    • Function Analysis of Human ATP7B in Copper Transport by Using Transgenic Rat

      Abstract:目的 利用转基因大鼠模型 ,研究人Wilson病致病基因ATP7B在铜代谢通路中的功能。方法 将构建的 7 1kb含有鸡 β肌动蛋白启动子的人正常ATP7BcDNA ,经显微注射法导入Wilson病动物模型LEC(long_evanscinnamon)大鼠受精卵的雄性原核。以RT_PCR和Westernblot分析其在转基因大鼠各组织内的表达谱 ;用抗大鼠铜蓝蛋白抗体检测肝细胞全铜蓝蛋白 (holoceruloplasmin)的合成 ;同时对 6~ 30周龄的转基因大鼠血清铜蓝蛋白亚铁氧化酶活性、血清铜浓度、胆汁的铜分泌量、以及肝、肾和脑组织中的铜含量进行分年龄段连续测定和统计学分析。结果 在转基因大鼠体内 ,ATP7B在各种组织中获得广泛表达 ,其中肝和肌肉中的表达水平较高 ,肝细胞恢复了全铜蓝蛋白的合成 ,血清中的铜含量及胆汁的铜排泄量增加 ,肝、肾组织中的铜蓄积减少。结论 完整表达人ATP7B基因产物的转基因大鼠的肝细胞 ,通过CPN介导的铜向血清的分泌和调节铜向胆汁的排泄 ,恢复了ATP7B参与铜转运功能 ,Wilson病的铜蓄积与ATP7B基因的突变直接相关。

    • Experimental Reseach of Effects of Chronic Compression on the Rat Sciatic Nerve

      Abstract:Objective To investigate the effect of chronic compression on the rat sciatic nerve's function and histomorphology. Methods According to the rat model of sciatic nerve compression designed by Mackinnon, the thirty two Sprague Dawley male rats were made the model of sciatic nerve compression as compression group; sixteen rats had the same operation but the silitube was took out as blank control group ;another 8 rats without operation as normal group. Sciatic Function Index(SFI) was determined in both blank control group and compression group every week.At the 2nd,4th week,2nd,4th,6th,8 th week and 2nd week,motor nerve conduction velocity(MNCV) was detected in blank control group,compression group and normal group respectively, with the sciatic nerve(for compression group, the compressed nerve)taken out to perform histomorphology analysis.Results\ At the 4th week, SFI and MNCV of blank control group had no significant difference to that of normal group( P >0 05),but that of compression group had significant difference to that of blank control group and normal group( P <0 05).Myelin sheath was first affected after compression.The changes of its morphology appeared,then follwed by changes of the thickness,at last the axon was affected. Demylination appeared at the 4th week. Conclusions\ The rat sciatic nerve compression model designed by Mackinnon had the strain injury to the nerve.And the compressed nerve showed histological changes of myelin sheath and axon.

    • Genetic Monitoring to Wild Derived TW Inbreeding Mouse

      Abstract:Objective To monitor the genetic quality of the new wild derived inbred strain of mice trapped originally in China. Methods According to national standard, GB/T14927 1-2001, GB/T14927 2-2001 and the procedure from International Laboratory Animal Commission, 25 biochemical markers for genetic monitoring of TW (Tianjin Wild) inbred strain were performed, and skin grafting test was also carried out. Results Measurement of 25 biochemical markers gene loci was performed by electrophoresis and the results revealed that all observed gene loci were homogenous without any polymorphism and mutation. Of the 25 loci observed, 8 were found to show the rare subtypes. The skin grafting test showed that no dropping graft was found during 100 days after transplantion, implying the TW was of isohistogenicity. Conclusion\ The TW inbred strain mouse is high homogenous in the genetic characteristics and it is possible for the TW inbred strain to be a standard model of genetic monitoring of biochemical markers.

    • Isolation of Babesia microti-like Parasites from Rodents in hu-RBC-SCID Mice

      Abstract:Objective To isolate and identify piroplasms from rodents in Japan by using NOD/shi scid mice, and to test its nature to infect human RBC. Methods The rodents' blood specimens were inoculated into NOD/shi scid mice, whose circulating erythrocytes(RBC) had been replaced with human RBCs(hu RBC SCID mice). Results Babesia microti isolated from rodents proliferated within the human RBCs in the hu RBC SCID mice, and one type was identical to the parasites from human. Conclusion The isolates from rodents were demonstrated to be infective for human erythrocytes by inoculation into hu RBC SCID mice.

