    银屑病(Psoriasis)作为一种在全球范围内广泛流行的慢性炎症性皮肤病,以表皮过度增生、异常角化以及免疫细胞浸润为主要特征,对患者的生活质量和心理健康造成显著负面影响。该病的发生与遗传易感性、免疫失调以及环境因素紧密相关。尽管目前在银屑病发病机制的研究上已取得了重要进展,但现有的治疗手段仍存在疗效有限和副作用明显等问题,因此深入剖析其病理网络并开发新型干预策略迫在眉睫。在此背景下,咪喹莫特(Imiquimod)诱导的银屑病动物模型凭借其高度的可重复性和良好的病理模拟性,成为研究银屑病的关键工具。本综述系统梳理了咪喹莫特诱导银屑病模型的核心作用机制,详细阐述了其通过核心免疫炎症轴的级联反应、下游协同机制的多元调控网络以及宿主与环境因素的交互作用等途径发挥作用的分子基础;基于该模型的研究已成功验证了多种靶向疗法和天然产物对银屑病的治疗效果,充分彰显了其在治疗干预研究中的重要应用价值;此外,对咪喹莫特诱发银屑病模型存在的局限性进行了探讨,并对其未来的研究方向进行了展望,以期为银屑病的深入研究和治疗提供参考。

    Abstract:

    Psoriasis, a chronic inflammatory skin disease prevalent worldwide, is primarily characterized by epidermal hyperplasia, abnormal keratinization, and immune cell infiltration, exerting a significant negative impact on patients’ quality of life and mental well-being. The onset of this disease is closely associated with genetic susceptibility, immune dysregulation, and environmental factors. Although significant progress has been made in the research on the pathogenesis of psoriasis, the existing treatment methods still have problems such as limited efficacy and obvious side-effects. Therefore, it is extremely urgent to conduct an in-depth analysis of its pathological network and develop novel intervention strategies. Against this backdrop, the imiquimod-induced psoriasis animal model has become a crucial tool for studying psoriasis due to its high reproducibility and excellent pathological simulation. This review systematically summarizes the core mechanism of action of the imiquimod-induced psoriasis model, elaborately expounds on the molecular basis of its action through pathways such as the cascade reaction of the core immune-inflammatory axis, the multi-regulatory network of downstream synergistic mechanisms, and the interaction between host and environmental factors. Research based on this model has successfully verified the therapeutic effects of various targeted therapies and natural products on psoriasis, fully demonstrating its important application value in therapeutic intervention research. In addition, the limitations of the imiquimod-induced psoriasis model are discussed, and the future research directions are prospected, aiming to provide references for in-depth research and treatment of psoriasis.
