Abstract:目的 使用COBE Spectra血液成分分离机大量采集实验猕猴外周血单核细胞(PBMCs),继而可从中分离得到外周血造血干细胞(PBSC),为进一步利用猕猴开展免疫学研究、基因治疗提供足够的目的 细胞,探讨采集的关键技术,建立安全、有效的采集方法.方法 体重为4-5奴的实验猕猴5只,采集前一个月内分3次进行自体或异体血液于4℃储备共120 mL,用于采集时填充管路.5只猴采集前接受rhGM-CSF 20μg/ks皮下注射动员4～5 d,麻醉动物后行股动脉穿刺,选择自动外周血干细胞收集程序(Auto-PBSC)进行采集.采集结束后管路中血液以10 mL/min回输给动物3～5 min.结果 生长因子连续注射第4天外周血白细胞数增至最高,收获细胞数量随循环血量和采集次数增加而增多.经动员的所有猴能够采集到需要的PBMC,最多达9.9×108,采集次数1～3次,循环血量达750～1420 mL,实验结束后1只猕猴因心脏衰竭死亡.结论 人用血细胞分离机可用于4～5 kg实验猕猴PBMC的大量采集.由于动物不同于人体,为保证采集成功需要选用适合于猕猴的程序,采集前做好储血和生长因子动员准备,稳定的麻醉保定,提高抗凝剂比例,积极处理并发症是关键.

Abstract:目的 探讨兔眼增殖性玻璃体视网膜病变模型的建立方法.方法 ①体外培养兔眼视网膜色素上皮细胞;②兔眼3组,每组6只,分别在玻璃体内注射0.1 mL的生理盐水、1×106细胞及2×106细胞,在不同时间段进行裂隙灯显微镜、间接检眼镜、眼底照像和B超检查,观察成模情况.结果 注射后28 d,生理盐水组成模0眼;1×106细胞组成模5眼,其中I级眼1只,Ⅱ级眼3只,Ⅲ级眼1只;2×106细胞组成模6眼,其中Ⅱ级眼2只,Ⅲ级眼4只.结论 兔眼玻璃体内注射2×106同种视网膜色素上皮细胞建立增殖性玻璃体视网膜病变模型,符合病变发展规律,而且稳定可靠,成模较快,简单易行.

Abstract:目的 建立一种操作简便、性能稳定的新生大鼠高氧肺损伤动物模型.方法 ①设计制造能自动控制氧浓度的高氧动物饲养舱.②将孕期满21 d出生的SPF级新生大鼠随机分成4组,即:高氧组Ⅰ(入高氧舱中饲养1-7 d),高氧组Ⅱ(人高氧舱中饲养14 d),以及相应的空气对照组Ⅰ、Ⅱ,每组14只.舱内氧浓度≥90%,每天23 h.计算肺系数,HE染色与病理学观察.结果 高氧组与相应的对照组比较:高氧组大鼠体重轻,肺系数大,HE染色显示部分肺泡萎缩、肺间质及肺泡腔有水肿、出血,中性粒细胞浸润;肺泡发育明显滞后,辐射状肺泡计数明显少.结论 本动物饲养舱性能稳定,操作简便;复制的新生大鼠的肺病理变化符合高氧肺损伤的改变.
