    目的 建立豚鼠的甲流感染模型并评价激素甲强龙对感染的干预作用。方法 将 4 ~ 6 周龄 雌 性SPF 级豚鼠(200 ~ 300) g 分为 4 组:正常对照组,模型组,甲强龙 1 组和甲强龙 2 组,除对照组外,豚鼠经乙醚麻醉后进行滴鼻接种 A /California /7 /2009(CA7)病毒,分别于攻毒后 3 d 和 5 d 给予激素甲强龙,大剂量 3 d 后 1 /4 剂量给 3 d,检测豚鼠感染的多项指标,观察期为 14 d。结果 成功建立了豚鼠的甲流感模型;甲 强 龙 1 组 豚 鼠 肺 部 炎症减轻较模型组和甲强龙 2 组明显,存活率较这两组均降低,甲强龙 2 组肺部炎症较模型组减轻,而较甲强龙 1 组重,豚鼠的存活率较这两组均无差异。结论 豚鼠能感染 A /California /7 /2009(CA7)病毒,感染后 5 d 比感染后 3 d开始给予激素的干预效果好。

    Abstract:

    Objective To establish a guinea pig model of influenza A /California /7 /2009(CA7) virus infection and evaluate the intervention with hormone methylprednisolone for viral pneumonia due to this infection. Methods Female Hartley strain guinea pigs ( 200-300 g ) were divided into four groups: control,model,methylprednisolone group 1 and methylprednisolone group 2. The animals were anesthetized by inhalation of ether and inoculated intranasally inoculated with 0. 3 mL of virus CA7. Methylprednisolone was administered beginning at postinfection day 3 or 5,recommended dose lasting for 3 days and then quarter of recommended dose for another 3 days. The animals were observed daily for 14 days for clinical signs of infection and survival. Results The guinea pig model for A /California /7 /2009(CA7)virus infection was established successfully. The lung inflammation of methylprednisolone group 1 was alleviated compared with that of both model group and methylprednisolone group 2,but the survival rate was lower compared with that of both two groups. The lung inflammation of methylprednisolone group 2 was alleviated in comparison with that in the model group but aggravated compared with that of methylprednisolone group 1,while the survival rate was not affected. Conclusion Guinea pigs can be infected with influenza A virus A /California /7 /2009,and it is better to administer methylprednisolone on dpi 5 than dpi 3.
