    目的 检测化气消浊中药复方对ZDF(Zucker diabetic fatty)大鼠血糖、血脂及细胞因子的影响,探讨化气消浊法对糖脂代谢的调节作用及机制。方法 雄性ZDF (fa/fa)大鼠采用高脂饲料饲喂造成糖尿病模型后,按血糖水平随机分为模型组(8只)和中药组(8只);ZDF (fa/+)大鼠为正常组(8只)。采用化气消浊代表方“气化通调方”连续灌胃6周,给药期间监测大鼠血糖变化,第6周进行口服糖耐量试验。6周后取材并留取血清检测血脂四项、游离脂肪酸;Elisa法检测胰岛素水平;HE染色观察胰腺病理改变;蛋白芯片法检测细胞因子表达并绘制聚类热图。结果 与正常组相比,模型组血糖、血脂四项、游离脂肪酸、胰岛素、炎性细胞因子(CXCL3、IL-2、IL-1α等)水平显著升高,中药组血糖、血脂四项、游离脂肪酸、胰岛素、炎性细胞因子水平较模型组降低。模型组胰岛细胞脱颗粒,中药组胰岛病理损害较模型组缓解。结论 化气消浊法代表方“气化通调方”可一定程度上调节ZDF大鼠的糖脂代谢,缓解慢性炎症损伤。

    Abstract:

    Objective This study aimed to explore the effects of a Chinese medicine formula on regulating glucose, lipid and cytokines in ZDF rats by transforming Qi and eliminating turbidity. Methods ZDF (fa/fa) rats were randomly assigned into the diabetic group (DM, n=8) and the Chinese medicine-administrative group (TCM, n=8) according to the levels of glucose after feeding on high-fat diet. ZDF (fa/+) rats were used in the control group (NC, n=8). Qihuatongtiao, the typical formula that function by transforming Qi and eliminating turbidity, was then administrated to the rats in the TCM group for six weeks. Levels of blood glucose were monitored and an oral glucose tolerance test was performed in the last week. Rats’ serum was used for testing the levels of glucose (BG), triglyceride (TG), cholesterol (TC), high density lipoprotein (HDL), low density lipoprotein (LDL), free fatty acid (FFA) and insulin (FINS). Pathological changes of the pancreas were observed after staining by HE. Levels of cytokines were tested with Raybio cytokine arrays. Results The levels of BG, TG, TC, HDL, LDL, FFA, FINS and pro-inflammatory cytokines of rats in the DM group were significantly higher than those in the NC group, while the levels of them in the TCM group were significantly lower than those in the DM group. There were degranulated islets in the pancreas of rats in the DM group, while less degranulated islets in the TCM group. Conclusion Qihuatongtiao, the Chinese medicine formula function by transforming Qi and eliminating turbidity, regulated glucose and lipid, and alleviated inflammatory lesions in ZDF rats.
