    铜死亡(cuproptosis)是一种具有铜依赖性的新型程序性细胞死亡方式,继发于铜过载诱发的线粒体功能受损。过量的铜离子靶向结合三羧酸循环(tricarboxylic acid cycle, TCA循环)中的硫辛酰化蛋白,促进其异常寡聚化,诱导铁硫簇(Fe-S簇)蛋白解离,最终触发级联蛋白毒性应激反应,打破细胞内稳态,导致细胞死亡。研究表明,铜稳态失衡可诱导铜死亡,这一过程与肿瘤的发生、发展和预后密切相关。口腔癌作为头颈部常见的恶性肿瘤,具有高复发率和高耐药性的临床特点,研究发现口腔癌组织中的铜含量显著高于正常组织,这为基于铜死亡的口腔癌诊疗策略提供了全新视角。本文通过综述铜死亡的作用机制,详细探讨铜死亡及其配合物在口腔癌诊断、治疗和预后评估中的研究进展,以期为口腔癌精准诊疗策略提供新的思路与依据。

    Abstract:

    Cuproptosis is a novel form of copper-dependent programmed cell death that occurs secondary to mitochondrial dysfunction induced by copper overload. Excessive copper ions specifically bind to lipoylated proteins in the tricarboxylic acid (TCA) cycle, promoting their abnormal oligomerization and inducing the dissociation of iron-sulfur (Fe-S) cluster proteins. This ultimately triggers a cascade of proteotoxic stress responses, disrupting intracellular homeostasis and leading to cell death. Research has shown that dysregulation of copper homeostasis can induce cuproptosis, a process closely associated with the occurrence, progression, and prognosis of tumors. As a common malignant tumor in the head and neck region, oral cancer is clinically characterized by high recurrence rates and significant drug resistance. Studies have found that the copper content in oral cancer tissues is significantly higher than in normal tissues, providing a brand-new perspective for the development of cuproptosis-based diagnostic and therapeutic strategies for oral cancer. By reviewing the mechanism of cuproptosis, this article elaborates on the research progress of cuproptosis and its complexes in the diagnosis, treatment, and prognostic evaluation of oral cancer, aiming to provide new ideas and evidence for the precise diagnosis and treatment of oral cancer.
