Abstract:bjective To estabolish vascular dementia animal model with ligation of bilateral common carotid arteries, the volume of blood flow in various regions of rat brain was determined. Methods The changes in volume of flood flow in different brain regions were measured by laser Doppler blood flow monitor within 10 minutes after ligating the bilateral common carotid arteries of rats. Results The average of volume of brain blood flow decreased (lobus frontalis 67.83%; lobus parietalis 56.82%; lobus occipitalis 16.16%; basal nucleus of Meynert 51.21%; caudate putamen 41.18%; field CA1 of the Ammon's horn 51.83%; field CA2 of the Ammon's horn 41.21%) after the ligation. Conclusion The volume of blood flow in the brain regions closely related to the ability of learning and memory all declined significantly.

Abstract:用01%、03%、05%和075%四种不同浓度的蜂胶乙醇提取物(LiquidofEthanotExtratedfrompropolis)EEP来净化自发感染螨虫的ICR小鼠。发现不同浓度的EEP对体表螨虫均有不同程度的净化作用,但05%浓度的EEp净化效果最佳。净化后的小鼠精神状态较好,被毛光洁贴身,无异常反应,镜检未查见螨虫。结果表明EEP天然、无毒〔1〕,价廉易得,是一种高效、简便、理想的实验动物体表螨虫的生物净化剂。

Abstract:ix germfree KM mice were obtained by hysterectomy or caesarian section, the young were kept in a germfree environment which is called isolator, and were fed with artificial milk by hand. All newborns were confirmed to be germfree by sterility testing. The feces, bedding and swab are examined for germfree detection, all test samples are negative, this result indicate that the germfree animal feeding are successful. Except above, we analysied nutrition of artificial milk and compared with mother milk of mice.

Abstract:目的对Ⅱ型胶原蛋白(CⅡ-A)和弗氏完全佐剂(A-A)大鼠关节炎模型在大体外观和足部组织病理学切片等方面进行观察比较。方法分别采用Ⅱ型胶原蛋白和弗氏完全佐剂诱导建立大鼠关节炎模型,利用排水法对大鼠足部体积进行测定,并将大鼠后足进行组织病理学切片观察。结果从大体外观和足部病理切片上两种大鼠模型均显示出有明显的病变,但CⅡ-A大鼠与A-A大鼠比较,滑膜增生及软骨破坏等继发性病变特征更为明显,关节炎持续时间也较长,更接近于人的类风湿性关节炎。结论CⅡ-A大鼠模型与A-A相比是研究RA较为理想的动物模型。

Abstract:Objective The aged rats, basal forebrain-injured rats and the rats injected with scopolamine are ofter used as animal models of alzheimer's disease. A behavioral comparison among three animal models were carried out in the study. Method Water maze and open field were applied in above three animal model groups, youth control group and sham operation control group. Results The ability of learning and memory in the aged rats, basal forebrain-injured rats and scopolamine-injected rats decreased significantly. The ability of spatial cognition in basal forebrain-injured rats and scopolamine-injected rats significantly declined. For the scopolamine-injected rats the excitability in new environment abnormally increased, (P<0.05). Conclusions The behavioral changes in basal forebrain-injured rats are more similar to the early clinical manifests of alzheimer's disease than those in aged rats and scopolamine-injected rats.

Abstract:采用细胞外微电极技术,记录离体灌流的蟾蜍椎旁交感神经节细胞膜电位,观察川芎嗪对嘌呤受体介导反应的调制作用。三磷酸腺苷(300μmol/L)可引起神经节细胞膜去极化(n=62)、超极化反应(n=27)以及去极化之后伴随超极化过程的双相反应(n=9)。P2受体拮抗剂台盼蓝(500μmol/L)可抑制三磷酸腺苷的去极化反应(n=8);P1受体拮抗剂氨茶碱(200μmol/L)可抑制三磷酸腺苷的超极化反应(n=7)。滴加川芎嗪(1~5mmol/L),神经节细胞膜未出现明显的电位变化。外源性环-磷酸腺苷(250μmol/L)可模拟三磷酸腺苷的超极化反应(n=9)。川芎嗪(3mmol/L)可抑制三磷酸腺苷的去极化反应,使其幅值减少539±95%(n=14,P<001),并能加强三磷酸腺苷所致超极化反应,使其幅值增大1054±245%(n=12,P<001)。在同一标本上,川芎嗪使环一磷酸腺苷的超极化反应加强(n=4)。此外,川芎嗪可抑制三磷酸腺苷引起的双相反应中的去极相,而增大其后的超极相(n=3)。

