Model Foundation on Rat Osteosarcoma Using Orthotopic Implantation and Observation on Its Biological Characteristics

    目的建立大鼠UMR106骨肉瘤细胞株的同种异体原位移植模型。方法采用完整瘤组织块原位移植到大鼠右胫骨骺端,分不同时期处死,观察其生长及转移特性。同时检测其血清中ALP水平的变化。结果肿瘤原位移植成功率为91.67%,移植后1.2周肿瘤从髓内向髓外生长,3周骨皮质破坏侵犯软组织但无肺转移,4周肺部可见少量小转移灶,5周骨皮质破坏更加明显,肺转移灶明显增加,6—7周大鼠双肺广泛转移因呼衰而死亡。ALP水平1.2周逐渐升高,3周时达最高峰,4.6周逐渐下降,7周接近一平台期,但仍高于同期正常水平。结论该模型成功率高,成本低,实用性强,模拟了人类骨肉瘤临床发病和发展过程,可作为研究骨肉瘤较理想的模型。

    Abstract:

    Objective To establish an osteosarcoma model of rat and to study its biological characteristics.Methods Intact tumor pieces were inoculated into the right tibia of rat,the rats were killed in different time and observed the growth of tumor and metastasis.At the same time,serum alkaline phosphatase was detected.Result The tumor growth rate was 91.67%,from the first week to the second week,the tumor grew out from medullary cavity,then it widely destroyed cortical bone and invaded soft tissue on third week but there was no pulmonary metastasis.Pulmonary metastasis took place on the fourth week,on the fifth week the cortical bone was destroyed more obviously than before and the quantity of pulmonary metastasis increased,and the rat finally died for respiratory failure on the sixth or seventh week.The level of ALP began to step up from the first week to the second week,reached a peak on the third week.It fell off from the fourth week to the sixth week,and approached a platform on the seventh week which was still higher than normal level of same time.Conclusion The tumor growth rate is high,and the animal is available easily.So the model can be widely used for scientific research.The model simulated the clinical process of human osteosarcoma,and could be a satisfactory model for studying osteosarcoma.
