    摘要:

    目的 研究肉桂枝叶水提物不同极性部位对气虚大鼠的补气作用及机制,为肉桂补气的药效物质基础提供参考。方法 将90只Wistar雄性大鼠随机分为空白组、模型组、阳性组、乙酸乙酯部位高剂量组、乙酸乙酯部位低剂量组、正丁醇部位高剂量组、正丁醇部位低剂量组、水部位高剂量组、水部位低剂量组,每组10只。除空白组外,其余各组均采用“饮食不节+疲劳”法构建气虚动物模型,造模同时,灌胃给药,每天1次,连续给药21 d。末次给药后,记录大鼠的体质量、负重游泳时间,测定血清中抗肿瘤坏死因子-α(anti-tumor necrosis factor-alpha,TNF-α)、白介素-2(interleukin-2,IL-2)、白介素-12(interleukin-12,IL-12)、肌酸激酶(creatine kinase,CK)、乳酸(lactic acid,LD)、乳酸脱氢酶(lactic dehydrogenase,LDH)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malonaldehyde,MDA)、白蛋白(albumin,ALB)及肝脏中二磷酸腺苷(adenosine diphosphate,ADP)、三磷酸腺苷(adenosine triphosphate,ATP)等指标,测定胸腺指数、脾脏指数、肝脏指数,使用HE染色法对各组大鼠的肝脏进行病理观察。结果 与模型组比较,各给药组大鼠的气虚症状及肝脏组织损伤均得到了不同程度改善;乙酸乙酯部位、正丁醇部位均可显著延长气虚大鼠的负重游泳时间(P<0.01);乙酸乙酯部位高剂量可显著升高胸腺指数、TNF-α、IL-2、IL-12、ADP、ATP、SOD、ALB含量(P<0.05,P<0.01),显著降低CK、、LD、LDH、MDA含量(P<0.05,P<0.01);乙酸乙酯部位低剂量可显著升高IL-2、ATP、ADP、ALB含量(P<0.05,P<0.01),显著降低MDA、LD、LDH含量(P<0.05,P<0.01);正丁醇部位高剂量可显著升高胸腺指数、IL-2、IL-12、ATP、ADP、ALB、SOD含量(P<0.01),显著降低TNF-α、CK、LD、LDH、MDA含量(P<0.05,P<0.01);正丁醇部位低剂量可显著升高SOD、IL-1、ALB、ATP、ADP含量(P<0.05,P<0.01),显著降低CK、LDH、LD、MDA含量(P<0.05,P<0.01);水部位高剂量显著升高IL-12、ATP含量(P<0.05),显著降低TNF-α、LDH、MDA含量(P<0.05,P<0.01);水部位低剂量可显著降低LD含量(P<0.05)。结论 肉桂枝叶水提取物不同极性部位可不同程度改善大鼠气虚症状及保护气虚大鼠的胸腺、脾脏、肝脏,且可调控气虚大鼠多种因子水平,从而提高其免疫力、减少脏器损伤等多种效果以达到补气、延长疲劳发生时间、增强机体的运动耐力等作用。

    Abstract:

    Objective To study the qi-tonifying effect and mechanism of different polar parts of water extract of Cinnamomum cassia branches and leaves on qi-deficiency rats, and to provide reference for the pharmacodynamic material basis of Cinnamomum cassia qi-tonifying. Methods Ninety Wistar male rats were randomly divided into blank group, amodel group, positive group, ethyl acetate fraction high dose group, ethyl acetate fraction low dose group, n-butanol fraction high dose group, n-butanol fraction low dose group, Water fraction high dose group, Water fraction low dose group, with 10 rats in each group. Except for the blank group, the other groups used the ' improper diet + fatigue ' method to construct the animal model of qi deficiency. At the same time of modeling, the rats were given intragastric administration once a day for 21 days. After the last administration, the body weight and weight-bearing swimming time of the rats were recorded. Anti-tumor necrosis factor-alpha ( TNF-α ), interleukin-2 ( IL-2 ), interleukin-12 ( IL-12 ), creatine kinase ( CK ), lactic acid ( LD ), lactic dehydrogenase ( LDH ), superoxide dismutase ( SOD ), malondialdehyde ( MDA ), albumin ( ALB ) in serum and adenosine diphosphate ( ADP ), adenosine triphosphate ( ATP ) in liver were measured. The thymus index, spleen index and liver index were measured, and the liver of each group was observed by HE staining. Results Compared with the model group, the qi deficiency symptoms and liver tissue damage of the rats in each administration group were improved to varying degrees. The ethyl acetate fraction and n-butanol fraction could significantly prolong the weight-bearing swimming time of Qi deficiency rats ( P < 0.01 ). The high dose of ethyl acetate extract could significantly increase the thymus index, TNF-α, IL-2, IL-12, ADP, ATP, SOD and ALB contents ( P < 0.05, P < 0.01 ), and significantly reduce the contents of CK, LD, LDH and MDA ( P < 0.05, P < 0.01 ). The low dose of ethyl acetate fraction could significantly increase the contents of IL-2, ATP, ADP and ALB ( P < 0.05, P < 0.01 ), and significantly reduce the contents of MDA, LD and LDH ( P < 0.05, P < 0.01 ). The high dose of n-butanol fraction could significantly increase the thymus index, IL-2, IL-12, ATP, ADP, ALB and SOD contents ( P < 0.01 ), and significantly reduce the contents of TNF-α, CK, LD, LDH and MDA ( P < 0.05, P < 0.01 ). The low dose of n-butanol fraction could significantly increase the contents of SOD, IL-1, ALB, ATP and ADP ( P < 0.05, P < 0.05 ). Conclusion The different polar parts of the water extract of Cinnamomum cassia branches and leaves can improve the symptoms of qi deficiency in rats to varying degrees and protect the thymus, spleen and liver of qi deficiency rats, and can regulate the levels of various factors in qi deficiency rats, thereby improving their immunity and reducing organ damage. Various effects such as qi supplementation, prolonging the time of fatigue, and enhancing the body 's exercise endurance.
