    目的 采用液相悬浮芯片系统同时测定实验兔圆小囊中IL-1β、IL-1R1、IL-8、IL-8RA和IL-15各基因的表达情况,并对该方法进行评价。方法 利用Affymetrix的Panomics QuantiGene Plex 2.0 Assay中bDNA信号放大和多磁珠分析技术,来同时检测两种实验兔圆小囊中多重mRNA并定量。建立实验兔免疫相关白介素基因的液相悬浮芯片检测方法。结果 可同时检测IL-1β、IL-1R1、IL-8、IL-8R和IL-15各基因的含量,并发现WHBE兔IL-15基因的相对表达量显著高于JW兔(P<0.05),IL-1R1基因的相对表达量显著高于JW兔(P<0.01),IL-8RA基因在WHBE兔中的相对表达量也高于JW兔(P<0.05)。结论 建立了实验兔白介素基因的液相悬浮芯片检测方法,WHBE兔的IL-15、IL-1R1和IL-8RA基因表达量较高,可能与WHBE兔独特的免疫学特性有关。

    Abstract:

    Objective To measure IL-1β, IL-1R1, IL-8, IL-8R and IL-15 genes expression in rabbit sacculus rotundus using the Liquid suspension array system, and evaluate the method. Methods The simultaneous measurement and quantity of multiple genes mRNA expression in rabbit sacculus rotundus was detected by the Panomics QuantiGene Plex 2.0 Assay of Affymetrix using the bDNA signal amplification and beads analysis techniques. Then, this method was applied in the detection of rabbit interleukin gene. Results Content of IL-1β, IL-1R1, IL-8, IL-8RA and IL-15 gene can be detected simultaneously. Gene expression of IL-15 in WHBE rabbit is significantly higher than JW rabbit (P<0.05), and gene expression of IL-1R1 in WHBE rabbit is significantly higher than JW rabbit (P<0.01). Also, the relative expression quantity of IL-8RA gene in WHBE rabbit is higher than JW rabbit (P<0.05). Conclusion Liquid suspension array method for detecting rabbit interleukin gene was established. Gene expressions of IL-15, IL-1R1 and IL-8RA in WHBE rabbit were higher, and all the results reveal WHBE rabbit may have unique immunological properties.
