    目的 通过系列行为学测试,比较中药白香丹胶囊(BXD)、5-羟色胺 3 受体(5-HT3R)特异性激动剂及其拮抗剂、大麻素受体 1(CB1R)特异性激动剂及其拮抗剂对经前期烦躁障碍(PMDD)焦虑症大鼠的干预效应。 方法 大鼠经卵巢摘除、激素诱导动情周期和居住入侵 3 种复合因子构建 PMDD 焦虑症动物模型。后续实验分两批进行:第 1 批大鼠分为对照组、模型组、5-HT3R 激动剂(1-phenylbiguanide, PBG)组、CB1R 激动剂(WIN55212-2, WIN)组、PBG+WIN 组、BXD 组;第 2 批大鼠分为对照组、模型组、5-HT3R 拮抗剂(Granisetron, GRA)组、CB1R 拮抗剂(Rimonabant, RIM)组、GRA+RIM 组、BXD 组。 药物干预结束后,利用旷场行为测试(OFT)和高架十字迷宫行为测试(EPM)评价各组大鼠焦虑样行为。 结果 相较于对照组,模型组大鼠的居住入侵(RIT)混合攻击行为得分显著升高(P<0. 01)、OFT 中央区进入次数及停留时间显著降低(P<0. 05)、EPM 开放臂进入次数及停留时间百分比显著降低(P<0. 05);相较于模型组,PBG 组、RIM 组、BXD 组的 RIT 混合攻击行为得分显著降低(P<0. 01)、OFT 中央区进入次数及停留时间显著升高(P<0. 05)、EPM 开放臂进入次数及停留时间百分比显著升高(P<0. 05)。 结论 中药 BXD、5-HT3R 特异性激动剂 1-phenylbiguanide、CB1R 特异性拮抗剂 Rimonabant 可有效纠正 PMDD 焦虑症模型大鼠的焦虑样行为。

    Abstract:

    Objective To compare the intervention effects of the Baixiangdan capsule ( BXD), the 5- hydroxytryptamine 3 receptor ( 5-HT3R)-specific agonist and its antagonist, and the cannabinoid receptor 1 ( CB1R)- specific agonist and its antagonist on premenstrual dysphoric disorder (PMDD) rats. Methods An animal model of PMDD anxiety was established using three compound factors: ovariectomy, hormone-induced estrous cycle and residential invasion. Follow-up experiments were carried out in two batches. The first batch of rats was divided into a blank group, a model group, a 5-HT3R agonist (1-phenylbiguanide, PBG) group, a CB1R agonist (WIN55212-2, WIN) group, a PBG+WIN group and a BXD group. The second batch of rats was divided into a blank group, a model group, a 5-HT3R antagonist (Granisetron, GRA) group, a CB1R antagonist (Rimonabant, RIM) group, a GRA+RIM group and a BXD group. After drug intervention, the anxiety-like behavior of rats in each group was evaluated using the open-field test ( OFT) and elevated plus maze (EPM). Results Compared with the control group, the mixed aggressive behavior score of resident- intruder test (RIT) in the model group was significantly higher (P<0. 01), and the number of entries into the central area of the OFT and the percentage of staying in the EPM open arms were significantly lower (P<0. 05). Compared with the model group, the RIT mixed aggression scores of the PBG, RIM and BXD groups were significantly lower (P<0. 01), the number of entries and residence time in the central area of the OFT were significantly higher (P<0. 05), and the number of entries and residence time percentage of the EPM open arm were significantly higher (P<0. 05). Conclusions BXD, 5- HT3R agonist PBG and CB1R antagonist RIM can effectively correct the anxiety-like behavior of PMDD model rats.
