    • Expression and Significance of VEGF Receptors in the Nude Mouse Models with Various Human Tumors

      Abstract:Objective To study the expression of vascular endothelial growth factor (VEGF) receptors flt\|1 and KDR in the nude mouse models with various human tumors subcutaneously transplanted by immunohistochemical method.Methods\ Tumor tissues were collected from the transgenic mice and washed,fixed,parraffined,sliced continuously and immunohistochemically stained.Results\ flt\|1 was determined to be strongly positive or positive in the vascular endothelial cells and tumor cells of 11 nude mouse models with various human tumors, that is,human gastric adenocarcinoma SGC,human malignant melanoma A375,human breast cancer Bcap\|37,human glioma U251,human intestinal adenocarcinoma LOVO,human hepatoma SMMC\|7721,human cervial cancer,human gastric adenocarcinoma SPC\|A1,human non\|small cell lung cancer H128,human ovarian cancer 3AO,human hepatoma graft,while KDR was determined to be positive or weakly positive in the vascular endothelial cells and tumor cells of 6 nude mouse models with various human tumors, that is,human gastric adenocarcinoma SGC,human maliganant melanoma A375,human breast cancer Bcap\|37,human glioma U251,human intestinal adenocarcinoma LOVO,human ovarian cancer 3AO.Conclusion\ Coexpression of VEGF receptors in tumor vascular endothelial cells and tumor cells raises the possibility of autocrine stimulation and emphasizes the importance of the reaction of VEGF and VEGF receptors to tumor growth.

    • The Efficient Derivation of Embryonic Stem Cell Colonies in Kunming Mouse

      Abstract:Objective To investigate the efficiency of isolation and cloning of ES cell colonies in Kunming mouse. Method\ Three culture conditions were adopted,namely, PMEF feeder layer, NIH3T3 feeder layer and LIF without feeder layer. Results\ According to the formation rate of ES cell colonies, the two kinds of feeder layer are better than LIF only, the blastocysts are better origin for ES cells than morulae and there are no differences between the two kinds of feeder layer. Conclusion It is a more efficient way to isolate and clone ES cell colonies from Kunming mouse blastocysts by PMEF or NIH3T3 feeder layer.

    • Study on Functions of Four Mouse Genes Relative to Feeding Amount, Drinking Amount and Protein Metabolism

      Abstract:Objective The purpose is providing some scientific bases for further research of nutriology and nutritional disease science on molecular biological level by studying on functions of mouse genes relative to feeding amount,drinking amount and protein metabolism. Method\ Employing antisense oligo nucleotide technology and animal behavioral tests,the authors studied gene function relative to animal meals and nutritional metabiosm on the whole body level of animal.The authors selected four genes having obvious differences of feeding amount with control groups in the past research,and used BALB/c mice to do experiments.Results\ The feeding and drinking amounts of D1,Pe and Tr antisense groups have obvious differences with control groups,and the proteins metabolism of D1 and Tr antisense groups have obvious differences with control groups.The feeding and drinking amount of D1 is much greater than control groups and the tendency is downwards,but the apparent digestibility is lower than control groups.This indicates that its function of proteins metabolism is dropping.The feeding and drinking amount of Tr is also much greater than control groups but the tendency is upwards and the apparent digestiblity is greater than control groups.This illustrates that its function of proteins metabolism is going up.Conclusion D1 gene is probably relative to improving nutritional metabolism function and Tr gene has the function of inhibiting nutritional metabolsim.Moreover,D1,Pe and Tr are relative to other behavior such as locomotion,sense of pain and memory;P3 has no relation to other behavior.

    • Rat Model of Type 2 Diabetic Mellitus and Its Glycometabolism Characters

      Abstract:Objective To develop a rat model of type 2 diabetic mellitus. Method\ 8 weeks old SD rats were injected with low dose STZ after high calorie given for two months.Then insulin\|glucose tolerance test,islet immunohistory and other corresponding tests were performed and analysed.The data were compard to the others such as purely low dose STZ,high dose STZ and high calorie.Results\ In this rat model,insulin sensitivity decreased,and the synthesis and excretion of insulin were lower than that of purely high calorie group but still higher than that of normal one.Conclusion\ This model owned the characters of mild impaired islet function and insulin resistance which resembled the clinical phenotype of type 2 diabetes,and it would help deepen the mechanical investigation of diabetes and its chronic complication.

    • Seasonal Variation Influences on Reproductive Functions in Female Rhesus Monkey

      Abstract:Objective To study the seasonal variation influences on reproductive functions in female rhesus monkey. Method\ Seasonal biological variations in skins,menstrual cycle,level of hormones were assasyed by random sampling of statistics method and radioimmune technique.Results\ 1.The periodic changes in sexual characteristics were demonstrated,including the sexual skins and normal menstrual cycles of all female rhesus monkeys during the breeding season.During the non\|breeding season some female rhesus monkeys displayed the sexual skins and abnormal menstrual cycles,a few female rhesus monkeys showed no menstrual cycles for a longer time.2.Gonadotropin and steroidogensis displayed significant cyclical secretion in the breeding period,while no significant cyclical secretion in the non\|breeding season.Conclusion\ It is suggested that female rhesus monkeys show remarkable sexual skins,menstrual cycles and hormones change between the breeding and non\|breeding seasons,which could lead to different reproductive functions of female rhesus monkey.

