    目的制备来源于巴马香猪皮肤的无细胞真皮,研究其生物学特性,为其异种无细胞真皮应用于烧伤及创伤修复临床形成积累基础研究资料。方法研究两种不同的酶(0.1%dispaseII和0.25%胰蛋白酶)处理去表皮,结合去污剂0.5%Triton X-100处理不同时间制作无细胞真皮的方法,随后对不同方法制作的无细胞真皮进行常规石蜡切片,显微镜下观察其结构,并进行微生物检测和细胞毒性检测。结果胰蛋白酶法去除表皮彻底,且基底膜保留完整,dispaseII也能完整地去除表皮,但对基底膜有破坏;0.5%Triton X-100室温下振荡处理24h能完全去除真皮中的细胞成分及皮肤附属器且对皮肤胶原结构没有破坏;在所用试剂中加入0.02%的NaN3能够起到完全抑菌的作用;制作的无细胞真皮无细胞毒性。结论无细胞真皮内已经完全没有细胞成分及皮肤附属器,抗原性显著降低,无细胞无毒性,可替代异体无细胞真皮基质作为创伤修复的真皮支架。

    Abstract:

    Objective To prepare BaMa minipig's acellular dermal substitute in order to ultilize it into xeno-transplant in clinical use.Methods Epidermis of BaMa minipig's skin was removed by 0.1% dispaseII or 0.25% typsin for one night,then dermis was treated with 0.5% triton X-100 for 24h,succussed or not succussed.The prepared acellular dermal matrix was examined macroscopically with gross observation,light microscopy,bacteris culture test and cytotoxicity test.Results Epideris could be removed thoroughly either in 0.1% dispaseII or 0.25% typsin for one night,but the basal memberane was integrity in 0.25% typsin and in 0.1% dispaseII the basal memberane was not integrity.Cellular components were elimated thoroughly treated by succuss in 0.5% triton X-100 for 24h.There were not any bacteria and ctyotoxity.Conclusion Prepared BaMa minipig's acellular dermal substitute had no cellular components,bacteria and cytotoxity,which would be an ideal dermal substitute.
