    类器官已成为肿瘤研究的重要技术平台,但在模拟肿瘤组织结构和功能方面依旧存在着问题。基因编辑技术的发展,尤其与肿瘤类器官相结合时,为精准、全面地模拟体内肿瘤模型特征提供了新的途径。通过基因编辑技术在肿瘤类器官中引入特定基因变异或修正突变,可以深入解析肿瘤的发生发展机制,探索潜在的治疗靶点,并加速药物筛选过程,为肿瘤个体化治疗提供了新的思路。本文综述了肿瘤类器官的形成及基因编辑策略应用于肿瘤类器官研究的技术要点,重点阐述了基因编辑技术在肿瘤类器官上的独特应用及前景。同时,提出准确模拟体内微环境,推动类器官基因编辑技术的标准化和稳定性,优化基因编辑效率,可以加速肿瘤类器官在精准医学研究中的应用。

    Abstract:

    Organoids have become an important technological platform in cancer research, but there are still challenges in simulating the primary tumor tissue structure and function. The development of gene editing technology, especially when combined with tumor organoids, provides a new approach to accurately and comprehensively simulate the characteristics of in vivo tumor models. Introducing specific gene mutations or correcting mutations in tumor organoids through gene editing technology can deeply analyze the mechanisms of tumor initiation and progression, explore potential therapeutic targets, accelerate the drug screening process, and provide new insights for personalized cancer treatment. This article reviews the formation of tumor organoids and the technical aspects of gene editing strategies, emphasizing the unique applications and prospects of gene editing technology in tumor organoids. Meanwhile, it is proposed that accurately simulating the in vivo microenvironment, promoting the standardisation and stability of organoid gene editing technology, and optimising the efficiency of gene editing can accelerate the application of organoids in precision medicine research.
