    目的 探讨木犀草素(luteolin,LUT)对甲状腺乳头状癌TPC-1 细胞侵袭迁移的影响及其可能的机制?方法 采用不同浓度的木犀草素(0?20?40 μmol/ L)与人甲状腺乳头状癌TPC-1 细胞共同培养,实验分为三组:空白对照组(加入等量的二甲基亚砜DMSO)?LUT 20 μmol/ L?LUT 40 μmol/ L?MTT 法检测细胞的生存率,Transwell 小室实验检测细胞侵袭?迁移能力,利用Western blotting 和qRT-PCR 分别检测细胞中磷酸化蛋白激酶B(p-AKT)?基质金属蛋白酶2?9(MMP2?MMP9)的蛋白和MMP2?MMP9 mRNA 表达的变化水平?结果 与空白对照组相比,不同浓度的木犀草素对TPC-1 细胞的增殖活性具有明显的抑制作用(均 P < 0. 05);LUT 浓度为20μmol/ L?40 μmol/ L 组均明显抑制了TPC-1 细胞的侵袭?迁移能力(均 P < 0. 05);与空白对照组相比,LUT 作用于甲状腺乳头状癌细胞TPC-1 后,p-AKT?MMP2?MMP9 蛋白和mRNA 的表达水平明显降低(均 P < 0. 05)?结论 木犀草素可能通过PI3K/ AKT 这一信号通路抑制甲状腺乳头状癌TPC-1 细胞侵袭迁移能力,从而在甲状腺乳头状癌治疗及预后中发挥着重要的作用?

    Abstract:

    Objective To investigate the effects of luteolin on the invasion and migration of papillary thyroid carcinoma TPC-1 cells and its possible mechanism. Methods Various concentrations of luteolin (0, 20, and 40 μmol/ L)were applied to human papillary thyroid carcinoma TPC-1 cells. The experiment included three groups: blank control group (with the same amount of dimethyl sulfoxide), 20 μmol/ L group and 40 μmol/ L group. The survival rate of cells was detected by MTT assays, and cell invasion and migration abilities were analyzed by Transwell assays. Western blotting and qRT-PCR were used to measure the expression levels of phosphorylated protein kinase B ( p-AKT), and matrix metalloproteinase (MMP)-2 and -9. Results Compared with the blank control group, luteolin at various concentrations had a significant inhibitory effect on the proliferation of TPC-1 cells (all P < 0. 05). Luteolin concentrations of 20 and 40 μmol/ L significantly inhibited the invasion and migration of TPC cells. Compared with the blank control group, the expression levels of p-AKT, MMP-2, and MMP-9 proteins and mRNAs were significantly decreased in TPC-1 cells after luteolin treatments (all P < 0. 05). Conclusions Luteolin inhibits the invasion and migration of papillary thyroid carcinoma TPC-1 cells through the PI3K/ AKT signaling pathway, which might play an important role in the treatment and prognosis of papillary thyroid carcinoma.
