Selective Elimination of CD8+Lymphocytes from Rhesus Macaques of Chinese Origin Using Anti-CD8+ Monoclonal Antibody

    Abstract:

    Objective To identify the effectiveness of McAb(cM-T807) anti-huCD8~+T cell on eliminating the CD8~+ lymphocytes from Chinese original rhesus monkeys.Methods The anti-CD8~+T cell McAb was injected into two rhesus monkeys at 0 d,3 d and 7 d,respectively.Peripheral blood and lymph node samples were collected from the two monkeys at different time after injection.The CD3/CD4/CD8 from both peripheral blood lymphocytes(PBL)and lymph nodes(LN) cell suspension were differentiated by FACS and the effectiveness of elimination of CD8~+ lymphocyte was detected.Results The results showed that 99.9% and 90.0% reduction of CD8~+cells in peripheral blood and lymph nodes from two Chinese-origin rhesus macaques were detected from 24 and 168 hours after injection,respectively,and the duration of low CD8~+ levels maintained for 27 days in PBL and in LN for 21 days.Conclusions CD8~+T cells can be eliminated effectively from PBL and LN in Chinese original rhesus monkeys by injection of McAb cM-T807.
