Abstract:Objective To study gene therapy through the production of HBsAg transgenic mice model. Methods The pronuclei of FVB mice with exogenous gene, pcDNA HBsAg, were microinjecred, and the injected embryos were transplanted into the recipients. Transgenic positive mice were confirmed by PCR and Southern Blot. ELISA titer was analyzed in transgenic positive mice after gene immunization with pcDNA HBsAg. Results HBsAg transgenic mice were identified by PCR and Southern Blot. The specific antibodies were stimulated after gene immunization. Conclusion HBsAg transgenic mice were produced successfully and gene immunization can stimulate specific antibodies in transgenic mice.

Abstract:目的用PCR技术检测转人组织蛋白酶K基因鼠系,筛选携带目的基因的阳性小鼠。方法用PCR技术,对显微注射后出生的转人组织蛋白酶K基因小鼠、首建鼠的F1、F2、F3及F2代与野生型ICR小鼠的回交后代进行检测。结果共检测显微注射后出生的小鼠25只,阳性为1只,阳性率为4%;检测F1、F2、F3代小鼠分别为60、77和69只,阳性仔鼠数为35、56和63只,阳性率分别为550%、727%和913%;对回交后代进行检测,未见纯合的F2代小鼠。结论外源基因(人组织蛋白酶K基因)已经整合到小鼠的染色体上,并且按孟德尔遗传定律中的分离定律进行遗传。

Abstract:目的建立大鼠骨肉瘤动物模型。方法体内连续传代筛选UMR_106细胞系获得高成瘤率的细胞,接种于SD大鼠皮下。肿瘤组织行常规病理HE染色并镜检,同时检测大鼠血清中碱性磷酸酶的活性水平。结果一周内可见肿瘤形成,荷瘤动物存活时间为28~63d,肿瘤组织镜下可见典型骨肉瘤病理特征,成骨作用明显,荷瘤鼠血清碱性磷酸酶水平显著升高。结论成功建立了可移植大鼠骨肉瘤动物模型,为骨肉瘤的肿瘤免疫研究提供条件。

Abstract:目的观察抗辐射药物E838对IRM_2小鼠、ICR小鼠的辐射防护作用,为研究新型抗辐射药物提供动物模型。方法小鼠在照射前3、2、1d腹腔注射药物,对照组腹腔注射茶油,24h后经137Csγ射线80Gy全身一次性照射,观察照射后30d小鼠的生存天数及保护指数。结果E838100、150、200μg02ml提高IRM_2小鼠的存活率分别为55%、85%、75%,保护指数为199、256、246。提高ICR小鼠的存活率分别为20%、35%、30%。保护指数为178、217、189。结论E838对受γ射线照射小鼠有明显的防护作用。

Abstract:目的考察我室研制的软膏剂猪皮生物敷料对大鼠深二度烧伤创面的愈合作用。方法利用SPF级SD大鼠制备深二度烧伤动物模型,以灭菌油纱布敷料为对照组,观察利用软膏剂猪皮生物敷料对创面愈合作用的影响。结果实验组创面覆盖后渗出明显减少,愈合时间平均为213d,愈合时间较对照组提前了59d。实验组伤后7、9、12、15、18及21d的创面愈合率较烧伤对照组高(P<001)。结论软膏剂猪皮生物敷料促进烧伤创面愈合,对于临床上需要包扎的创面,这种膏剂猪皮敷料是一种很好的选择。

Abstract:目的建立新生乳鼠脑缺氧缺血动物模型。方法将临产(妊娠21d)雌性大鼠,在无菌环境条件下,剖开腹部,用止血钳完全阻断子宫体、子宫角的动脉血管,造成新生乳鼠急性脑缺氧缺血,致使脑部发生病理改变以及存活乳鼠行为的变化。结果随着完全阻断血管时间的延长,新生乳鼠死亡数增加,两者具有直线正相关关系(P<005)。成活的乳鼠与正常分娩的乳鼠,在翻身、爬行、对应激刺激(触摸、扎刺、高低温环境变化)等行为对比差异明显,而且身体发育增长缓慢。结论结扎临产大鼠子宫,可造成新生乳鼠急性脑缺氧缺血动物模型。此法操作简单、快速,脑缺氧缺血彻底,乳鼠脑部病理改变明显。存活的乳鼠出现程度不同的神经麻痹疾病症状。此项实验,可为人类探讨新生儿急性缺氧缺血疾病的病理学、生理学、药物治疗学等诸多方面,提供理想可靠的模型动物。

Abstract:Objective To study the effects of PEC, the oral liquid,which consists of Panax quinquefolium L, Epimedium brevicornum Maxim, Schisandra chinensis Bail and Cervus eplaphus L, on immune system in mice. Methods Lymphocyte proliferation assay and hemolysin concentration (HC 50 ) test were used,clearance rate of charcoal particles and DCH response induced by DNCB were determined. Results Application of 10-20 ml/kg of PEC promoted the lymphocyte proliferative activity, that of 2 5-10 ml/kg of PEC increased the production of serum specific antibody hemolysins, as well as it also enhanced clearance rate of charcoal particles, increased the phagocytic activity of the monocytes when it was given orally for 6-10 days. 2 5-10 ml/kg of PEC improved obviously the DCH response induced by DNCB in immune_suppression mice when it was given orally for 10 days. Conclusion PEC showed the strong immune_stimulating activities in mice.\;

Abstract:Objective To study the effects of gingko on atherosclerosis in rabbits. Methods\ 24 rabbits with atheroscleroses were divided into gingko group(high fat food gingko extract) and control group (high fat food).Results\ Compared with control,the serum TC,TG,MDA in gingko group decrased obviously( P <0.01,<0.05) and the HDL C,SOD level increaed markedly( P <0.01) 8 weeks later.Conclusion\ Gingko is good for preventing and rehabilitating the cardiovascular disease and may be a kind of useful antilipemic agents.

