    人源化造血干细胞(Humanized hematopoietic stem cells,Hu-HSCs)模型是模拟人源T细胞分化和功能的关键模型,然而重建小鼠由于背景为重度免疫缺陷,其胸腺发育受阻或没有胸腺,缺乏T细胞分化成熟的场所,导致重建小鼠人源功能性T细胞不足。尽管移植人胎儿胸腺组织能够支持人源化小鼠体内T细胞的有效发育,但供体组织稀缺和伦理问题限制了其广泛应用。因此,亟待开发新型胸腺组织克服上述局限性。本文系统总结了Hu-HSCs小鼠模型的基本特征及传统胎胸腺移植诱导的人T细胞免疫应答能力;重点综述了新型胸腺移植在促进T细胞发育、调节人类T细胞介导的免疫应答方面的作用,揭示其潜在机制及对提升人体免疫力的重要意义,期望为开发新的人源化小鼠模型提供理论依据和技术支持。

    Abstract:

    The Humanized hematopoietic stem cells (Hu-HSCs) model is a key model for simulating the differentiation and function of human T cells. However, due to the background of severe immune deficiency in reconstructed mice, thymic development is hindered or there is no thymic gland, lacking the site for T cell differentiation and maturation. It leads to insufficient reconstruction of human functional T cells in mice. Although the transplantation of human fetal thymus tissue can support the effective development of T cells in humanized mice, the scarcity of donor tissues and ethical issues have limited its wide application. Therefore, it is urgent to develop new types of thymus tissue to overcome the above limitations. This article systematically summarizes the basic characteristics of the Hu-HSCs mouse model and the human T-cell immune response ability induced by traditional fetal thymus transplantation. This paper mainly reviews the role of the novel thymus transplantation in promoting T cell development and regulating the immune response mediated by human T cells, reveals its potential mechanism and significance in enhancing human immunity, and is expected to provide theoretical basis and technical support for the development of new humanized mouse models.