Abstract:In order to maintain embryonic stem cells (ES cells) in a proliferative, unidifferentiated state in vitro. Mitomycin C-treated primary mouse embryo fibroblasts (PMEFs) were used as the feeder cells in media of DMEM supplement with LIF for co-culture with ES-D3 cells. ES cell colonies grow in small clusters. Many laboratories including ours, find that culture on feeder layers of PMEFs yields high quality ES cells with greater consistency. Some techniques that maintaining ES cell in a pluripotent state were be discussed in detail.

Abstract:本文采用改进的体外生物测定法分析恒河猴月经周期中促卵泡刺激素(FSH)、促黄体生成素(LH)的变化情况。结果表明:雌猴的FSH峰出现在月经周期的第102±10天,峰值:572±48ng/ml;LH峰出现在月经周期的第120±16天,峰值:963±97ng/ml,且为滤泡期、黄体期平均值的7~8倍。证实了在雌性恒河猴的月经周期中,其LH分泌高峰与FSH分泌高峰不在同一时间出现的现象。

Abstract:In this study, the effect of natural ceramide has been tested on primary cultures of cortical neurons. In order to study if the cortical neurons undergoing apoptosis, some markers such as the leakage rate of lactate dehydrogenase(LDH) and the MTT metabolic rate were assessed, also the morphological changes were observed by Giemsa staining. DNA ladders were determined by gel electrophoresis, as well as the cell ultrastructurales were observed under electronmicroscope. The results showed that 500 to 1000nM of ceramide could induce apoptosis of culture cortical neurons in dose-dependent and time-dependent way after 12 hrs of incubation. The ultrastructural changes are consistent with the changes observed by Giemsa staining or DNA ladders on gel electrophoresis.

Abstract:M53 stardand strains were detected using one-step method amplipifiction and southern boltting a pair of oligonuoleotide primers and probe, were designed, synthesized and purified a specific, sensitiv and rapid detected meothod on agarose gel eletrophoresis products showed the 710bp specific band of M53 genomice DNAs. of 50 subjects were detected from SD rats, the results suggest that test-rate of PCR was higher than that of isolates cultivate method. The amplified products were confirmed by southern boltting, using alkaline phophatase (AP) labeled oligonuoeleotide probe and the specificity DNAs sequence by southern blotting onto Nylon Hybond-N membranes, and the negtive cotrans were no signals. By PCR specific detection, as little as 10pg of Mycoplasma Pulmonis DNAs yielded positive results. These results indicated that one-step PCR assay was rapid, specific, sensitive, and suitable for the detection of rats and mice.

Abstract:ransgenic technique is one part of the development engineering or embryonic manipulation. This techniques include collecting embryos, in vitro culture of embryos and transferring embryos as a basic technology. Production of transgenic mice is one kind of application technology. The foreign DNA ware microinjected into nucle of fertilized one cell eggs and integrated into apparently random sit in the host genome. We produce transgenic mice by microinjection with NchW (Human prion protein gene),pCSLN(Sialidase-L gene),prbo(Bovine prion protein gene),CXTGAP(Human GM 2 activation protein gene),Prsh (Sheep prion protein gene) DNA fragments into the pronucleus of fertilized eggs. The DNA integrates on chromosomes at random and we got the positive result from the pups by PCR-Southern method. The average positive rate of transgenic mice is 16.38%. The present work proved that the main requirement of succeed on production transgenic mice should include precise technical skill, animal chosen and husbandry,manipulation

Abstract:目的为建立溶血性贫血(HA)和再生障碍性贫血(AA)两种贫血动物模型,研究HA和AA的发病机理。方法50只杂种健康家兔随机分为两组,25只皮下注射盐酸苯肼制备溶血性贫血(HA)模型;另25只行60Coγ-射线亚致死量全身照射后输注同种异体胸腺淋巴细胞诱发AA型。结果HA组成功率100%,死亡率56%(14/25只);AA组成功率为92%(23/25只)。结论两种模型的发生时间较稳定,实验重复性好,结果可靠,这为HA和AA发病机理的探讨及实验性研究奠定了基础
