    目的 探讨菲牛蛭(Poecilobdella manillensis)对角叉菜胶(Kappa-carrageenan,KCG)诱导小鼠尾部血栓形成的抑制作用。方法 连续7d以菲牛蛭提取物(JC-17)2.5、25、100 mg/kg和肝素钠50 mg/kg腹腔注射处理昆明小鼠,于末次给药1h后采用0.5 %角叉菜胶诱发小鼠尾部形成血栓。结果 48 h Model组小鼠黑尾率为94.91 ± 3.927,表明造模成功,L、M、H组较Model组的小鼠黑尾率呈梯度下降趋势;提取物显著延长凝血酶原时间(PT)、降低纤维蛋白原(FIB)含量;Model组脾脏TNF-α、IL-1β、IL-6含量升高,其中TNF-α最高,为433.8±12.1 pg/mL,而给药组均显著抑制炎症因子含量。结论 菲牛蛭提取物能有效抑制小鼠尾部血栓形成,同时具有抗炎作用,提示该提取物在炎症相关性血栓疾病的防治中具有潜在应用价值。

    Abstract:

    Objective To investigate the inhibitory effect of Poecilobdella manillensis (PM) extract on kappa-carrageenan (KCG)-induced tail thrombosis in mice. Methods Kunming mice were intraperitoneally administered PM extract JC-17 (2.5, 25, or 100 mg/kg) or heparin sodium (50 mg/kg) for 7 consecutive days. One hour after the last dose, tail thrombosis was induced by 0.5% KCG injection. Results At 48 h, the tail blackening rate in the Model group was 94.91 ± 3.93%, confirming successful thrombosis induction. The PM-treated groups (L, M, H) exhibited a dose-dependent reduction in tail blackening. The extract significantly prolonged prothrombin time (PT) and reduced fibrinogen (FIB) levels. In the Model group, splenic TNF-α, IL-1β, and IL-6 levels were elevated, with TNF-α reaching the highest concentration (433.8 ± 12.1 pg/mL). All PM-treated groups showed significant suppression of these inflammatory cytokines. Conclusion PM extract effectively inhibits KCG-induced tail thrombosis and exhibits anti-inflammatory activity, suggesting its potential therapeutic value for inflammation-associated thrombotic disorders.