    • Insulin Resistance and Hypertension Rat Model Induced by Fructose-rich and Its Features

      Abstract:Objective The aim of the study was to create a rat model(Fructose Fed Rats, FFR)of insulin resistance (IR) and hypertension ,and to investigate its features. Methods Six week old male Sprague Dawley rats were fed with either normal rat chow groups(60% vegetable starch, 11% fat and 29% protein, n =35) or fructose rich diet groups(66% fructose, 12% fat and 22% protein, n =35).In all rats, The euglycemic hyperinsulinemic glucose clamp(GC)technique was performed to estimate insulin sensitivity. Blood pressure was elevated with tail cuff method. Results It was showed that blood pressure of the FFR groups was significantly increased compared with control groups. The average rate of glucose clamp,as a measure of insulin sensitivity (M value), and weight were significantly lower in FFR compared with control ( P =0.012-0.002) after 4 weeks or 8 weeks. TG was significantly decreased in the control groups compared with FFR groups ( P =0.036).However, FFA and FINS were the same as TG,with no significant diffrence( P =0.066,0.074). Conclusion This rat model is characterized by typical insulin resistance and hypertension. It may provide a advantageous tool for investigations of insulin resistance and hypertension.

      Abstract:目的 诱导慢性移植物抗宿主病狼疮样小鼠模型。方法 选用 (DBA 2×C57BL 6J)F1小鼠 ,通过尾静脉注射其母鼠DBA 2淋巴细胞诱导模型 ,观察 12周。结果 F1小鼠注射母鼠淋巴细胞后 2周产生自身抗体 ,出现蛋白尿 ,4周血脂、血肌酐、尿素氮轻度升高 ,肾脏病理仅有系膜细胞轻度增生 ,未见间质损害。 8周血生化指标明显改变 ,肾脏病理示肾小球系膜细胞中度增生 ,间质炎细胞浸润和肾小管大量蛋白管型 ,10~ 12周各项观察指标改变明显 ,病理出现局灶或弥漫肾小球硬化 ,免疫荧光示IgG、IgM、C3沿毛细血管壁及系膜区沉积。结论 慢性移植物抗宿主病狼疮样小鼠模型类似人类狼疮性肾炎 ,是良好的狼疮肾炎模型

    • Mensuration of Main Visceral Weight in Clean Mongolian Gerbils

      Abstract:Objective The weight of 11 kinds of organ and body weight in 75 Mongolian gerbils were mensurated in order to study and build biology indexes system of main organs in Mongolian gerbils and provide the parameters for animal experiments. Methods 38 female and 37 male Mongolian gerbils aged 2-3 months were selected and their body weight and main organ weight were separately weighed, and the data were analyzed in statistics.Results There were no significant differences in main organ weight by sexes( P >0 05),and the growth of main organs of animal community was in a good harmony through the analysis of Kendall coefficient. The linear analysis about body weight and organs showed there were obvious linear relations except thymus,thyroid,brain and testis.Conclusions There are no significant differences in the main organ weight of Mongolian gerbils aged 2-3 months by sexes, and whether there is linear relation between body weight and organs lies on different organs.

      Abstract:目的 建立杂种犬肺内源性、外源性急性呼吸窘迫综合征 (ARDS)模型。方法 健康雄性杂种犬 12只随机分为肺内源性ARDS模型组和肺外源性ARDS模型组 ,每组 6只。采用静脉注射油酸形成肺外源性ARDS模型 ,应用十六烷磺基丁二酸钠盐气管内吸入形成肺内源性ARDS模型。动态观察ARDS模型犬气体交换指标和肺不同部位 (肺尖叶、肺心叶和肺膈叶 )支气管肺泡灌洗液 (BALF)中部分炎性介质 (TNF α ,IL 1β,IL 6 )的变化。 结果 肺损伤后模型犬氧合指数明显恶化 ,肺内源性ARDS模型肺尖叶和肺心叶BALF中炎性介质水平明显高于肺外源性ARDS模型。结论 本实验成功建立了肺内 /肺外不同源性ARDS模型 ,肺外源性ARDS和肺内源性ARDS的肺脏不同部位炎性介质释放具有明显的差异

The choosing of a suitable model is the key to increasing the success rate of operation according to the aim of study. This paper will present the history and progress of rat intestinal transplantation models from the aspects of donor procedure, vascular anastomosis,intestinal anastomosis, reconstruction of lymphatic drainage, combined liver/intestine transplantation and compare the features of various models.

      Abstract:犬瘟热病是由犬瘟热病毒 (CDV)感染犬和其他食肉动物的多发性、致死性传染病。CDV属于副黏病毒科麻疹病毒属有囊膜的负链线性RNA病毒 ,基因组长约 16kb。基因组中包括 6个非重叠基因区。它们的编码基因按 3′~ 5′端顺序依次为N P M F H L系列。最新研究表明CDV两个糖蛋白 (H ,F)是宿主免疫系统的主要目的抗原 ,产生中和抗体的重要抗原之一。N蛋白是保守性较强的免疫原性蛋白 ,此外N蛋白上含有T细胞表位。H ,F和N蛋白在犬瘟热的免疫预防方面起重要作用
