    目的 本研究旨在培育载脂蛋白 E(ApoE) 基因敲除兔,探究其表型特征及繁育性能,建立 ApoE 基因敲除兔的育种技术体系。 方法 采用 CRISPR/ Cas9 基因编辑技术和胚胎移植技术构建 ApoE 基因 敲除兔,通过基因型筛选健康的 ApoE 基因敲除兔作为种兔,结合近交或回交方式进行选种扩繁,对 ApoE 基因敲除兔进行血常规、血脂生化检测和病理检查表型分析,并评估其繁殖性能,从而筛选出遗传稳定且表型明显的 ApoE 基因敲除兔。 结果 成功获得 ApoE -/-兔群体,正常饮食下,其总胆固醇(TC)和低密度脂蛋白胆固醇 (LDL-C)显著高于新西兰兔(WT)(P<0. 01),表现出脂质代谢紊乱和动脉粥样硬化的病理特征,其繁殖性能及生长发育与新西兰兔(WT)无显著性差异。 结论 本研究成功制备并筛选出遗传稳定、表型明显的 ApoE 基因敲除兔,建立了有效的育种技术体系,为心血管及相关代谢疾病的机制研究提供了重要的动物模型。

    Abstract:

    Objective To develop apolipoprotein E ( ApoE) gene-knockout rabbits, investigate their phenotypic characteristics and reproductive performance, and establish a breeding technology system for ApoE geneknockout rabbits. Methods ApoE gene-knockout rabbits were constructed using CRISPR/ Cas9 gene-editing technology with embryo transplantation. Healthy ApoE gene-knockout rabbits were screened by genotyping to serve as breeding stock and their selection and expansion were conducted by inbreeding or backcrossing. Phenotypic analysis included routine blood tests, lipid biochemical assays, and pathological examination. Reproductive performance was also evaluated to select genetically stable ApoE gene-knockout rabbits with distinct phenotypes. Results An ApoE -/-rabbit population was successfully established. Under a normal diet, total cholesterol and low-density lipoprotein cholesterol levels were significantly higher in ApoE -/- than in New Zealand rabbits ( WT) ( P< 0. 01), exhibiting pathological features of lipid metabolism disorder and atherosclerosis. There were no significant differences in reproductive performance and growth development between ApoE -/- and New Zealand rabbits (WT). Conclusions This study successfully produced and selected genetically stable and phenotypically distinct ApoE gene-knockout rabbits, establishing an effective breeding technology system. These rabbits provide an important animal model for studying the mechanisms of cardiovascular and related metabolic diseases.
