    目的 构建慢病毒介导 HNRNPK 下调的 HepG2 细胞稳转株,研究 HNRNPK 沉默对肝癌细胞增殖与迁移功能的影响。 方法 以构建的慢病毒感染 HepG2 细胞并筛选阳性细胞获得稳定表达的细胞株,并检测 HNRNPK 表达效率。 通过 CCK-8 法与细胞克隆形成实验研究 HNRNPK 下调对细胞增殖的影响。 通过划痕实验与 Transwell 实验研究 HNRNPK 下调对细胞迁移的影响。 结果 成功构建了慢病毒介导 HNRNPK 沉默的 HepG2 细胞稳转株,HNRNPK 沉默显著抑制了 HepG2 细胞的增殖与迁移能力。 结论 HNRNPK 下调能够在体外抑制 HepG2 细胞的增殖与迁移。

    Abstract:

    Objective To establish a HepG2 cell line with downregulated heterogeneous nuclear ribonucleoprotein K (HNRNPK) mediated by lentivirus and study the effects of HNRNPK downregulation on cell proliferation and migration. Methods HepG2 cells were infected with packaged lentivirus, and positive cells were selected to obtain stable transgenic cell lines. Then detected expression efficiency of HNRNPK. The effect of HNRNPK downregulation on cell proliferation was detected using cell counting CCK-8 and colony formation assays. The effect of HNRNPK downregulation on cell migration was detected using the wound healing and Transwell assays. Results The HepG2 cell line with downregulated HNRNPK mediated by lentivirus infection was successfully constructed. HNRNPK-downregulation inhibited the proliferation and migration of HepG2 liver cells. Conclusions HNRNPK-downregulation inhibits the proliferation and migration of HepG2 liver cells in vitro.
