    目的:观察电针(electroacupuncture,EA)对糖尿病神经痛(diabetic neuropathic pain,DNP)大鼠体重、空腹血糖、热痛阈、背根神经节(dorsal root ganglion,DRG)瞬时受体电位香草蛋白4(Transient Receptor Potential Vanilloid 4,TRPV4)的影响。方法:采用向腹腔注入链脲佐菌素(streptozotocin,STZ)构建DNP大鼠模型。第15-21 d,在大鼠双侧足三里(ST36)和昆仑(BL60)穴予以每日一次30分钟的2 Hz电针治疗。检测各组大鼠体重、空腹血糖和热痛阈。采用免疫荧光检测大鼠L4-L6 DRG中TRPV4的表达。检测TRPV4激动剂GSK1016790A对电针干预DNP大鼠体重、空腹血糖、热痛阈的影响。结果:第7 d后,与正常组相比,模型组体重明显下降(P<0.01),空腹血糖明显上升(P<0.01);第21 d,与模型组相比,电针组热痛阈明显提高(P<0.01);大鼠L4-L6 DRG上的TRPV4免疫荧光结果显示:模型组的阳性细胞表达较正常组均显著升高(P<0.01),电针组大鼠L4-L5 DRG TRPV4的阳性细胞表达较模型组均显著下降(P<0.01);TRPV4激动剂GSK1016790A组可翻转电针对DNP大鼠热痛阈下调作用(P<0.01)。结论:电针能减轻链脲佐菌素诱导的DNP,其作用机制可能与其通过抑制DRG中TRPV4的表达有关。

    Abstract:

    【t】 Objective: To observe the effects of electroacupuncture on body weight, fasting blood glucose, paw withdrawal latency, transient receptor potential vanilloid 4(TRPV4) in dorsal root ganglia(DRG) in rats with diabetic neuropathic pain(DNP). Methods: The DNP rat model was constructed by intraperitoneal injection of STZ, and electroacupuncture was applied to bilateral Zusanli and Kunlun points once daily for 30 min from D15 to D21. Measured the body weight, fasting blood glucose, hot plate latency. The expression of TRPV4 in rat L4-L6 DRG was detected by immunofluorescence. The effects of TRPV4 agonist GSK1016790A on body weight, fasting blood glucose and heat pain threshold of DNP rats treated with electroacupuncture were detected. Results: After the 7th day, compared with the normal group, body weight was significantly decreased(P<0.01)in the model group and the fasting glucose was significantly increased(P<0.01).
