    目的 探讨小陷胸汤对高血脂症(hyperlipemia,HLP)小鼠血管内皮损伤的影响。 方法 选用 36 只 C57BL/ 6 小鼠,随机分为对照组,模型组,低剂量组(0. 03 g / mL 小陷胸汤)、中剂量组(0. 06 g / mL 小陷胸汤)、高剂量组(0. 12 g / mL 小陷胸汤)和洛伐他丁组(2. 5 mg / kg 洛伐他丁),每组 6 只,通过高脂饲料喂养 8 周建立 HLP 模型并给予相应药物进行干预。 给药期间观察各组小鼠一般行为学状态,每周称重一次;HE 染色观察主动脉血管组织病理学变化;免疫组化法检测小鼠主动脉组织血管假性血友病因子( von Willebrand factor,vWF)的表达;qRT-PCR 检测小鼠主动脉组织血管内皮生长因子( vascular endothelial growth factor, VEGF)、酪氨酸蛋白激酶 Eph 受体 B4 (EPH receptor B4,EphB4)及其胞膜附着型配体 B2(EphrinB2)的 mRNA 水平;Western blot 检测小鼠主动脉组织 VEGF、EphB4 和 EphrinB2 蛋白水平。 结果 与对照组相比,模型组小鼠活动量和摄食量减少,毛发较差,体重增加,主动脉组织中 vWF、VEGF、EphB4、EphrinB2 表达水平升高;与模型组相比,小陷胸汤各剂量组和洛伐他丁组小鼠活动量和摄食量增加, 毛发光亮, 体重减少, 主动脉组织 中 vWF、 VEGF、 EphB4、 EphrinB2表达水平降低(P<0. 05);与低剂量组比较,中、高剂量组小鼠主动脉组织中 VEGF、EphB4 和 EphrinB2 mRNA 及蛋白表达水平降低(P<0. 05)。 结论 小陷胸汤可抑制 HLP 的发展,对小鼠血管内皮具有保护作用。

    Abstract:

    Objective To explore the effect of Xiaoxianxiong Decoction on vascular endothelial injury in mice with hyperlipidemia ( Hyperlipemia, HLP). Methods Thirty-six C57BL/ 6 mice were randomly divided into control, model, low dose group (0. 03 g / mL Xiaoxianxiong Decoction), medium dose (0. 06 g / mL Xiaoxianxiong Decoction), high dose group (0. 12 g / mL Xiaoxianxiong Decoction), and Lovastatin (2. 5 mg / kg lovastatin) groups. Six rats in each group were fed a high fat diet for 8 weeks to establish the HLP model and administered corresponding drugs for treatment. During the administration period, the general behavioral state of mice in each group was observed and mice were weighed once a week. HE staining was used to observe histopathological changes of aortic vessels. Immunohistochemistry was used to detect expression of von Willebrand factor ( vWF) in aortic tissues of mice. qRT-PCR was used to measure mRNA levels of vascular endothelial growth factor (VEGF), EPH receptor B4 (EphB4), and Ephrin-B2 (EphrinB2) in aortic tissues of mice. Western blot was used to measure the protein levels of VEGF, EphB4, and EphrinB2 in aortic tissues of mice. Results Compared with the control group, the activity and food intake of mice in the model group was decreased, hair was poor, weight had increased, and the expression levels of vWF, VEGF, EphB4, and EphrinB2 were increased in aortic tissue. Compared with the model group, the activity and food intake of mice in each dose group of Xiaoxianxiong Decoction and the Lovastatin group were increased, hair was brighter, weight was reduced, and the expression levels of vWF, VEGF, EphB4, and EphrinB2 were decreased in aortic tissue (P< 0. 05). Compared with the low dose group, mRNA and protein expression levels of VEGF, EphB4, and EphrinB2 were decreased in medium and high dose groups ( P< 0. 05). Conclusions Xiaoxuai decoction inhibits the development of HLP and protects the vascular endothelium of mice.
