    • Effects of 60Co-γRadiation on Feed for Laboratory Animals

      Abstract:分别用3 000、 4 000、 5 000、 6 000、 7 000、 8 000Gy60Co—γ射线辐照实验鼠料和实验兔料后,对辐照饲料的细菌总数进行了测定。经回归分析得到辐照剂量(x,Gy)与饲料中细菌总数(Y,g-1)的回归方程,鼠料为y=6 212.9e(-6.659 2 x/10 000),兔料为 y=8 243.9 e(-6.593 2 x/10 000)。用上述方程进行估计,生产无菌鼠料所需辐照剂量为60 000 Gy,生产无菌兔料所需辐照剂量为86 000 Gy。

      Abstract:【摘要】用BV DIAdot Kit和BV EIA抗原片对本中心100份经HSV-1抗原片检测为阳性,150份HSV1抗体阴性的食蟹猴血清进行B病毒抗体检测。结果发现,BV DiAdot Kit对150份阴性血清的阳性检测出率为8.7%,而B、EIA的阳性检测率为18.o%,二者的符合率90.7%。后者阳性检测率明显高于前者。用上述两种方法对100份HSV-1抗体阳性血清( - )进行检测发现,BV DLAdotKit和BV EIA的阳性检出率均为100%。后者价格低廉且对HSV-1抗体阴性血清的阳性检出率较高,值得在国内猕猴生产和出口单位大力推广。

      Abstract:As enlargement of poultry industries, demand of SPF eggs has been increased quickly. 2 000 breeder eggs(l800 of female line and 200 of male line) were introduced from America in order to meet marketing requirements in 1995. New colony had been hatched and maintained in separated barrier system until completing animal health tests twice at 10 and 16 weeks old of chickens. After several generations, the production performances of new colony were much better than original data, particular in laying rate and fertility, from 83% and 93% to 85% and 98% separately.

    • Study of Immune Endurance of Speciality on Allogeneic-Bone-Marrow-Transplantation (ALLO-BMT) in Survival Mice

      Abstract:目的通过对异基因骨髓移植(ALLO-BMT)存活小鼠及其皮片移植的观察,以判断ALLO-BMT小鼠是否建立了特异性的兔疫耐受机制。方法(1)给A组受者CB6F1(H-2bd)小鼠注射生理盐水,给B组受者CB6F1(H-2bd)小鼠移植供者C57BL/6j(H-2b)小鼠的骨髓细胞;(2)将供者C57BL/6j(H-2b)和BALB/c(H-2d)小鼠的尾部皮片原位移植给ALLO—BMT后存活60 d以上的受者CB6F1(H-2bd)小鼠和未经任何处理的CB6F1(H-2bd)小鼠,观察100 d。结果(1)A组受者CB6F1(H-2bd)小鼠平均存活时间(10.6土1.3)d显著短于B组的受者CB6F1(H-2bd)小鼠平均存活时间(36.9±10.8)d(p<0.01)。(2)II组中进行ALLO-BMT后存活60 d以上的受者CB6F1只接受来自C57BL/6j小鼠的皮片而排斥来自BALB/c/小鼠的皮片,而I组中未经任何处理的 CB6F1小鼠排斥来自 C57BL/6j和 BALB/C /c小鼠的皮片。结论经 γ射线照射并移植有C57BL/6j 小鼠异基因骨髓且能长期存活的CB6F1小鼠建立了较完全的供者型免疫耐

    • Genetic Monitoring of the IRM-2 Inbred Strain Mouse

      Abstract:Objective The aim of this study is to examine the genetic characteristics of the IRM-2 mouse which is a new inbred strain mouse hybridized by the ICR(paternal strain) with 615(maternal strain) and has progressed to 38th generation since 1990. Methods Genetic monitoring of the IRM-2 inbred strain mouse was performed by skin grafting test, coat color test and biochemical marker analysis. Results The skin grafting test showed that no dropping graft was found during 100 days after transplanting, implying the IRM-2 was of isohistogenicity. Analysis of coat color genes indicated that hair color of first generation hybrid crossed between F38 and BALB/c was cassia bark as same as the IRM-2, suggesting that the coat color genes of the IRM-2 of were homogenous. The gene type is AAbbCCDD. Measuring of 25 and 13 biochenmical markers gene loci on 13 chromosomes was performed by electrophoresis for F23 and F38 respectively. The results revealed that all observed gene loci were homogenous without any polymorphism and mutation. Conclusion The IRM-2 inbred strain mouse is high homogenous in the genetic characterishcs.

