Abstract:Objective: To investigate effects of sodium houttuyfonate on lung Expression of PI3K and AKT1 and mTORmRNA in rats with chronic obstructive pulmonary disease(COPD), and reveal the possible mechanism of the COPD was treated by sodium houttuyfonate. Methods:Twenty-four Male Wistar rats were randomly divided into normal control group, model control group, dexamethasone and sodium houttuyfonate group(N=6). The COPD model of rats were bulit by the Lipopolysaccharide tracheal instillation and smudging. The expression of PI3K and AKT1 and mTORmRNA were measured by real time PCR . While the histopathology of the lung tissue was observed by light microscope.Results: Compared with the normal control group, the expression of PI3K and AKT1 were obviously increased and mTORmRNA were obviously decreased in model group(P<0.01; P<0.05).Compared with model group, the expression of PI3K and AKT1 were obviously decreased and mTORmRNA were obviously increased in sodium houttuyfonate and dexamethasone group(P<0.01; P<0.05). Compared with dexamethasone group, the expression of mTORmRNA were obviously increased in sodium houttuyfonate group(P<0.05). Light microscope observation indicates that there were local pulmonary consolidation and a large number of neutrophils infiltrated in the alveolar cavity. pulmonary interstitial fibrous tissue hyperplasia with great quantities in the model group. while the pathological manifestations were much more ameliorated than those of the model group, the mild interstitial pneumonia and only a small amount of fibrous tissue hyperplasia were showed in the sodium houttuyfonate and the dexamethasone group. Conclusion:Sodium houttuyfonate lessens the injury of lung tissue and has protective effects on COPD rats ,the mechanism is possibly related to the down-regulatation the expression of PI3K and AKT1mRNA and up-regulatation the expression mTORmRNA in COPD rats .
