    目的 系统评估长期食用转 cry1Ab / cry2AjG10evo-EPSPS 基因玉米(双抗 12-5 玉米)对食蟹猴多代生殖与子代(包括 F1 代和 F2 代)早期发育的安全性。 方法 将 36 只性成熟食蟹猴( F0 代,6 雄 30 雌)随机分为常规饲料(CK)组、转基因玉米(GM)组及非转基因亲本对照玉米(Non-GM)组,按 1 ∶ 5 雄 ∶ 雌 进行后宫式繁殖。持续记录 F0 与 F1 代的生殖性能(怀孕、流产、出生及出生后死亡数),并对 F1 及 F2 代仔 猴进行新生仔猴外观、神经行为功能发育、体格发育及血液生理生化指标的系统检测。 结果 F0 代 GM 组怀 孕胎次(32 次)与 CK 组(26 次)、Non-GM 组(29 次)无显著差异P>0. 05);F1 代总体存活数(GM 组 19 只、 CK 组 20 只、Non-GM 组 14 只)组间差异无统计学意义(χ2 = 3. 141,P= 0. 237)。 F1 代繁殖性能及 F2 代存活情况组间同样未见显著差异(P= 1. 000)。 所有子代新生仔猴外观均未见畸形,神经行为功能发育评分、体格生长趋势及血液基础生理指标(24 项血常规、18 项血清生化)在 3 组间均无显著性差异P>0. 05)。 结论 长期食用双抗 12-5 玉米未对食蟹猴 F0 代生育能力及子代早期发育产生不良影响,为该类生物育种产品的食用安 全性提供了重要的非人灵长类动物实验依据。

    Abstract:

    Objective To systematically evaluate the multi-generational reproductive and developmental safety of long-term consumption of genetically modified maize expressing cry1Ab / cry2Aj and G10evo-EPSPS protein (double-resistant 12-5) in cynomolgus monkeys. Methods Thirty-six sexually mature cynomolgus monkeys ( F0 generation, 6 males, 30 females) were divided randomly into conventional diet (CK), genetically modified maize (GM), and non-genetically modified parental maize (Non-GM) groups. Animals were bred in a harem system at a male-to-female ratio of 1 ∶ 5. The reproductive performances of the F0 and F1 generations were recorded, and F1 / F2 offspring were assessed systematically for fetal appearance, neurobehavioral development, physical growth, and hematological / serum biochemical parameters. Results There was no significant difference in F0 pregnancy rates (GM group: 32, CK group: 26, Non-GM group: 29; P>0. 05) or overall F1 offspring survival (GM group: 19, CK group: 20, Non-GM group: 14; χ2 = 3. 141, P= 0. 237) among the groups. There was also no significant difference among the groups in terms of reproductive performance of the F1 generation or survival of the F2 generation ( P = 1. 000). No fetal malformations were detected in any offspring. Behavioral neurodevelopmental scores, physical growth trends, and baseline physiological parameters (including 24 hematological and 18 serum biochemical indices) showed no differences among the three groups. Conclusions Long-term consumption of doulde-resistant 12-5 maize had no adverse effects on parental fertility or early offspring development in cynomolgus monkeys, providing important non-human primate evidence for the safety of this genetically modified crop.
