    目的 观察不同年龄自发性高血压大鼠( SHR) 心脏 AT2R 的表达水平及心肌胶原含量,探 讨 AT2R在高血压发生、发展过程中的作用。方法 1 月龄组( S1) 、2 月龄组( S2) 、3 月龄组( S3) 、6 月龄组( S6) 和 9 月龄组( S9) 雄性 SHR 共五组,每组各 6 只,各组均有 相 应 月 龄 的 Wistar-Kyoto 大 鼠( WKY) 作 对 照。采 用 RBP - I 型 大 鼠血压心率测定仪测量大鼠动脉收缩压( SBP) ; 放免法( RIA) 测定血浆血管紧张素Ⅱ( AngⅡ) ; 免疫组化染色结合计算机图像分析方法测定心脏 AT2R 的表达水平,天狼星红胶原染色大鼠的心脏切片。结果 1. SHR SBP 随着月龄的增加呈持续上升( P < 0. 05) ,SHR 的 SBP 均高于相应配对的 WKY 组( P < 0. 05) 。2. 一个月后 SHR 血浆 AngⅡ浓度均高于 S1( P < 0. 05) ,一个月 后 SHR 血 浆 AngⅡ浓度均高于相应配对 的 WKY 组( P < 0. 05) 。3. SHR 心 脏AT2R 免疫染色阳性面积比随着月份的增加而降低,SHR 心脏 AT2R 免疫染色阳性面积比均低于相应配对的 WKY组( P ﹤ 0. 05) 4. SHR 心肌中的胶原含量随着月龄的增加而增加。结论 SHR 心脏 AT2R 表达水平比 WKY 低,并随着年龄的增加而降低。SHR 心肌中的胶原含量随着月龄的增加而增加,而 WKY 无类似趋势。

    Abstract:

    Objective To investigate the expression of AT2R and myocardial collagen in heart of different age spontaneously hypertensive rats( SHR) and to explore the role of these factor in the development of hypertension. Methods Male SHR( n = 30) were allocated to five groups by age: 1) one-month-old group( S1) ; 2) two-month-old group( S2) ;3) three-month-old group( S3) ; 4) six-month-old group( S6) ; 5) nine-month-old group( S9) . Sex and age matched WistarKyoto normotensive rats( WKY) served as controls. Blood pressure was measured. Plasma concentration of angiotensinⅡ ( Ang Ⅱ ) was measured by radioimmunoassay ( RIA ) . The expression of AT2R in heart was examined by immunohistochemistry and computer image analysis. Collagen of the heart was stained by sirus red to observe the myocardial fibrosis. Results 1. An age-dependent increase of SBP was observed in SHR before six months( P < 0. 05) ,the SBP was higher in the different age SHR than that in the matched WKY( P < 0. 05) . 2. Plasma AngⅡconcentration was higher in S2,S3,S6 and S9 than that in S1( P < 0. 05) ,plasma AngⅡ concentration was higher in S2,S3,S6 and S9 than that in the matched WKY ( P < 0. 05 ) . 3. The quantification of AT2R immunohistochemical staining showed: An age-dependent decrease of the AT2R was observed in heart in SHR( P < 0. 05) ,which was lower in S9 than that in S1( P < 0. 05) ,The AT2R levels were lower in SHR than those in the matched WKY( P < 0. 05) . 4. The content of collagen in the heart of SHR showed: An age-dependent increase of the myocardial fibrosis was observed in SHR ( P < 0. 05 ) . Conclusions The expression of AT2R protein in heart of SHR is decreased compared with WKY. The content of collagen in the heart of SHR is increased compared with WKY. An age-dependent varies of AT2R protein and the content of collagen was observed in SHR.
