Abstract:Objective To study the effects of chemical medicine combined with immunopotentiator on sarcoma 180 Cells.Methods Kunming mice were used as laboratory animal models which were all with sarcoma 180 cells transplanted subcutaneously. Test group was administered FK309 combined with BCG. Then the authors did the ordinary blood test and serumal biochemical test,observed pathological tissue slices,and calculated inhibition rate of the growth of tumor. Results Total protein and albumin of test group were higher than those of control group. Examination of tissue slices showed that necrotic degree of tumor of test mice was worse than that of controls. The inhibition rate of sarcoma on the fifth,tenth,fifteenth days was 14.3%, 21.4% and 36.4% respectively. Statistical assay indicated that there was no significant difference (P<0.05) between the mean weight of tumor of both groups until the fifteenth day(P<0.05) . The mean weight of mice had no conspicuous difference.Conclusion FK309 with BCG may be of value in the inhibition of the development of tumors.

Abstract:Objective To study the histopathological changes in lungs spontaneous lesions of breeding %macaca mulatta.% Methods Lung samples from 2-20 years monkeys were investigated by routine histological technique.Results Various spontaneous lesions,totaling 17 kinds in different degrees,were found in 84.6% monkeys of the study,including pneumonia,inflammatory cell infiltration,degenerations,abscess,proliferation of fibrous tissue, and fibrosis.The degree and the freqency of lesions increased with age.Conclusion The spontaneous lesions of lung were often observed in %macaca mulatta% and its interference effect should be seriously taken.

Abstract:Objective To breed transgenic mice containing xylE gene and to screen homozygous transgenic mice. Methods Two founders(G 09 and G 031) of transgenic mice containing a %xylE% mutational target gene were selected for breeding. G 09 and G 031 were mated with nontransgenic ICR mice respectively and F 1 offsprings were produced. F 2 and F 3 offsprings were generated by inbreeding of the same litters of F 1 or F 2 respectively. Results Their offspring's phenotype had no detectable abnormal. Detection of line 31 indicated that the %xylE% target gene could stably transmit to the next generations. One homozygous male mouse of line 31 by PCR had been confirmed. Through mating the mouce to nontransgenic partners, all the 28 offsprings (four litters) were transgenic by PCR-Southern detection. Conclusions 1.Transgene could be stably transmitted to F 3. 2.One homozygous transgenic mouse was screened within 4 generations of line 31.;

Abstract:Objective To observe the effect of conjugated linoleic acids on serum lipids level in rats, according to serum TC,TG,HDL-TC.Methods 50 male Wistar rats were divided into five groups(normal control group, high fat control group,three conjugated linoleic acids groups).The rats were fed with high fat diets and different conjugated linoleic acids oil and four weeks later,serum lipids level are measured. Results Compared with high fat control group,serum TC,TG levels decrease significantly while HDL-TC level kept steady.Conclusion It is suggested that conjugated linoleic acids can reduce serum lipids level.

Abstract:Objective To evaluate the effect of seawater immersion after open abdominal trauma on several plasma enzymes. Methods Sixty Wistar rats were divided into two groups. A model of an open abdominal injury was established. Control group(n=30) was simple abdominal trauma. Experimental group(n=30) had seawater immersion after chest trauma. Blood samples were taken at three different time intervals for assessing creatine kinase (CK), lactate dehydrogenase (LDH), alanine aminotransferase (ALT) and aspartate aminotransferase (AST). Results The plasma CK, LDH, ALT and AST were significantly higher in experimental group than that in control group. Conclusion Plasma enzymes including plasma CK, LDH, ALT and AST have significantly changes in experimental animals and more severe with time.

Abstract:Objective To investigate whether there is any injury in kidney after intestine ischemia-reperfusion in dogs in accordance with the concentration change of nitric oxide(NO) and superoxide dismutase(SOD), and expression of c-Fos,bax,bcl-2 in Kidney of dogs. Methods The superior mesenteric artery was ligated for 30 min to establish intestine ischemia-reperfusion model in dog, and the NO and SOD in vein blood were assayed both before ligation and reperfusion in 0,30,60 min, 4 d, 7 d; then the expressions of c-Fos,bax,bcl-2 in kidney in experimental and control groups were studied at 0,30,60 min, 1 d,4 d,7 d. Results After intestine ischemia-reperfusion the concentration of NO from vein blood increased except 4 d group, and the concentration of SOD increased significantly (P<0.01); the expression of c-Fos was extremely high (P<0.01); the expression of bax was gradually increased, and the significant difference appeared in 30 min and late groups (P<0.01); the expression of bcl-2 didn't appear in 0 min, but appeared in 30 min and late groups (P<0.01). Conclusion The free radical increase after intestinal ischemia-reperfusion resulted in the change of c-Fos, bax,bcl-2 expression in the kidney, which may lead to kidney injure.

