    目的 探究金钗石斛提取物(Dendrobium nobile Lindl,DNL)在 1-甲基 v-4-苯基-1, 2, 3, 6-四氢吡啶 (1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine,MPTP)诱导的小鼠帕金森病(Parkinson’ s disease,PD)模型中的神经保护作用。 方法 将小鼠随机分为对照组、模型组,美多芭组(阳性药组)、DNL(100、200、300 mg / kg)组,通过腹腔注射(i.p)MPTP(30 mg / (kg·d),7 d)建立亚急性小鼠 PD 模型,进行步态分析实验、爬杆实验、悬挂实验等评估 DNL 对 PD 小鼠运动异常的影响;酶联免疫吸附测定法(ELISA)检测各组小鼠纹状体区和皮层区肿瘤坏死因子-α (tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)和白细胞介素-1β( interleukin-1β,IL-1β)表达的变化;蛋白质印迹法(Western blot)检测各组小鼠纹状体 caspase-3 和 caspase-9 的表达情况。 结果 模型组与对照组相比,推进指数、支撑时相的步态指标有显著变化(P<0. 05);与模型组相比,美多芭组、DNL(100、200、300 mg / kg)组在左后、右前、右后推进指数,左后、右前、右后支撑时相均有明显改善(P<0. 05);与对照组相比,模型组在爬杆上半段和总时间增加(P<0. 01、P<0. 001),调头时间和下半段时间无明显变化(P>0. 05);与模型组相比,DNL (300 mg / kg)组可以改善上半段时间(P<0. 05),美多芭和 DNL(100、200 mg / kg)组无明显改善(P>0. 05);与对照组 相比,模型组的悬挂评分降低(P<0.05),与模型组相比,DNL(300 mg / kg)可以增加悬挂评分(P<0. 05),美多芭和 DNL(100、200 mg / kg)无明显改善(P>0. 05);与对照组相比,模型组小鼠纹状体中 TNF-α、IL-1β、IL-6 含量增加(P< 0.05),caspase-3 和 caspase-9 蛋白表达水平显著增高(P<0. 001);与模型组相比,美多芭、DNL(300 mg / kg)组炎症因子指标均降低(P<0. 05),皮层中各组含量无差异(P>0. 05);caspase-3 和 caspase-9 蛋白表达水平均降低(P< 0. 05)。 结论 DNL 能够改善 MPTP 诱导的 PD 小鼠行为变化,减轻脑组织中的炎症反应,抑制神经细胞凋亡,在治疗 PD 药物及功能食品研发方面具有较好的潜力。

    Abstract:

    Objective To investigate the neuroprotective effect of Dendrobium nobile Lindl (DNL) in mice with 1-methyl-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced Parkinson’s disease (PD). Methods After establishment of the MPTP-induced subacute PD mouse model (30 mg MPTP / ( kg·day) i.p, 7 d), the effects of DNL on the motor dysfunction in PD mice were evaluated by gait analysis, the pole test, suspension test and open field test. The effects of DNL on the expression levels of tumor necrosis factor-α (TNF-α), interleukin-6 ( IL-6) and interleukin-1β ( IL-1β) in PD mice were determined by ELISA. The effects of DNL on the expression levels of caspase-3 and caspase-9 in PD mice were assessed by Western blot. Results Compared with the control group, gait indexes, such as the propulsion index and duty cycle, were significantly changed in the model group (P<0.05) compared with model group, whereas Madopar and DNL (100, 200 and 300 mg / kg) groups showed significant improvements (P<0.05, P<0.01 and P<0.001). In the pole test, compared with the control group, climbing in the top half of the pole time and total time of the model group were significantly increased (P<0.01 and P<0.001), but the time of turn around and the time of the lower half of the pole were not changed significantly (P>0.05). Compared with model group, the DNL (300 mg / kg) group showed improvement in the top half of the pole time (P<0.05), but Madopar and DNL ( 100 and 200 mg / kg) groups had no significant improvement (P>0.05). In the suspension test, compared with the control group, the suspension score of the model group was significantly decreased (P<0.05) compared with that of the model group and the suspension score of the DNL (300 mg / kg) group was increased (P<0.05), but there was no significant improvement in Madopar and DNL (100 and 200 mg / kg) groups (P>0.05). Compared with the control group, the levels of TNF-α, IL-1β, IL-6, caspase-3 and caspase-9 were increased in the striatum of mice in the model group (P<0.05, P<0.001) compared with model group and those in the DNL (300 mg / kg) group were significantly lower (P<0.05), but there were no significant differences in the content of cortex among the groups (P>0.05). Conclusions DNL significantly improved MPTP-induced behavioral changes in PD mice, reduced the inflammatory response and neuronal apoptosis in brain tissue, and has a good potential for the development of PD drugs and functional food.
