    目的 比较灵芝、枸杞、云芝、黄芪、香菇五种植物粗多糖对 C57BL/ 6 小鼠骨髓来源树突状细胞 ( bone marrow-derived dendritic cells, BMDC)的体外诱导功能。 方法 采用 GM-CSF 体外诱导和培养 C57BL/ 6 小鼠 BMDC 细胞,体外培养 6 d 后,分别加入灵芝、枸杞、云芝、黄芪和香菇五种植物粗多糖进行培养,24 h 后,采用流式细胞术检测细胞表面 CD80、CD86 及 MHCII 分子的表达,采用 ELISA 检测 BMDC 细胞培养上清中细胞因子 IL-6、IL- 12 和 TFN-α 的含量。 结果 云芝多糖可显著诱导 BMDC 细胞表明的 CD80、CD86 和 MHCII 分子表达上调,并刺激 BMDC 细胞释放 IL-6,IL-12p40 和 TFN-α。 枸杞多糖、灵芝多糖和香菇多糖仅显著性的诱导 IL-6 释放增加。 结论 云芝多糖对 BMDC 具有较强的诱导作用,具有潜在的疫苗佐剂活性。

    Abstract:

    Objective To investigate the adjuvant activity of polysaccharides from Ganoderma lucidum, Lycium barbaru, Versicolor, Astragalus and Lentinan on bone marrow-derived dendritic cells ( BMDCs) from C57BL/ 6 mice. Methods Bone marrow cells from C57BL/ 6 mice were cultured in the presence of granulocyte-macrophage colony- stimulating factor GM-CSF. After 6 days, immature BMDCs were collected and co-cultured with the polysaccharides of G. lucidum, L. barbaru, Versicolor, Astragalus or Lentinula. CpG oligodeoxynucleotides were used as a positive control. The cell surface expression of CD80, CD86 and MHCII were detected by fluorescence-activated cell sorting analysis. The release of cytokines interleukin (IL)-6, IL-12 and tumor necrosis factor TFN-α were measured by enzyme-linked immunosorbent assay. Results Versicolor polysaccharides significantly upregulated the surface expression of CD80, CD86 and MHCII on BMDCs, as well as the release of the cytokines IL-6, IL-12 and TFN-α compared with the control group. Conclusions Versicolor polysaccharides promoted the maturation of BMDCs in vitro and may have potential as an adjuvant for vaccine preparations.