Abstract:Objective To observe the effect of new drug Xisichuan on experimental asthma in guinea pigs. Methods Experimental asthma models were induced by acetylcholine and histamine nebulization or ovalbumin injection investigated. Results and Conclusion Xisichuan has distinct bronchic dilatation effect on acetylcholine and histamine induced guinea pigs model. It can partly prevent the bronchial constriction induced by ovalbumin immune sensitivity in guinea pigs.

Abstract:Objective Chlamydia psittaci is now suspected as a primary predisposing factor eliciting a sever respiratory syndrome. Methods The broiler blood samples suffered from respiratory symptom were screened in Beijing and Tianjing areas during 2001 to 2002.The lung, heart and kidney tissue from infected broiler were fixed,stained,implanted and observed under microscope; meanwhile the lung tissue suspension from infected broiler was inoculated into the SPF egg on day 7. The yolk membrane of the dead chick embryo were collected during day 4 to 9,dyed by Lugols Iodine and by Chlamydia Immunofluorescence Assay. In order to discriminate Chlamydia psittaci from Chlamyida trachomatis,the activity of isolated pathogen was inhibited by infusing Penicilln and sodium sulfadiazine.The isolated pathogen was injected into broilers of 15 day,observering the clinical pathology. Results The serological response was seen after the clinical signs, and the positive rate varied from 10%-30%.The activity of pathogen was demonstrated positive in fluorochrome.The same pathological change could be replicated by injecting the isolated pathogen. Conclusion The isolated pathogen may be an Chlamydia psittaci, whose conbined infection with E.Coli can be responsible for the severe respiratory disease with high mortality in broilers.

Abstract:目的 研究上下方角膜缘上皮非对称性损害对角膜曲率的影响。方法 将兔的 11 1点方位的角膜缘上皮保留 ,其他方位角膜缘在外围保留 0 2mm宽的上皮带 ,其余上皮全部刮除 ,造成角膜上下方干细胞储备的差异。采取角膜地形图测量、组织学等方法观察角膜形态变化。结果 角膜中央区出现锥形凸起 ,锥形凸起处屈光值 (5 7 2 5± 2 73屈光度 )比术前角膜对应点屈光值 (5 2 2 3± 1.91屈光度 )明显增大 ,差异有显著意义 (P <0 0 1) ,组织学观察角膜锥形凸起处有代偿性增生的上皮细胞团。结论 角膜中央代偿性上皮增生可以显著地改变角膜曲率 ,角膜缘干细胞的分布异常是造成角膜中央上皮代偿性增生的原因之一。

Abstract:目的 观察Cx43基因异常引起心功能障碍的超声心动图表现。方法 对胎龄为14.5d、15.5d或16.5d的Cx43基因剔除、CMV43和FZ转基因小鼠胚胎,进行多普勒超声心动图检测、心脏形态学检查和基因型测定。结果 cx43基因异常小鼠胚胎主要表现为心室收缩期射血峰值增高(IPSV)或/和舒张期房室瓣口血流频谱呈单峰(MIP)。IPSV增高与肺动脉口狭窄相一致;而舒张期MIP则与右心室肥厚或右心室扩大相符合。结论 超声心动图在小鼠心脏病模型的研究方面具有较高的价值;Cx43异常可导致肺动脉口狭窄和右室舒张功能障碍。

Abstract:Objective To study the anatomic feature and the function of kidney in Smad3 gene knock-out mice. Methods The blood parameters of kidney drainage were measured by anatomical method and by VITALAB. Results There were significant difference in mice between types and age of Smad3 gene knock-out mice the homozygous mutants . The parameters of Cre,avoirdupois, kidneys and adrenal gland were significant differences between three types in 35 days mice :the parameters of the UA and BUN were higer than those of heterozygous mutants ,and the parameters of avoirdupois and visceral coefficient were lower than those of wildtypes ,but the parameters of Cre and BUN were higher than wildtypes. The parameters of UA of 70 days homozygous mutants were higher than wildtypes, but UA of 6 months lower. There were no significant differences in homozygous mice ,but parameters of Tbili ,Cre and BUN were higher in 6 months, the UA was higher in 70 days. Conclusion The metabolite outcomes were different in different ages of the mice . There were 12.5% one-kidney-mice which were produced by homozygous mutants,but the adrenal glands in existence. It was showed that Smad3 gene konck-out affected the formation of the kidneys.

