Abstract:目的 研究肾癌细胞致敏的小鼠腹股沟淋巴结T细胞在体外经抗4 - 1BB(肿瘤坏死因子受体家族成员)单克隆抗体诱导刺激后IFN γ的表达水平。方法 以BCG和高聚生作为佐剂,用6 0 Co射线灭活的小鼠肾癌细胞Renca刺激小鼠腹股沟淋巴结,致敏后的淋巴结(肿瘤引流淋巴结tumor draininglymphnode,TDLN)T淋巴细胞在体外联合使用IL 2、抗CD3、抗CD2 8、抗4 1BB小鼠单克隆抗体激活,然后利用流式细胞术(FACS)检测细胞内IFN γ(干扰素Ⅱ型)的表达水平。结果 应用4 - 1BB单抗刺激的实验组有抗瘤活性的T细胞比例明显提高,其中T淋巴细胞分泌IFN γ的水平达到4 8.2 3% (P <0 .0 1)。结论 抗4 1BB单抗体外刺激肿瘤细胞致敏的小鼠腹股沟淋巴结T细胞后,T淋巴细胞内表达IFN γ水平提高。该工作为肾癌的肿瘤疫苗研制提供了实验依据

Abstract:目的 探讨大鼠胰岛细胞分离纯化和培养的方法。方法 采用胰导管逆行灌注Hanks液分离成年大鼠胰岛,Ⅴ型胶原酶消化,不连续密度梯度Ficoll 4 0 0纯化胰岛。用台盼蓝和双硫腙染色检测胰岛活性和纯度,葡萄糖刺激胰岛素释放试验检测胰岛功能。结果 胰岛细胞的收获量为5 4 0±15 0个胰岛 胰腺,活性>90 % ,纯度>90 % ,高糖刺激后胰岛素释放量为低糖刺激的2倍多。结论 胰导管灌注Hanks液,Ⅴ型胶原酶消化和不连续密度梯度葡聚糖纯化胰岛,可以获取数量多,活性和纯度好的胰岛

Abstract:目的观察不同电荷性质氨基酸对清醒兔血压及其心血管系统一氧化氮合成反应的影响。方法L-广脯氨酸、L-广丝氨酸、L-广丙氨酸、L-广甘氨酸、L-广精氨酸、L-广赖氨酸的生理盐水溶液,以及L-广精氨酸与L-广脯氨酸混合物的生理盐水溶液,L-广赖氨酸与L-广脯氨酸混合物的生理盐水溶液,通过颈内静脉插管注入处于清醒状态的实验兔体内,颈总动脉插管观察血压的变化;测定氨基酸注入前后血清中的亚硝酸盐水平。结果①L-广脯氨酸、L-广丝氨酸、L-广丙氨酸和L-广甘氨酸具有升压效应,L-广精氨酸和L-广赖氨酸具有降压效应,L-广精氨酸或L-广赖氨酸与L-广脯氨酸在血压调节上存在相互拮抗的作用。②L-广脯氨酸、L-广丝氨酸和L-广丙氨酸注入引起血清亚硝酸盐测定值下降,L-广甘氨酸注入对血清亚硝酸盐测定值没有影响;L-广精氨酸和L-广赖氨酸注入引起血清亚硝酸盐测定值的升高;L-广精氨酸或L-广赖氨酸与L-广脯氨酸同时注入亦引起血清亚硝酸盐测定值的升高。结论机体内存在某种电荷依赖性的血压调节机制,该机制在一定程度上通过促进或抑制心血管系统一氧化氮合成反应发挥其血压调节作用。

Abstract:目的给新生鸡雏进行铅染毒,观察铅引起的红细胞核浓缩情况。方法设立标准饲料对照组、标准饲料染毒组、低Ca对照组以及低Ca染毒组。染毒后的不同时间点,测定血铅浓度及细胞核浓缩情况。结果标准饲料铅染毒组第2天血铅浓度达到最大值57.7μg/g;低钙铅染毒组血铅浓度第4天达到最大值37.2μg/g;标准饲料染毒组与低Ca染毒组同时在第4、6、8天观察到显著的核浓缩,核浓缩在染毒第6天达到高峰。约20%红细胞核浓缩,同时两染毒组的无核红细胞显著增加。观察低Ca对照组与低Ca染毒组的红细胞DNA电泳,低Ca染毒组DNA呈梯凳状,被分割成大小不等的DNA片段。结论铅直接作用于鸡红细胞,引起细胞核浓缩。预示红细胞核浓缩是脱核的一个过程。

