后肢静脉性溃疡大鼠模型的构建与分析

    目的 采用结扎左髂总静脉+局部皮肤全层切除的方法,构建一种稳定的、可行性高的后肢静脉性溃疡的大鼠,为研究下肢静脉性溃疡提供标准化的动物模型。 方法 将40 只雄性SD 大鼠随机分为3 组,模型组15 只(分离并结扎左髂总静脉后剪去局部全层皮肤)、对照组15 只(只分离左髂总静脉后剪去局部全层皮肤)、空白组10 只(不做处理),观察10 d,期间观察大鼠的成模率、一般状态、静脉彩色多普勒超声图像与创面变化。10 d后各组大鼠予以常规腹腔麻醉并解剖取材,HE 染色观察创面组织细胞形态学变化;免疫组化检测血管P-选择素与ICAM-1 的表达水平;ELISA 检测血浆IL-6 与TNF-α 水平。 结果 (1)与对照组和空白组比较,模型组后肢静脉彩超显示有血栓形成。(2)与对照组比较,模型组的创面面积更大(P<0. 05)、愈合率更低(P<0. 05)。(3)HE 染色显示,与空白组比较,模型组与对照组创面组织均出现炎症细胞浸润,模型组炎症程度更明显。(4)免疫组化结果显示,与对照组和空白组比较,模型组血管P-选择素与ICAM-1 的表达水平显著上调(P<0. 05)。(5)ELISA结果显示,与对照组和空白组比较,模型组血浆IL-6 与TNF-α 水平显著上调(P<0. 05)。 结论 采用结扎左髂总静脉+局部皮肤全层切除的方法,可以成功构建后肢静脉性溃疡的大鼠模型。

    Abstract:

    Objective To construct a stable and highly feasible rat with hindlimb venous ulcer model, test its physiological and pathological indicators, and provide a standardized animal model for the study of venous leg ulcers. Methods Forty male SD rats were randomly divided into three groups. Fifteen rats in the model group (in which the left common iliac vein was isolated and ligated, and the partial full-thickness of the skin was cut off); 15 in the control group (the left common iliac vein was isolated, and the partial full-thickness of the skin was cut off); and 10 rats in the blank group (no treatment) were observed for 10 days, during which the model rate, general state, venous color Doppler ultrasound images, and changes to the wound surfaces were observed. After 10 days, HE staining was used to observe morphological changes to wound tissue cells. Expression levels of vascular P-selectin and ICAM-1 were detected by immunohistochemistry, and the levels of plasma IL-6 and TNF-α were detected by ELISA. Results (1) Compared with ultrasonography. (2) Compared with the control group, the model group’s wound area was larger (P<0. 05) and the healing rate was lower (P<0. 05). (3) HE staining showed inflammatory cell infiltration in the wound tissue of both the model group and the control group, and the degree of inflammation was more obvious in the model group than the blank group. (4) Immunohistochemistry showed that, compared with the control and blank groups, the model group had significantly up-regulated expression levels of vascular P-selectin and ICAM-1 (P<0. 05). (5) ELISA result showed that, compared with the control and blank groups, the model group’s levels of plasma IL-6 and TNF-α were significantly upregulated (P<0. 05). Conclusions A rat model of hindlimb venous ulcer can be successfully constructed by ligating the left common iliac vein combined with partial full-thickness skin excision.