      Abstract:采用60Co一γ射线3.0 Gy照射后第4 d开始给予环磷酰胺50.0 mg·kg-1及氯霉素62.5mg·ks一1,共3 d,获得了AA模型。外周血象示三系细胞均有显著减低(P<0.05或P<0.01),骨髓增生明显低下,造血细胞容量<20%,脂肪细胞明显增多,各项指标均符合再生障碍性贫血。此方法简便、周期短、成功率高。

      Abstract:In this experiment we selected six Chinese No.l miniature pigs to produce 51 pigs by laparohysterectomy-birth in the isolators, 7 pigs were shifted out because of too light body weight or dead fetus. 44 pigs existed with the surgery etistense percentage of 86.3%. After stopping lactation 36 pigs existed (81 .8%). In 10 weeks of feeding period, we checked and tested feces, feeding material, drinking-water, wind inlet /outlet and feces outlet for the anaerobic bacteria, aerobic bacteria, fungus, separately at 21d, 42d, 63d by the methods of USP, NB, SB, and all the test results were negative. Pig Pasteurella bacteria, Swine erysipelas bacteria, foot and mouth disease (FMD) virus, swine fever virus, pig mini-virus, toxoplasma were detected with all the negative results both on the mother pig before laparohysterectomy-birth and on the new-born pigs at 45d after the birth. The new-born pigs fulfill the SPF standards.

      Abstract:检测6月龄和20月龄大鼠血中二甲基精氨酸和降钙素基因相关肽浓度的变化。结果发现,20月龄大鼠血清二甲基精氨酸浓度显著升高,而血浆降钙素基因相关肽浓度无明显变化。提示老龄大鼠血中二甲基精氨酸浓度升高可能是促进血管内皮功能不全的重要原因。

      Abstract:Objective To compare the effects of supporting mouse blastocyst development and differentiation in sequential culture media and non-sequential culture media. Methods Zygotes were cultured in media Q-three for 48h after 48h of culture with media Q-one, while zygotes were cultured only in media Q-one in control group. Results The percentage of total blastocysts and the percentage of hatching blastocysts in sequential culture group were significantly higher than in non-sequential culture group, 89% vs. 12% and 38% vs. 0 respectively. Morever, the formation of expanding blastocyst was morphologically better in sequential media. Conclusions The sequential culture system was beneficial for mouse embryo development and differentiation from early embryo stage to blastocyst stage.

      Abstract:To maintain the clean animal facility needs much energy. We compared different kinds of energies during the time of design and reconstruction of Nanwei Road Clean Animal Facility, and adopted some measures for reusing the energy that was from cool or warm waste air. The result showed that the environmental indicators of the clean animal facility reached the National Standard for Laboratory Animals. We saved money obviously by using the economized energy measures in summer and winter when the animal facility was operated.

      Abstract:目的从免疫学的角度对分离的B病毒147株与单纯疱疹病毒在蛋白多肽水平上进行分析。方法利用Westernblot技术对新分离的B病毒147株及单纯疱疹病毒感染的细胞多肽进行分析。结果得到了八条含有B病毒特异抗原表位的多肽。结论该方法可以明确区别R、和HSV-l、HSV-2,并为寻找特异抗原进行表达奠定了基础。

      Abstract:目的测定正常成年近交系小鼠IRM-2的一些血液学和血清生物化学参数.方法用自动光电血球计数仪测定分析了IRM-2小鼠的血细胞成分及相关参数;用半自动生化分析仪测定了血清常用的13项生化指标;并进行了常规法骨髓细胞的分类.结果雌、雄小鼠各10只,平均生化指标及活度分别为:ALT:39.1 U/L 和44.1 U/L, AST:128.2 U/L 和113.0 U/L, ALP:117.7 U/L 和76.5 U/L,GLU:4.2 mmol/L和4.7 mmol/L,TG:0.79 mmol/L和 0.73 mmol/L,CHO:2.9 mmol/L和 3.5 mmol/L, TP: 29.4 g/L 和 28.1 g/L, ALB:21.2 g/L 和18.9 g/L,BUN:5.29 mmol/L和 5.0 mmol/L,CRE:52.1 μmol/L 和 51.0 μmol/l, Ca:6.8 mg/dl和 7.0 mg/dl,P:7.4 mg/dl和7.7 mg/dl, TBIL:38 μmol/L和 38 μmol/L.外周血检测为RBC:9.5×1012/L和9.1×1012/L, WBC:10.3×109/L和 9.5×109/L,PLT:485(109/L 和604×109/L,粒细胞(包括杆状核和分叶核)分别占21.2%和 31.7%,白细胞中的淋巴细胞分别占78.7% 和68.2%.骨髓象中粒系统占45%,红系统占22%.结论从这些结果可以看出,IRM-2近交系小鼠的血液生理、血液生化指标较稳定,个体差异较小,适合用作辐射损伤动物模型和肿瘤放射治疗动物模型及其他研究.

      Abstract:近40年来,人体的一些重要细胞组织低温保存和移植的逐步成功,低温技术在治疗肿瘤等方面的应用,动植物细胞、胚胎、组织低温保存的发展,已经给医学、生物学、农业、畜牧业等带来了巨大的效益,并使低温生物学、低温医学成为人们关注的新兴交叉科学。在实验动物方面,随着生命科学的飞速发展,实验动物的品种、品系数量也在不断地增加。因此采用胚胎冷冻保存技术,建立实验动物的胚胎库,将是保存实验动物品系和我国特有的动物资源的行之有效的方法。同时也为进行胚胎发育机理研究、转基因动物培育、动物模型培育等创造了条件。
