    为了解决临床同种器官移植中供体器官短缺问题,研究者们开始探索异种器官移植这一方法的可行性。2002年,全球首例GTKO猪的问世及其表现出的较好的免疫排斥结果效果,证明了基因编辑对异种移植的重要性。目前,随着体细胞克隆技术和基因编辑技术的不断完善,研究者逐步确立了以基因编辑猪为异种移植供体的研究方向。本文综述了近年来全球具有代表性的“猪-人”异种器官移植案例,阐述了每一个案例的实验结果与基因编辑的类型;针对不同类型的免疫排斥反应,分析了不同基因编辑策略对异种移植的影响和机理机制;进一步总结了编辑基因的选择与组合策略,提出用于异种移植的“工具猪”概念,并对相关研究中存在的问题进行了讨论,期望为优化异种移植供体猪的基因编辑策略提供新的思路。

    Abstract:

    To address the shortage of donor organs in clinical allogeneic organ transplantation, researchers have started to explore the feasibility of xenotransplantation. In 2002, the world"s first GTKO (α-1,3-galactosyltransferase knockout) pig was created, and its demonstrated reduction in immune rejection effects proved the significance of gene editing in xenotransplantation. At present, with the continuous improvement of somatic cell cloning technology and gene editing technology, researchers have gradually established the research direction of using gene-edited pigs as donors for xenotransplantation. This review summarizes representative global cases of "pig-to-human" xenotransplantation in recent years, elaborates on the experimental results and types of gene editing for each case; For different types of immune rejection reactions, it analyzes the impacts and mechanisms of different gene editing strategies on xenotransplantation; Furthermore, it summarizes the strategies for selecting and combining edited genes, The concept of "tool pigs" for xenotransplantation is proposed, and the problems existing in related studies are discussed, expecting to provide new ideas for optimizing the gene editing strategies of donor pigs for xenotransplantation.