Abstract:目的 观察体外状态下新西兰白兔骨髓中成纤维细胞集落形成单位 (CFU F)及成骨性的CFU F集落数量 ,为探讨骨髓的成骨能力提供实验依据。方法 将实验动物分为幼年组 (6个月~ 1岁 )、壮年组 (2 5~ 3 2岁 )两组 ,在无菌状态下 ,在每只动物的股骨上穿刺点抽取骨髓 0 8ml,每个培养瓶 (2 5ml)接种含 8× 10 5个有核细胞的骨髓量 ,在体外培养 12d时计数CFU F集落数量。计数后将培养瓶中CFU F集落进行ALP染色 ,在肉眼下计数着色的CFU F集落 (即成骨性CFU F集落 )数量。结果 按照集落着色的深浅 ,本研究把CFU F集落分为 3种 :①强阳性集落 ;②弱阳性集落 ;③阴性集落。两组成骨性CFU F集落数量约占CFU F集落总数的 1 4 ,主要为弱阳性集落 ,占成骨性CFU F集落的 70 %~ 80 % ;幼年组与壮年组强阳性集落分别占CFU F集落总数的 1 13和 1 17,幼年组明显高于壮年组 ,两组差异有显著性 (P <0 0 5 )。由于每个CFU F集落均来源于一个骨髓基质干细胞 ,通过集落形成率可推算出新西兰白兔骨髓中每 110 0 0~ 130 0 0个有核细胞中有一个骨髓基质干细胞 ,后者仅占骨髓细胞的百万分之一。结论 骨髓中骨髓基质干细胞数量极少 ,成骨性骨髓基质干细胞就更少了 ,但就骨髓细胞总量来说骨髓中骨髓基质干细胞也是相当可观的。

Abstract:Objective To observe the changes of aging data of Wistar rats in the subacute aging model induced by the subcutaneous injection with D-galactose.Methods 20 male Wistar rats were divided randomly into two groups.Each group contained 10 rats for the control or aging model.The control rats were injected subcutaneously with 10 ml PBS/kg each day,and the rats of the aging model with 48 mg D-galactose/kg each day.After 42 days with continuous injection,the blood specimens were collected for the detection of the data relative to the aging. Results Compared with the control , the weight of the aging model rats increased slowly. The thymus of these rats shrivelled clearly,and their weight decreased as well (P<0.01).In addition,the superoxide dismutase(SOD) activity in the serum of these aging rats declined obviously(P<0.01). Conclusion The aging model could be established by the mimic aging reaction induced with D-galactose.

Abstract:Objective To explore and improve operative management of pancreas transplantation in miniature swine. Methods The 40 miniature swine were divided into donors and recipients and pancreas transplantation was performed 20 times in all. Miniature swine were anesthetized with ketamine and maintained with ketamine and etomidate. Meanwhile, mean artery pressure(MAP), central venous pressure (CVP) and blood gas (BG) were monitored. And the 400-800 ml blood was obtained from abdominal aorta of donor and transfused to recipient in the operation. Dexamethasone and dopamine were given in the reopening of inosculated blood vessels. Results The rate of successful operation was 90%. Recipient's MAP were significantly decreased after the reopening of inosculated blood vessels,compared with MAP before the reopening(P<0.05). Other monitored data had not markedly changed. It was more superior and convenient to obtain blood from abdominal aorta than inferior vena cava of the donor. The average transfusion was 400 ml to a recipient and injected dexamethasone and dopamine had contributed to stabilization of circular system in the reopening of inosculated blood vessels. Conclusion Enhancement of monitoring and management of circular system, timely transfusion and stabilization of blood pressure could improve the rate of successful operation and were crucial for pancreas transplantation in miniature swine, especially after the reopening of inosculated blood vessels.

Abstract:Objective To investigate the method to establish experimental animal model of ventricular septal defect (VSD) in pigs. Methods Thoracotomy was performed on 15 young pigs under general anaesthesia. A spool-like ring amounted on a specially designed centesis tool was put into right ventricle via an incision on right auricle which was pushed to puncture through the septum. The ring was then left on the septum to create the artificial VSD. 3 days after the operation, echocardiography was conducted to verify the existence of VSDs. The animals were catheterized one month later to obtain hemodynamic data. Results 9 pigs survived the operation with successful acquisition of VSDs. Doppler echocardiography demonstrated the transseptal shunts with peak velocities ranged from 1.7-4.4m/sec. Cardiac catheterization showed the Qp/Qs of 1.68-2.12 although the pulmonary pressures remained in normal limits. Conclusion The authors report their experience in the establishment of experimental VSD model in pigs in revised Synhost method. The preliminary result shows that the VSD model in pigs is feasible and worth improving further in order to provide a useful animal model for the pathophysiological study of intracardiac left to right shunt in the future.

Abstract:目的 建立IRM— 2小鼠自发性肿瘤模型和移植性肿瘤模型 ,为肿瘤研究提供实验模型。方法 采用瘤块接种法及悬液接种法。 (1)无菌环境取 2mm3瘤块 ,接种于小鼠腹股沟皮下。 (2 )将瘤块研磨 ,加生理盐水稀释至 1× 10 7 ml,取细胞悬液 0 2ml(含 1× 10 6 细胞 )注入腋窝皮下 ,观察肿瘤生长情况。结果 接种恶性淋巴瘤连续传 70代 ,成瘤率 10 0 % ,移植津白Ⅱ号高乳腺癌瘤株连续 4 5代、移植T 739肺腺癌瘤株 5代 ,成瘤率均为10 0 % ,且生长稳定 ,传代周期固定。结论 该自发性恶性淋巴瘤具有较强的特异性 ,证明该模型是成立的。
