    本文综述了湖北省农科院生物技术研究所有关转基因猪研究的主要进展。先后将POMT—PGH,PSV40-PGH,POMT-PGH-Micro satellite DNA,PMHR,hDAF,pBHSA,pSHSA等7种不同结构的外源基因,注入4027枚猪的受精卵原核,获得原代表达的转基因猪83头,转基因的效率在0.9%~3.4%之间,平均2.2%。通过几个主要技术环节的改进,提高了转基因的效率;建立了外源基因整合的定性检测(PCR,Dot blot和Southern blot)和定位检测-染色体原位杂交技术;进行了相应外源基因的蛋白质表达检测;研究了转基因猪的遗传规律。

    Abstract:

    This paper summarizes recent progress in transgenic swine in Biotech Institute of Hubei Academy of Agriculture Science. The Seven individual foreign genes with different construction, that is, POMT-PGH, PSV40-PGH, POMT-PGH-Micro satellite DNA, PMHR, hDAF, pBHSA, pSHSA, was microinjected into 4027 embryos. 83 founders were produced. Transgenic rate was 0.9 to 3.4% and 2.2% on average. Through improving main technological process, we improved transgenic rate, established the technology of hybrid in situ of chromosome, detection of foreign gene quantity by PCR and Dot blot and Southern blot and Northern blot for expression protein, investigated the genetic regulation of transgenic swine.
