Abstract:Human peripheral blood lymphocytes (hu-PBL), were transferred to SCID(severe combined immunodeficient) mice by intraperitoneal injection, thus hu-PBL-SCID mice model was successfully established. Significant levels of human immunoglobulin were detected in sera of hu-PBL-SCID mice 4 weeks after engraftment. 8 kinds of immunonological phenotype of human lymphocyte were detected in the spleens of hu-PBL-SCID mice by flow cytometry 4 weeks after engraftment. These data demonstrated that proliferation or reconstitution of human lymphocytes appeared in the SCID mice. It suggested that and there was no difference of metastases between hu-PBL-SCID mice and unreconstituted SCID mice receiving transplanted human lung carcinoma PLA-801DL. The human lymphocytes in the hu-PBL-SCID mice have no significant antitumor effect.

Abstract:本文观察了用抗氧化剂维生素E预处理后,三氯乙烯(3000mg/kg B-W-) 一次性经口染毒24h 大鼠肝脏的超氧化物歧化酶、谷胱甘肽过氧化物酶等抗氧化酶活力及丙二醛含量的变化,结果表明三氯乙烯染毒组肝脏中丙二醛含量、超氧化物歧化酶活力及血清谷丙转氨酶、谷草转氨酶活力均高于对照组(P< 0-01) ;而维生素E干预组的丙二醛含量、超氧化物歧化酶活力及血清谷丙转氨酶、谷草转氨酶活力均分别低于三氯乙烯染毒组(P< 0-01) ,说明三氯乙烯急性染毒可引起肝脏脂质过氧化反应及肝损害,肝脏超氧化物歧化酶活力升高可能是机体受自由基及脂质过氧化反应刺激而诱导产生的一种适应性反应,维生素E对三氯乙烯所致的肝损害有一定的保护作用。

Abstract:目的 介绍一种急性水肿性胰腺炎(AEP) 向急性出血坏死性胰腺炎(AHNP) 演变的大鼠模型,并对其超微结构进行了观察。方法 30 只SD大鼠随机分为正常组、AEP组和左旋精氨酸(L- Arg) 干预组。通过阴茎背静脉注射雨蛙素诱导AEP模型后,再经该静脉注入L- Arg 1600mg/kg,以观察血清生化指标和胰组织的光镜、透射电镜下改变。结果 血清淀粉酶、脂肪酶水平在AEP组、L- Arg 干预组均明显高于对照组,其中血清淀粉酶在两组间无显著性差异,而L- Arg 干预组的血清脂肪酶却明显高于AEP组;AEP组的血清、胰组织NO浓度明显低于对照组,而L- Arg 组明显高于对照组和AEP组。光镜及透射电镜证实,L- Arg 干预AEP大鼠后,导致了胰实质出血、坏死和腺泡细胞内亚细胞结构的破坏。结论 L- Arg 导致AEP加重为AHNP,与NO 的毒性有关;此大鼠模型不但有血清淀粉酶等生化指标的改变,而且组织病理特点也较为典型,是研究急性胰腺炎重型化机理的良好模型。

Abstract:The sever combined immunodieficiency mice (Scid) was selected and injected infection larvae of wuchereria bancrofti for animal model established. The results showed that bancrofti filaria was easy infected scid and grow, develop into adults to continue producing microfilaria in Scid. The collecting rate of adult worms is as high as 14.8% around 6-30%. The microfilaria in circulation blood not only can be check up in all of Scid infected, adult worm also can be collected, meaning 100% of scid was successful for infected with bancrofti filaria. It indicated that due to immunodeficiency of Tand B lymphatic cells of Scid mice for antibody producing, it is a valuable laboratory animal with following characters easy infected, high microfilarial density, high collected rate of worm. It is good laboratory animal model for bancrofti filaria.

