Safety Evaluation of Long-Term Consumption of Genetically Modified Maize on Multi-Generational Reproduction and Early Development of Offspring in Cynomolgus Monkeys

    目的 系统评估长期食用转cry1Ab/cry2Aj和G10evo-EPSPS基因玉米(双抗12-5)对食蟹猴多代生殖与子代早期发育的安全性。方法 将36只性成熟食蟹猴(F0代,6雄30雌)随机分为常规饲料组(CK)、转基因玉米组(GM,70%掺入量)及非转基因亲本玉米组(Non-GM),按1:5雄雌比例进行后宫式繁殖。持续记录F0与F1代的生殖性能(怀孕、流产、出生及死亡数),并对F1及F2代子猴进行新出生猴外观、行为神经功能、体格发育及血液生理生化指标的系统检测。结果 F0代GM组怀孕胎次(33次)与CK组(26次)、Non-GM组(29次)无显著差异(P > 0.05);F1代总体存活数(GM组19只、CK组20只、Non-GM组14只)组间无统计学差异(χ2 = 3.141, P = 0.237)。F1代繁殖性能及F2代存活情况组间同样未见显著差异(P = 1.000)。所有子代胎猴外观均未见畸形,行为神经发育评分、体格生长趋势及血液基础生理指标(24项血常规、19项血清生化)在三组间均保持一致。结论 长期食用双抗12-5玉米未对食蟹猴亲代生育能力及子代早期发育产生不良影响,为该类生物育种产品的食用安全性提供了重要的非人灵长类动物实验依据。

    Abstract:

    Objective To systematically evaluate the multi-generational reproductive and developmental safety of long-term consumption of genetically modified maize expressing cry1Ab/cry2Aj and G10evo-EPSPS protein (double-resistant 12-5) in cynomolgus monkeys. Methods Thirty-six sexually mature cynomolgus monkeys (F0 generation, 6 males, 30 females) were randomly divided into a conventional diet group (CK), a genetically modified maize group (GM, 70% incorporation), and a non-genetically modified parental maize group (Non-GM). Animals were bred in a harem system at a 1:5 male-to-female ratio. Reproductive performance of the F0 and F1 generations was recorded, and F1/F2 offspring were systematically assessed for fetal appearance, neurobehavioral development, physical growth, and hematological/serum biochemical parameters. Results No significant differences were observed in F0 pregnancy rates (GM: 33, CK: 26, Non-GM: 29; P > 0.05) or overall F1 offspring survival (GM: 19, CK: 20, Non-GM: 14; χ2 = 3.141, P = 0.237). Reproductive performance of the F1 generation and survival of the F2 generation also showed no significant intergroup differences (P = 1.000). No fetal malformations were detected in any offspring. Behavioral neurodevelopmental scores, physical growth trends, and baseline physiological parameters (including 24 hematological and 19 serum biochemical indices) were consistent across all three groups. Conclusion Long-term consumption of cry1Ab/cry2Aj and G10evo-EPSPS transgenic maize has no adverse effects on parental fertility or early offspring development in cynomolgus monkeys, providing important non-human primate evidence for the food safety of this genetically modified crop.
