    摘要:

    目的探讨谷氨酸转运蛋白GLAST、GLT-1不同亚型在癫痫发作过程中的表达变化。方法采用TaqMan探针实时定量PCR(real-time quantitative PCR)技术,检测癫痫发作后不同时点脑组织中谷氨酸转运蛋白GLAST、GLT-1表达量的变化。结果癫痫发作后不同时点GLAST、GLT-1在大脑中的表达量明显低于对照组(P〈0.01),且在大脑中不同时点的表达量具有明显的差异(P〈0.01);GLAST在小脑中不同时点的表达量无明显差异(P〉0.05),但均明显低于对照组(P〈0.01);GLT-1在小脑中无表达。结论癫痫发作后谷氨酸转运蛋白GLAST、GLT-1在大脑中的表达量明显减少;GLAST、GLT-1表达量的减少可能是癫痫发病的诱因之一。

    Abstract:

    Objective To investigate the expression of GLAST and GLT-1 in rats with epilepsy.Methods Glutamate transporter GLAST and GLT-1 genes expression were detected by real-time fluorescent quantitative PCR assay with TaqMan probe in different time after epileptic seizure.Results The expression amount of GLAST and GLT-1 in cerebrum were obviously lower than that of the control in different time after epileptic seizure(P < 0.01).Furthermore,the amount of expression had evidently difference in cerebrum in different time after epileptic seizure(P < 0.01).The expression amount of GLAST in cerebellum was not significantly different in different time after epileptic seizure(P> 0.05),but they were obviously lower than that in the control(P<0.01).However,GLT-1 was not expressed in cerebellum.Conclusion The expression amount decrease of GLAST and GLT-1 may be one of the factors of epileptic seizure.