Abstract:Objective To observe the effect of the dry powder of Opuntia in treatment of experimental hyperlipemia Wistar rats. Methods\ The curative effect of the dry powder of Opuntia on blood lipids in normal and hyperlipemia Wistar rats was observed. Results\ After the treatment with dry powder of Opuntia,the level of TC in nomal Wistar rats decreased, showing no significant difference in comparison with normal feed group ( P >0.05). The levels of TC,LDL C,AI( P <0.01),TG( P <0.05) were lower significantly in hyperlipemia Wistar rats after treated with high dose,middle dose,low dose of Opuntia. Conclusion\ The dry powder of Opuntia may regulate the level of blood lipids in normal and hyperlipemia Wistar rats.The effect is more obvious in hyperlipemia rats than in normal rats.

Abstract:Objective To measure and analyse the chief organ weight and coefficients of different species of experimental rabbits.Methods\ In 40 GPB rabbits,CW rabbits and experimental closed colony NZW rabbits bred by Nanjing General Hospital were measured the chief organ weight and coefficient of liver, heart, spleen, lung, pancrease, kidney, adrenal, testis, ovary, womb and oviduct with electronic scales.Result\ No significant differences were observed in all indications between male and female ( P >0.05) except the lung weight of GPB and the lung coefficient of NZW( P <0.05).Among the species,coefficients of liver,spleen,lung,pancrease,adrenal showed significant differences( P <0.05), and the others P >05.Between the GPB and CW,all organ coefficients were with no significant differences( P >0.05).Conclusion\ The chief organ weight and coefficients of experimental rabbits have some correlativity with species characteristics.

Abstract:Objective To acknowledge the Beagle's normal levels of hematology and blood biochemicals. Methods 10 blood biochemicals and 9 blood routine values of Beagles from different origins and ages were examined by BSALC partial blood automated chemical analyzer and German made Medicon 4000 fully automated hematology analyzer. Results The results were coordinated with those reported in the published articles, fluctuating in the rang of normal levels except the high level of ALP, compared with the ordinary human specimen. Conclusion The experimental animal of different origins and ages has a relation with the results. It's necessary to pay close attention to the coordination of the origins and ages of the experimental animals during the experiments.

Abstract:目的对小鼠卵巢玻璃化冷冻的方法和冷冻液进行研究,摸索出一种简便的小鼠卵巢冻存法。方法用玻璃化冷冻液EFS40对Lats1基因敲除杂合子小鼠卵巢进行玻璃化冷冻,冷冻效果通过解冻后的卵巢肾包膜下易位移植、卵巢囊原位移植和对移植后的成活卵巢进行组织学检查来判定。对出生的后代进行Lats1基因检测,来判定卵巢原位移植结果的可靠性。结果与结论解冻后的卵巢肾包膜下移植733%成活,并具有正常的功能;卵巢囊移植后卵巢存活率为467%,受体怀孕率为333%,平均产仔数为(37±22)只,后代中能检出Lats1基因;试验中还发现,受体的发情时间普遍提前10d左右。

Abstract:目的分析VcⅠ系獭兔(VcⅠ)、VcⅡ系獭兔(VcⅡ)、青紫蓝兔(QZL)、新西兰兔(NZR)和日本大耳白兔(JWR)5个品种(系)家兔群体内遗传变异和群体间遗传距离。方法每个品种(系)兔随机抽取16只,综合利用微卫星多重PCR、DNA池、复合点样和快速银染技术。结果建立起了一套快速分析家兔群体遗传变异的方法。5个品种(系)家兔群体内遗传变异情况为VcⅡ>VcⅠ>JWR>QZR>NZR,聚类结果为VcⅠ与VcⅡ亲缘关系最近,与JWR、QZL亲缘关系较近,与NZR亲缘关系最远。结论实验所得的各群体家兔遗传变异结果与Vc獭兔实际育种事实相符。

Abstract:Sperm mediated gene transfer (SMGT) is developed, a new method to generate transgenic animal based on traditional transgenic technique, because SMGT does not require special equipments, specific skills, and it merely needs simple operation. It is expected to be a potent way to generate transgenic animal. This review is a brief summary of basic principal, the most resent research progress and application perspectives of SMGT.

Abstract:Suture occuluded method for model of focal celebral ischemia in rats was reviewed according to the literature over 10 years. It is regarded as more ideal method at present and can be used in the basic and clinical study of cerebral and vascular diseases. The further study should be placed on controlling factors affecting the model duplication to make it more standardized, and to benefit the further discussion of pathological and physiological mechanisms in cerebral ischemia and the evaluation of the precise protective effect of traditional Chinese medicine on brain.
