Abstract:目的评价聚羟基脂肪酸(polyhydroxyalkanoates,PHA)、聚乳酸(polylacetic acid,PLA)和聚己内酯(polycaprolactone,PCL)三种膜性高分子材料在兔眼部的生物相容性。方法将24只新西兰兔随机分为4组,每组6只。PHA、PLA、PCL为实验组,材料植入兔右眼结膜下。假手术组结膜下钝性分离,但不植入任何高分子材料。使用裂隙灯显微镜观察并记录植入后不同时间手术眼的反应并评分。裂隙灯下观察材料的吸收时间。术后4周和16周取眼球,行HE染色、Masson染色和天狼猩红染色分别定性观察组织结构和炎症细胞、胶原纤维和胶原纤维的亚型与排列方向。结果术眼刺激性评分等级各组均不高于轻度刺激性。结膜下吸收时间PHA、PLA和PCL组分别是16周,12周和大于16周。组织学观察术后4周PHA、PLA和PCL组均形成材料包裹囊腔,囊壁以纤维组织为主,伴有毛细血管形成和炎性细胞浸润,以中性粒细胞为主。胶原纤维染色与假手术组无明显差异,以Ⅰ型和Ⅲ型为主,大致呈平行排列。术后16周PHA和PLA组材料已不可查及,包裹囊腔结构不规则,而PCL材料整体可查及,包裹囊腔规则。各组未见毛细血管,...

Abstract:目的研究Müller细胞在大鼠视网膜绿光损伤模型中的激活反应。方法 72只出生后8周(约230~280g)的成年雄性Sprague-Dawley大鼠随机分9组。一组作为正常对照,另8组暗适应24 h后置于波长为(530±10)nm的LED灯光箱中照射3 h,并分别于暗恢复3、6、12 h及1、2、3、7、15 d取眼球,光学显微镜下观察同一定位处视网膜组织形态、检测(TUNEL)凋亡细胞、免疫组织化学染色及蛋白免疫印迹分析。结果光损伤后光感受器内外节变短,外核层变薄,细胞排列紊乱;细胞凋亡出现在外核层,暗恢复3 h出现,3 d时达到高峰,7 d时消失;胶质细胞纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达量于暗恢复12 h开始升高,随暗恢复时间延长逐渐升高;谷氨酰胺合成酶(glutaminesynthetase,GS)总表达量未见明显改变,但可见蛋白一过性重分布,表达部位由内核层移至外核层。pSTAT3蛋白表达量于损伤后12 h出现一过性升高。结论绿光损伤视网膜激活Müller细胞,pSTAT3可能参与了此激活过程。

Abstract:目的研究不同浓度消旋山莨菪碱液对雏鸡形觉剥夺近视眼轴的影响。方法 3日龄雏鸡60只右眼遮盖半透明塑料膜,随机分为6组:给药组4组,不同浓度消旋山莨菪碱液(0.05%组,0.1%组,1%组,2.5%组),隔天行右眼球结膜下注射,每次给药50μL,共4次;对照组为生理盐水组,注射方法同给药组;单纯形觉剥夺组,不做干预。8 d后观察药物对眼轴的影响。结果 2.5%浓度组右眼与左眼眼轴差异显著小于低浓度组及对照组(P<0.05),给药组未发现组织病理毒性。结论消旋山莨菪碱对雏鸡近视眼轴增长有一定抑制作用,无毒副作用,有一定浓度依赖性。

Abstract:目的建立小鼠结核分枝杆菌耐利福平株静脉感染小鼠模型。方法 30只BALB/c雌性小鼠经尾静脉感染结核分枝杆菌耐利福平株每只106CFU,观察小鼠一般状况,并分别在感染后6周、10周、14周处死小鼠,进行脾、肺组织病理切片、抗酸染色、计脏器荷菌数。结果感染后小鼠体重呈逐渐上升趋势,脏器病理变化随时间推移由急性炎症逐渐转为慢性炎症反应,抗酸染色均为阳性,脏器荷菌数与感染剂量相对应,并至少可持续14周。结论成功建立了小鼠结核分枝杆菌耐利福平株静脉感染动物模型,为其进一步用于结核病防治研究奠定了一定的基础。

Abstract:目的检测转化生长因子-β1(transforming growth factor-β1,TGF-β1)对体外培养大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cell,BMMSCs)增殖及Slug表达的影响。方法采用密度梯度离心结合贴壁法分离、培养大鼠BMMSCs,用免疫组织化学方法对培养第3代的细胞进行鉴定。用MTT法检测不同浓度TGF-β1对细胞增殖的影响;免疫荧光和免疫印迹法检测TGF-β1处理前后Slug的表达情况。结果密度梯度离心结合贴壁法能有效分离、纯化大鼠BMMSCs,免疫组织化学法检测显示CD29、CD44表达阳性,而CD34、CD45表达阴性;低浓度TGF-β1对BMMSCs的增殖有促进作用,高浓度却抑制BMMSCs的增殖。TGF-β1处理24 h,Slug蛋白表达明显增强。结论一定浓度的TGF-β1可以促进BMMSCs的增殖,而且引起Slug蛋白增加。
