Abstract:目的 通过选择目前几个培养基生产厂家的几种常用生化培养基,利用已知标准菌株进行对比、优选,并提出应用时的一些注意事项,最终确定适合于实验动物细菌检测用的生化培养基。方法 将标准菌株接种血琼脂,取菌苔分别接种克氏双糖铁琼脂、硝酸盐培养基、尿素培养基、蛋白胨水、明胶液化培养基,37℃培养,观察结果。结果 鼠棒状杆菌和嗜肺巴斯德杆菌在各厂家的克氏双糖铁培养基上反应结果有较大差异;沙门菌在北京市双旋微生物培养基制品厂的尿素培养基上有淡淡的红色(阳性),影响了实验结果的判断;中国药品生物制品检定所的硝酸盐培养基的试验管和空白对照管均为阳性,实验结果无法判断;微量生化反应管与干粉培养基的效果一样。结论 各厂家生化培养基的质量有待进一步提高,以杭州市天河微生物试剂有限公司生产的生化培养基质量相对较好。在实际应用中,可参照本研究的结果,结合本实验室的使用情况选择使用。

Abstract:Objective To isolate L1210 cell lines by the single cell cloning technique and to compare some cytology characteristics of L1210 cells and clonal cells.Methods By scanning electron microscope we observed cell superficial structure; DNA content analysis by flow cytometry; the protein distribution analysis by immunohistochemistry. Results Clonal cells L2B9, L2H7,L2H8,L3B12 and L3E11 had a lot of tiny crests and tiny folds, but difference in density, L2C9 had abundant microvilli and L3F9 had massive prominency. DNA contents were 8.8,8.3,8.3,8.5,8.6,8.7,8.9,9.0pg for L1210 and seven sub-lines L2H7,L3F9, L2H8,L2C9,L3B12,L2B9, L3E11,respectively. Statistic analysis indicated significant difference ( P <0.01).Main range of the chromosome number of L1210 cells were 62-63, chromosome number of the clonal cells were 48,58,64,66,62,63,58 of L2B9, L2C9, L2H7,L3B12,L3E11,L3F9,L2H8 respectiverly. With immunohistochemistry different clonal cells had different reactions to 10 kinds of antibodies, and the reaction positive rates of clonal cells were higher than L1210 cells.Conclusion Seven sub -linesshoweddifferentproperties.

Abstract:Three strains of mice (ICR, B6C3F1 and C 57 BL/6J) of 28, 56 and 112 days old were in superovulation with PMSG and HCG. The techniques of in vitro fertilization, culture, and transplantion of two-cell embryos to pseudopregnant recipients were used. The highest numbers of oocytes were obtained from 28-days old superovulated mice in the three strains, being prominent discrepancy with 56-days and 112-days old mice ( P <0.01). There was no difference of the insemination rate among three strains( P >0.05).Approximately 91% of ova developed to the two-cell stage after in vitro fertilization, and about 51% of these living two-cell embryos from the three different strains developed into foetus through transplantation in recipients, with no discrepancy in rate of pregnancy( P >0.05).\;

Abstract:目的 对近交系小鼠体内受精的胚胎在三种不同处理温度(20℃、25℃、28℃)进行玻璃化冷冻保存。方法 不同温度下,胚胎在10%EG溶液中预处理5min,再投入EFS40玻璃化溶液中1min,平衡后冷冻。结果 与20℃和28℃相比,三个品系小鼠的胚胎都在25℃冷冻时获得最佳效果,解冻后培养观察发育率分别是87.77%、90.00%、87.18%。结论 该结果对近交系小鼠胚胎保存具有重要意义。

Abstract:目的通过观察YH-L2600型新风换气机在全新风净化环境中不同季节的显热交换效率,了解其节能效果.方法1999年9月1日至2000年8月31日,每天7、13、19时观察记录新、污风交换前后的温度和洁净区的温湿度,计算显热交换效率.结果不同月份的显热交换效率有明显的区别;全年节电68580kW*h.结论YH-L2600新风换气机安装在空调系统主送风管道中,其显热交换效率与自备风机是否运转及湿度无关;温差越大,显热交换效率越高,节能效果越明显;良好的管理与维护,可保持该机优越的显热交换效率.

Abstract:Objective\ Establishing KM\|hairless inbred strain by means of complete,brother\|sister inbreeding after twenty generations.Methods\ The coat color gene test,skin graft and biochemical marker gene tests were used.Results and Conclusion\ The coat color gene test showed that the strain had the geno type of aabbccDD.All mice with grafted skin survived.No heterzygote was found according to the results of the twewty\|five biochemical marker gene (including akp-1)tests.

Abstract:目的探讨自X期鸡胚培养方法,制备嵌合体鸡.方法分离供体麻鸡种蛋的胚盘细胞,注入受体来航鸡种蛋胚盘,然后用以下两种方法培养鸡胚①钝端开窗法,沿鸡蛋气室边缘开口,内壳膜开小孔,注射胚盘细胞后再封口,置孵化器内,温度38℃,翻蛋角90°;②两次倒蛋法,胚盘细胞注入受体蛋后,倒入代用蛋壳Ⅰ,置孵化器内,温度38℃,翻蛋角90°,72h后,倒入代用蛋壳Ⅱ,改翻蛋角30°.结果用钝端开窗法注射83只种蛋,出雏35只(42.2%),表型嵌合体7只,3只存活;用两次倒蛋法,注射72只种蛋,出雏10只(13.9%),表型嵌合体5只,1只存活.结论钝端开窗法较经典的两次倒蛋法出雏率明显提高,用该方法制备嵌合体鸡更为方便可行.

Abstract:Objective\ To determine the growing index of SPF miniature pig.Methods\ The Chinese No.1 miniature pig by laparohysterotomy was fed with the SPF feeding method in isolator,and its weight,body\|length,thorax circle,and body\|height were measured separately at new\|born day,30?d,60?d,compared with the miniature pig of same age fed in the natural circum\|stance.Results\ The body weight,length,height and thorax circle of pigs fed in natural circumstance were all larger than those of the SPF feeding miniature pigs ( P <0.01,0.01< P <0.05).The coefficients of correlation of the body weight\|length\|thorax circle and height of the SPF miniature pig were separately 0.978,0.977,0.982 in new\|born condition;0.900,0.916,0.982 at 30?d; 0.917, 0.896,0.913 at 60?d.Conclusions\ The above growing index of SPF miniature pig are basically in consistence with that reported in papers.
