    目的 研究七氟醚对新生大鼠神经干细胞增殖影响?方法 取出生7 d 的SD 大鼠48 只,体重15~ 20 g,随机分3 组:对照组(Con 组)?1% 七氟醚组(S1 组)?3% 七氟醚组(S2 组)?S1 组和S2 组分别吸入1%?3% 七氟醚4 h?各组新生鼠麻醉开始前腹腔注射5?溴脱氧尿苷100 mg/ kg,于麻醉结束后6 h 处死大鼠并分离脑室区和脑室下区组织?采用免疫组化法进行Nestin 与BrdU 染色,计算检测脑室区和脑室下区神经干细胞增殖率; Western blot 法检测Nestin?Cyclin D1?BMP2?Smad4 蛋白的表达?结果 与对照组比较,S1 组和S2 组大鼠脑室区和脑室下区组织Nestin 蛋白表达降低?神经干细胞增殖率降低,Cyclin D1 表达下降,BMP2?Smad4 表达上调( P <0.05)?结论 七氟醚可抑制新生大鼠神经干细胞的增殖,其机制可能与Cyclin D1?BMP2?Smad4 蛋白表达变化有关?

    Abstract:

    Objective To evaluate the effect of sevoflurane on the proliferation of neural stem cells (NSCs) in the ventricular/ subventricular zone of neonatal rats. Methods Forty?eight 7?day?old Sprague?Dawley rats, weighing 15 -20 g, were divided into three groups (n =16 each) using a random number table: control group (Con), 1% sevoflurane group (S1), and 3% sevoflurane group (S2). Rats in the control group inhaled air for 4 h. Rats in S1 and S2 groups inhaled 1% and 3% sevoflurane, respectively, for 4 h. Cell proliferation was detected by Nestin and 5?bromodeoxyuridine staining. Protein expression of Nestin, Cyclin D1, BMP2, and Smad4 in ventricular/ subventricular tissues was analyzed by Western blotting. Results Compared with the control group, the proliferation rate of NSCs was decreased significantly. Expression levels of Nestin and Cyclin D1 were downregulated, but expression levels of BMP2 and Smad4 were significantly upregulated after sevoflurane treatment ( P < 0.05). Conclusions Sevoflurane inhibits proliferation of NSCs in the ventricular/ subventricular zone of neonatal rats and the mechanism may be related to regulation of Cyclin D1, BMP2, and Smad4 expression.