Abstract:对四个单位保种的S180 细胞经KM 小鼠腹腔传代,体外培养加秋水仙碱,涂片、染色后,显微镜下计数染色体数目:腹水传代的S180 细胞,用70 % 乙醇固定,流式细胞仪测DNA 含量。结果如下:本学部( 本部) ,中国医学科学院药物所( 药物所) ,武汉大学保种中心( 武汉大学) 和北京市肿瘤所( 肿瘤所) 保种的S180 细胞株,其染色体均数分别为62-8 ±22-8 ,69-1 ±21-2,39-9 ±8-26 ,58-7±9-75 条。四单位S180 细胞株染色体数做方差分析表明,除肿瘤所与本部外,其它两单位比较均有显著统计学差异(P< 0-01) 。直方图分析显示主流染色体范围分别为56 ~60 ,61 ~65,41 ~45,61 ~65 条。流式细胞术DNA 含量分析表明肿瘤所S180 DNA含量最多,武汉大学保种的S180 细胞的DNA含量最少。这些结果均证明四个单位的S180 细胞株在一些方面已出现显著差异。

Abstract:The value of six blood enzymes and two plasma protein levels in spontaneous NIDDM Chinese hamster were measured and their relationships were determined. The results were: five blood enzymes and plasma total protein of NIDDM animals all have a higher level than those of control (P<0.01), which suggests that the spontaneous NIDDM inbredline Chinese hamsters pathogenesis glycaemia and protein metablic abnormality. Abstract:将6 - 羟基多巴胺(6 - OHDA) 分两点注入SD大鼠左侧中脑黑质内侧端,制成震颤麻痹模型,毁损后6 ~8 周,用胚龄14~16 天同种胚鼠中脑黑质细胞悬液植入模型鼠尾壳核。实验分正常对照组,模型对照组,单纯胚中脑黑质移植组(MS组) ,含层粘蛋白Laminin 胚中脑黑质移植组(LMS组) ,含雪旺氏细胞胚中脑黑质移植组(SMS组) 。于移植后2 ~4 个月分别测试旋转试验,MS组、LMS组及SMS组与模型组间均有显著差异(P< 0-01) 。动物经测试后处死。脑切片观察发现,损伤侧中脑黑质DA神经元95% 以上死亡,移植组织位于尾壳核背侧或突入侧脑室,LMS组移植区> SMS组移植区> MS组移植区。三个移植区均见TH、GABA 免疫阳性细胞,其形态及大小与正常对照侧中脑黑质所见相似。LMS组与SMS组多巴胺阳性细胞突起较MS组多且长。本实验证明中脑黑质移植物Laminin 与雪旺氏细胞均能促进DA能神经元突起生长。

Abstract:The human liver tissue was homogenized. It's mitochondria were separated and purified by fraction centrifuge and step ingredient centrifuge. The purified mitochondria were micro-injected into the cytoplasm of the feritlized eggs. There is sixty-one percent microinjected eggs survived immediately after the manipulation. Among them, fifty-three percent eggs pass the primary division. The results suggested the possibility of production of mice bring human mitochondria.

Abstract:To study mouse oocytes in vitro maturation and fertilization in different culture. Methods Compare the maturation and fertilization rate in vitro of mouse oocytes, which were culture in FSH, Insulin or BSA, and were fertilized in Whitter's medium. Results 1.IVM/IVF rate of Do (81.4%, 31.0%) were higher than those of COC (48.6%, 27.1%). 2.Mouse oocytes cultured with FSH, insulin and BSA, the IVM rate were 77.9%, 82.3% and 60.7%, the IVF rate were 77.2%, 72.6% and 26.7%, the 2-cell rate were 49.2%, 34.2% and 10.0% respectly. The group cultured with insulin (I-Group) have the highest rate of IVM, but after fertilization, fertilized oocytes and 2-cell embryos of I-Group are fewer than those cultured with FSH. 3.two groups cultured with BSA have lower rate in vitro fertilization rate (26.7%, 25.8%). 4.Fertilization and 2-cell rate in those oocytes with polar body 1 (85.9%, 22.4%) were higher than those of GV-stage oocytes (71.1%, 12.9%). Conclusions 1.In vitro maturation and fertilization rate of Do were significantly higher than those of COC (P m<0.01; P f<0.05). 2.The maturation and fertilization rate were improved when the mouse oocytes were cultured in FSH and Insulin mediums. 3.BSA affected the fertilization rate of mouse oocytes significantly. 4.The IVF rate and 2-cell rate of oocytes with PbI are higher than those of GV-stage oocytes (P f<0.05; P 2-cell <0.05).