      Abstract:目的 观察运行中清洁级实验动物设施的环境指标 ,指导科学实验和设施管理。方法 在不同季节、不同动物饲养量和不同管理情况下 ,按国家标准GB T 14 92 5 94测试相关环境指标。结果 全年温度 2 4~ 2 5℃ ,相对湿度 4 0 %~ 70 % ,换气次数 14 .5~ 17.0次 h ,空气洁净度 60 0 0~ 2 0 0 0 0级 ,梯度压差 2 2~ 2 8Pa ,噪声 3 9~ 4 8dB ,落菌数 1.4~ 4 .9个 皿 ,氨浓度 0 .4~ 3 .8mg m3,各项环境指标稳定 ,但个别指标未达到国家标准。结论 不同季节大气尘浓度的变化对清洁级实验动物设施的环境指标无影响 ,严格管理可保持动态动物实验室的洁净度

      Abstract:Objective To construct the retroviral recombinant plasmid vector encoding TK gene of the herpes simplex virus.Methods\ The specific primers were designed and synthesized to amplify the DNA fragment (PCR method) of HSV1 tk gene from the plasmid pHSV106. The length of the fragment was 1168bp.After digested by appropriate restriction endoenzymes(BamHI and EcoRI),the fragment of HSV1 tk gene was cloned into pLXSN according to its special orientation.Then the recombinant plasmid was identified by digesting of endoenzymes,PCR and DNA sequencing.Results\ The fragments digested by endoenzymes and amplified by PCR method were as large as the predicted results.DNA sequencing was the same with the sequence reported on the literature.Conclusion\ The retroviral recombinant plasmid pLXSN TK was successfully constructed.

      Abstract:目的 研究心气虚病证动物模型及其评价体系的构建。方法 移植心肌梗塞致心力衰竭大鼠的制作模型 ,运用中西医结合虚证和血瘀证的全国统一诊断标准、将其定性的问诊内容代以定量的同等意义的指标测试进行心气虚证动物模型评价。结果 本研究制作的动物模型再现了从一个正常大鼠→以血瘀为主要损伤→形成心气虚证的过程 ;心气虚证动物模型具有时相性、功能性和与功能相关的组织结构物质性改变。结论 将中西医临床和或基础研究中规范的、成熟的、统一的方法和标准引入中医动物模型的研究 ,并注重吸取中医已取得的临床经验和研究成果 ,是指导中医病证动物模型建立的基本思路

      Abstract:目的 研究天然配比油对家兔肝组织中脂肪酸的影响。方法 用GC法测定 5 0只家兔喂天然配比油 90d肝组织脂肪酸组分和含量的变化。结果 猪油对照组中 14∶0、18∶1和MUFA明显增高 ;三个配比油组α18∶3水平明显增高 ,14∶0明显降低 ,配 1组的α18∶3含量最高 ,配 2和配 3组的γ18∶3和 18∶2含量明显增高、18∶1和MU FA无明显变化。结论 补充三种配比油均可提高体内n3系多烯脂酸水平 ;配比 2和配比 3油还可提高n6 系多烯脂酸质量、抑制 18∶1升高 ,治疗高脂血症效果较好

      Abstract:Objective\ To compare the biological characteristics of H 22 cell lines from different institutes. Methods\ H 22 cell lines from three institutes,that is,Beijing H 22 from Institute of Pharmacology of Chinese Medical Academy,Shandong H 22 from Institute of Pharmacology of Shandong Medical Academy and Wuhan H 22 from China Center for Type Culture Collection in Wuhan,were kept in KM mouse abdominal cavity,then their comparative analyses of culture behavior in vitro, growth feature in vivo, pathomorphism and therapeutic outcome were carried on. Results\ The growing speeds of three cell lines in vitro and in vivo were different markedly.The growing speed of Wuhan H 22 was fastest ( P<0.01 ) with the best consistency.Shandong H 22 could inoculate successfully at lowest cell concentration of 10\ 4 cells per mouse.The tumor conformations of different H 22 were different.The mice bearing H 22 tumor were treated with CTX, showing the highest neoplastic inhibition rate (74.6%)in mice with Shandong H 22 . Conclusions\ The biological characteristics of H 22 from different institutes were markedly different,though they were of the same origin.\;

