和 p-STAT3 蛋白表达水平的变化。 结果 与模型组相比,联合干预后小鼠瘤体积明显变小,具有统计学意义(P<0. 01),体质量升高(P<0. 05),进食未发生明显变化。 瘤组织中增殖相关蛋白 Ki67 表达明显降低,TUNEL 标记的凋亡细胞明显增多;p-JAK2、p-STAT3 蛋白表达升高。 结论 黄芩水煎液可抑制 JAK2 / STAT3 信号通路,并增加 5-Fu 对小鼠肺癌的抑制作用。

    Abstract:

    Objective To investigate the inhibitory effect of Scutellaria baicalensis combined with 5-Fu on Lewis tumor bearing mouse lung cancer based on the JAK2 / STAT3 signaling pathway. Methods To detect the changes in body mass, food intake and tumor volume of Lewis tumor-bearing mice after intervention of Scutellaria baicalensis (20 mg / kg) combined with 5-Fu. Pathological changes in tumor tissue were observed by HE staining, expression levels of proliferation related protein Ki67 were observed by immunohistochemical staining, changes in cell apoptosis levels in tumor tissue were observed by TUNEL staining, and changes in expression levels of JAK2,p-JAK2, STAT3, and p-STAT3 proteins in tumor tissue were observed by immunohistochemical staining. Results Compared with the model group, the tumor volume of mice after combined intervention significantly decreased (P<0. 01) and body mass increased (i>P<0. 05), but there was no significant change food intake. The expression of proliferation related protein Ki67 in tumor tissue was significantly reduced, and the number of apoptotic cells labeled with TUNEL was significantly increased; The expression of p-JAK2 and p-STAT3 proteins is elevated. Conclusions Scutellaria baicalensis decoction can inhibit the JAK2 / STAT3 signaling pathway and increase the inhibitory effect of 5-Fu on mouse lung cancer.
