    摘要:

    目的 采用受精卵显微注射方法获得的3头hEPO转基因山羊进行检测和整合状况分析。方法 用荧光原位杂交的方法,以正常羊及正常人染色体标本为对照,通过生物素标记的探针与转基因羊染色体标本进行杂交。结果经信号放大及DAPI染色后,3头转基因羊分别在不同染色体上各有一对明显的荧光信号,而对照组无信号。结论hEPO基因在转基因山羊染色体上的整合是单位点整合,分别整合在不同的染色体上。

    Abstract:

    Objective To determine the hEPO transgene on the chromosomes of three transgenic goats obtained by zygote microinjection.Method\ Fluorescence \%in situ\% hybridization technique was used to detect the hEPO transgene.The biotin labeled probe was hybridized with the chromosomes of transgenic goat,normal control goat and human's.Results\ By signal amplification and DAPI staining,one pair of signals could be observed obviously on different chromosomes of these transgenic goats,while no signals produced on normal goat and human chromosomes.Conclusion\ hEPO transgenes were integrated into the chromosomes of these three goats with single site,and the integration was random on the goat chromosomes.
