    目的 恒河猴被广泛应用于感染性疾病的病理、药物和疫苗研究中。为阐述恒河猴Ⅲ型干扰素(IFNL)系统的生物学特征,本文对恒河猴生殖道IFNL及IFNLR1的表达进行了研究。 方法 建立基因特异的RT-PCR方法,对正常和SHIV/SIV感染过的恒河生殖道组织中IFNL1、IFNL3和IFNLR1 mRNA的表达进行观察,利用共聚焦显微术对IFNLR1的组织分布进行观察。 结果 对正常恒河猴阴道和子宫组织的PCR检测未见明确的IFNL1和IFNL3 mRNA扩增条带,但可明确检测到IFNLR1的mRNA和缺失第6个外显子的变体;IFNLR1阳性染色主要位于阴道粘膜上皮和子宫内膜被覆上皮及腺上皮中。在部分SHIV/SIV感染个体中,IFNL1和IFNL3的mRNA水平明显增高,而IFNLR1的mRNA水平几乎没有变化;但Mx1、Mx2和OAS等干扰素刺激基因的mRNA水平显著升高。 结论 这些数据表明恒河猴生殖道可表达IFNL1、IFNL3和IFNLR1,为利用恒河猴开展艾滋病等人类生殖道感染疾病研究提供了基础。

    Abstract:

    Objective Rhesus macaques have been widely used in studies on pathology, drugs and vaccines of human diseases,. To elucidate the biological characteristics of the type Ⅲ interferon system of these monkeys, the expression of IFNLs and IFNLR1 in the genital tract of rhesus macaques was examined. Methods Gene specific RT-PCR methods were established to observe the expression levels of IFNL1, IFNL3, and IFNLR1 mRNA in normal and immunodeficiency virus infected genital tract tissues of rhesus macaques, the distribution of IFNLR1 immunoreactivity in these tissues was examined by confocal microscopy. Results It was found that the levels of IFNL1 and IFNL3 mRNA in the uterus and vagina of normal rhesus macaques are under the threshold of the methods, while the mRNA of IFNLR1 and its variant that lacking exon 6 could be readily detected. The immunoreactivity to IFNLR1 could be detected primarily in the covering or glandular epithelia of the uterus and vagina. Among the SHIV/SIV infected rhesus macaques, mRNA levels of IFNL1 and IFNL3 were increased in some individuals, mRNA levels of IFNLR1 were not changed, and mRNA levels of ISGs (Mx1, Mx2 and OAS) were increased significantly. Conclusions These data indicated that the type Ⅲ interferon system is expressed in the reproductive tract of rhesus macaques as in humans, which provided a scientific basis for using rhesus macaques in studies of human infectious diseases like AIDS.
