    摘要:

    近年研究发现,蛋白质质量控制失衡在阿尔茨海默病发病中起重要作用。其中内质网应激和自噬溶酶体通路异常是关键的调控机制。而中药可通过调控内质网应激与ALP的相互作用,减少神经元损伤或死亡,缓解认知障碍。中药单体如二氢青蒿素、齐墩果醇酸、姜黄素、藁本内酯、黄芩素、姜黄素等可通过调控GRP78、CHOP、LC3、LAMP1、P62等关键分子,协调内质网应激与自噬溶酶体通路平衡;中药复方如黄连解毒汤、开心散、地黄饮子、加减薯蓣丸、黑逍遥散则经过PINK1/Parkin、ATF4/CHOP、XBP1-IRE1及mTOR等信号通路,减轻内质网应激并改善自噬溶酶体通路发展缺陷,从而发挥神经保护作用。因此,本文系统阐述内质网应激与自噬溶酶体通路相互作用在阿尔茨海默病中的潜力及中药调控机制研究进展,为中医药现代化及阿尔茨海默病防治提供新策略。

    Abstract:

    Recent studies have found that the imbalance of protein quality control plays a significant role in the pathogenesis of Alzheimer's disease. Among them, endoplasmic reticulum stress and abnormal autophagy-lysosome pathways are key regulatory mechanisms. Traditional Chinese medicine can reduce neuronal damage or death and alleviate cognitive impairment by regulating the interaction between endoplasmic reticulum stress and ALP. Monomers of traditional Chinese medicine such as dihydroartemisinin, oleanolic acid, curcumin, ligustilide, baicalein, and curcumin can coordinate the balance of endoplasmic reticulum stress and autophagy-lysosome pathways by regulating key molecules such as GRP78, CHOP, LC3, LAMP1, and P62. Traditional Chinese medicine formulas such as Huanglian Jiedu Decoction, Kaixin Powder, Dihuang Yinzi, Jiajian Shuyu Pill, and Heixiaoyaosan can alleviate endoplasmic reticulum stress and improve the development defects of autophagy-lysosome pathways through PINK1/Parkin, ATF4/CHOP, XBP1-IRE1, and mTOR signaling pathways, thereby exerting neuroprotective effects. Therefore, this article systematically expounds the potential of the interaction between endoplasmic reticulum stress and autophagy-lysosome pathways in Alzheimer's disease and the research progress of the regulatory mechanism of traditional Chinese medicine, providing new strategies for the modernization of traditional Chinese medicine and the prevention and treatment of Alzheimer's disease.