      Abstract:目的 对L12 10细胞及其 4株克隆细胞建立的肿瘤动物模型的某些生物学特性进行比较研究 ,从中筛选出基本符合L12 10细胞生物学特性且一致性更好的克隆细胞。方法 北京肿瘤所L12 10细胞和 4株克隆细胞腹腔接种DBA 2小鼠 ,观察产生腹水的性质、腹水瘤细胞浓度和致小鼠死亡时间 ;皮下接种DBA 2小鼠 ,观察瘤块的生长情况 ;腹腔注射化疗药物环磷酰胺 (CY) ,比较对CY治疗的敏感性。结果 腹腔接种DBA 2小鼠均能生长腹水 ,其中克隆细胞L2H8、L3B12产生血性腹水 ,L3F9的腹水微血性 ,L3E11与肿瘤所L12 10细胞为无血性腹水 ;腹水瘤细胞的浓度及小鼠生存时间亦有差别。肿瘤所L12 10细胞及克隆细胞L3F9、L2H8、L3E11、L3B12第 10天瘤重依次为 1 9± 0 4 6、1 5± 0 3 8、0 75± 0 5 2、2 6± 0 3 0、2 0± 0 3 3g ;用CY治疗的抑瘤率分别是 4 8 7% ,81 3 % ,86 0 % ,78 7%及 67 1%。结论  4株克隆细胞的生物学特性基本符合L12 10细胞 ,对化疗药物CY的敏感性均高于L12 10细胞

    • Comparison of Cardiac Function in Different Animal Models with Cardiac Failure

      Abstract:Objective\ To compare the characteristic of cardiac function in two different congestive heart failure animal models.Methods\ To establish different congestive heart failure animal models with abdominal aortocaval shunts and ligature of the left coronary artery respectively was established in rats. The cardiac function test was with Doppler echocardiography.Results\ All the rats in both groups had developed cardiac hypertrophy.Ejection fraction (EF) of rats with aortocaval shunts decreased,but cardiac output (CO) and blood pressure (BP) in this group maintained normal.While EF,CO and mean artery pressure (MAP) all reduced significantly in ligation rats,IRT increased in this group.Conclusions\ Abdominal aortocaval shunts leads to high output cardiac failure.Cardiac failure resulted from ligation of coronary artery is low output,which is more severe.Doppler echocardiography provides a simple,reliable easy\|follow\|up way to test cardiac function in rat.\;

    • Establishment and Evaluation of the Model of the Sciatic NerveFirearm Injury in Rabbit

      Abstract:Objective To establish and evaluate the model of the sciatic nerve fire\|arm injury of rabbit.Methods According to the mass of steel ball and the charge, 26 rabbits were divided into 5 groups at random, 0.38g steel balls were used in the groups A,B and C, whose charges were 0.35?g,0.65g and 1.20g respectively; 1.03g steel balls were used in the groups D and E, whose charges were 0.35g and 0.65g respectively. The target was the midpoint of the lateral body surface projection of the right sciatic nerve. Velocity,transmission energy,wound tract dimension,occurrence of fracture and blood vessel injury,length of the injured sciatic nerve and the pathologic changes of the injured sciatic nerve were observed.Results The steel ball velocity of groups A,B,C,D,E were 621 6,939 8,1299 8,594 6 and 816 1m/s respectively. The transmission energies of groups C and E were 183\^2 and 165\^2J respectively, which were too high; the transmission energies of groups A and D were 18\^7 and 44\^2J respectively, which were too low; The transmission energy of group B was 80\^0J, which was well suitable. More fracture and blood vessel injury were found in groups C and E. Typical sciatic nerve pathologic changes were found in groups B,C and E. Conclusion Under the condition of group B, typical sciatic nerve fire\|arm injury could be gained easily, and the long\|term pathological changes of the injured nerve could be observed.

      Abstract:目的 观察长爪沙鼠精子的形态和结构。方法 应用光学显微镜、透射电镜和扫描电镜进行观察。结果与结论 长爪沙鼠精子由头、颈、尾三部分组成 ,全长为 ( 15 6 3 2± 6 61) μm ,头长为 ( 10 4 3± 2 12 ) μm ,颈、尾部长为 ( 14 5 89± 5 14 ) μm ,具典型的啮齿类动物的精子形态 ;长爪沙鼠精子的尾部轴丝是 9+ 2模式 ,轴丝外围有 9条大小、形态不一的外周致密纤维 ,外周致密纤维从主段到末端逐渐变细 ,并在主段的近末端处分 4批终止 ;主段的纤维鞘具有腹侧纵柱、背侧纵柱、内嵴等结构。在精子尾部的末段 ,外层仍保留有纤维鞘的结构 ,但无外周致密纤维 ,轴丝仍然为 9+ 2模式

    • Breeding and Examination of C57BL/6J-HBV Transgenic Mice

      Abstract:Objective To find out the breeding rule of C 57 BL/6J-HBV transgenic mice and to establish a credible examination method of HBsAg. Methods Breeding characterization and expression of HBsAg of C 57 BL/6J-HBV transgenic mice in two different matings were compared. The nicety of serum diagnosis of HBsAg was conformed by immunohistochemistry, and the localization of expressed products in the liver cells was determined by laser scanning confocal microscopy.Results and Conclnsion The litter number of two different mating methods is nonsignficant( P >0.05),however the weaning number is significant(0.01< P <0.05).Positive rate of intercross mice is higher than that of backcross mice. Laser scanning confocal microscopy determines that HBsAg is expressed in the cell membrane and cytoplasm.
