    摘要:

    目的 建立抗精子抗体 (AsAb)阳性大鼠动物模型。方法 同种Wistar大鼠精子抗原人工免疫法建立模型 ,通过ELISA法检测血清AsAb ;光镜HE染色观察大鼠附睾组织形态学改变 ;考马斯亮蓝顶体染色法检测精子的顶体反应率 ;化学比色法检测精子的ATP酶活性。结果 模型AsAb阳性大鼠血清样品的AsAb符合实验要求 ,其精子的顶体反应率、Na+ -K+ ATP酶、Ca2 + -ATP酶活性与对照组相比均显著降低 (P <0 0 1;P <0 0 5 ;P <0 0 5 )。结论 成功建立抗精子抗体阳性大鼠动物模型。

    Abstract:

    Objective To establish an animal model of positive antisperm antibody (AsAb) in rats. Methods The animal model was set up by artificial immunity with iso-Wistar rat sperm antigen. The level of AsAb in serum was determined by ELISA. The morphological changes of the epididymis tissue in rats were determined by optical microscope with HE. The rats sperm was visualized by staining acrosome with the dye Coomassie brilliant blue. The changes of ATPase activity of the rats sperm were detected by chemical colorimetry. Results The AsAb of rats serum in the model accorded with the experimental demand. The percentages of acrosome reaction, Na -K ATPase activity, Ca 2-ATPase activity in AsAb positive rats sperm significantly decreased as compared with the control group (P<0.01; P<0.05; P<0.05).Conclusion The results demonstrated it was successful to establish an animal model of AsAb positive rats.