Abstract:To investigate the PCR (polymerase chain reaction, PCR) assay for the detection of Mycoplasma pulmonis in infected rats and try to set up a feasible, quick and sensitive method for the diagnosis. Method 14 samples of the supernatants of throat swab samples and the cultural supernatants of throat swab from infected rats are performed PCR analysis using animal mycoplasms routine primers and M. Pulmonis species-specific primers respectively. The PCR products are confirmed with 2% agarose gel electrophoretic analysis. The M 53 and ATCC19612 standard strain of Mycoplasma pulmonis are used as positive control. Results Using animal mycoplasma routine primers, 8/14 of the supernatants of throat swab samples are positive in the PCR assay whereas 14/14 of the cultural supernatants from throat swab specimen are positive. With M.Pulmonis species-specific primers, the supernatants of throat swab samples are negative in the PCR analysis but 3/14 of the cultural supernatants from throat swab samples are positive. The mycoplasma routine primers can detect the M 53 and ATCC19612 standard strain, on the other hand, M.pulmonis species-specific can only detect M 53 but ATCC19612 strain. Conclusions The PCR assay using mycoplasma routine primers is superior to isolated culture for detecting M. Pilmonis, which could save time and labor. Nevertheless, whether the M.Pilmonis species-specific primers which the foreign researchers suggested could be used to specially detect the M.Pulmonis in the infected rats, need to be further investigated.

Abstract:本研究以羔羊卵巢母细胞为材料,把体外受精技术应用到绒山羊育种工作中,观察了羔羊体外受精卵的体外发育及移植产羔的效果,以便探讨通过体外受精技术缩短绒山羊育种周期的技术途径。对406 枚羔羊体外受精卵用M199 hcG、M199 LH 和M199 hcG EGF三种培养基进行体外发育培养。结果是,2~4 细胞期胚的发育率平均分别为40-0 % 、43-6 % 和49-6 % ;桑椹胚和囊胚的发育率平均分别为17-5 % 、15-8 % 和18-8 % 。将49 枚F1 代羔羊2~8 细胞期胚移植给31 只受体母羊,结果有11 只妊娠(35-5 % ) 并产羔12 只(F2 代) ;将24 枚F2 代羔羊2~4 细胞期胚移植给15 只受体母羊,结果有2 只妊娠(13-3 % ) 并产F3 代羔羊2 只。

Abstract:Cell suspensions of LJH-OS were injected to nude mice subcutaneous. In tact tumor pieces, obtained from the third serial passage of subcutaneously growing xenografts, were implanted into the proximal tibia and the subcutaneous tissues of nude mice respetively. Animals were sacrificed and autopsied 4 and 7 weeks after transplantation and examined macroscopically for local tumor growth and metastasis. The tumor formation rate, the growth rate, the invasion and metastatic behavior of the orthotopic and ectopic xenografts were investigated and compared. We discovered the tumor with orthotopic transplatation had a faster growth rate than the s.c implantation. Tumors in the tibia invaded and destroyed the normal structure of the tibia bone, also infiltrated to the nearby tissues. The lung metastasis and reactive bone formation were observed only in the tibia transplatation model. It can be concluded the orthotopic transplantation of tumor tissue of the LJH-OS cell line into the tibia of nude mice resulted in local tumor growth and spontaneous metastasis which closely resembled the clinical situation. Therefore this model may be useful for experimental therapy studies and for investigations on the role of bone in regulating tumor growth, invasion and metastasis of osteosarcoma.