Abstract:目的 优化中国地鼠RAPD分析体系。方法 以30条引物对山医群体中国地鼠的两个家系基因组DNA在各种不同条件下进行RAPD ,分析比较扩增结果。结果 在下列反应体系及温度循环参数下结果较好:在2 5 μl的反应体系中含有10×buffer 2 5 μl,dNTP 0 2mmol L ,Mg2 2 0mmol L ,引物0 2 μmol L ,Taq酶1 5U ,模板DNA 15ng ,扩增程序为94℃预变性3min后进入循环体系,94℃变性30s ,36℃退火5 0s ,72℃延伸1min ,4 0个循环后接7min延伸。结论 以上述反应体系及温度循环参数进行中国地鼠RAPD ,扩增结果稳定

Abstract:Objective To establish a method of mouse artificial ejaculation for repeating collection of sperm from mice, which is carried out in many experiments with semen. Methods Three ways were compared including massage with hand (MH), artificial vagina pump (AVP) and electro-ejaculation (EJ). Results AVP and EJ could induce ejaculation, and their success rate, average semen quantity and motility showed no significant difference. Repeated tests got the same data. Mouse ejaculation was divided into four stages with different cell quantity. MH produced no ejaculation. Conclusion AVP and EJ were feasible ways for mouse ejaculation.

Abstract:Objective To explore the establishment and postoperative mangement of 2K2C(two kidneys two clips) renovascular hypertensive model of rats.Methods 150 male Sprague-Dawley rats weighing 80 to 120 g were selected to make the 2K2C renovascular hypertensive model.Within 10 weeks after the operation of renal arterial constriction,the postoperative complication,the mortality,the blood pressure were recorded and the spontaneous strokes were confirmed by brain slides examination. Results After the renal arterial constriction,all rats developed hypertension one month later and celiac hemorrhage and infection were the most common complications of the operation. The causes of deaths included postoperative celiac hemorrhage(two cases),hyperanaesthesia(3 cases),incurable severe infection(3 cases),renal failure(5 cases), severe malnutrition unknown causes(2 causes). Brain slides examination was made in two spontaneous hypertensive rats, with basal ganglia hemorrhages confirmed. Conclusion Proficient surgical technique is the key factor in making the 2K2C renovascular hypertensive model of rats and the intensive postoperative manegement can decrease the mortality.

Abstract:Ventilation and air- conditioning system of all fresh airflow is usually adopted in the SPF experimental animal facility. In order to retrieve the energy of heat or cool of the system's exhausted airflow, which can be used again to heat or cool the fresh airflow in advance, gravitative low temperature heat pipe transdcers was installed in the ventilation system. Several years running has proved that it can reach the purposes of retrieving energy.

Abstract:Selectins are a family of three cell adhesion molecules:L-,E-and P-selectin. They specialize in capturing from the bloodstream to the blood vessel and mediating roll of leukocytes on the endothelia cell surface. The leukocytes migrate into the inflamed tissue at last. This review focuses on the mechanisms that regulate the express and function of the selectins and their ligands.

Abstract:目的回顾应用果蝇作为学习记忆模式动物的研究工作。方法文献调研。结果经实验室筛选出dnc、rut、amn、rad、cabbage、turnip、radish、nalyot、turnip、nemyp153、ala等表现条件反射行为障碍的突变体,且已证实与cAMP信号转导途径有关。经现代免疫组化研究发现蕈状体/蘑菇体(mushroom bodies)有多种基因产物的优势表达,成为果蝇的嗅觉、视觉学习和记忆过程起关键作用的神经解剖区。果蝇的学习记忆与个体发育程度有关。果蝇幼虫训练形成的条件性嗅觉回避反应,在幼虫蜕变为成虫后(8d)仍然存在,这为研究长时记忆的机制提供了一个有用的模型。dnc和amn突变体不能形成这一长时记忆。结论遗传学背景较为清晰的果蝇,由于具有各类学习记忆的能力,已成为研究学习记忆机制的重要实验动物。

Abstract:This paper summarizes recent progress in transgenic swine in Biotech Institute of Hubei Academy of Agriculture Science. The Seven individual foreign genes with different construction, that is, POMT-PGH, PSV40-PGH, POMT-PGH-Micro satellite DNA, PMHR, hDAF, pBHSA, pSHSA, was microinjected into 4027 embryos. 83 founders were produced. Transgenic rate was 0.9 to 3.4% and 2.2% on average. Through improving main technological process, we improved transgenic rate, established the technology of hybrid in situ of chromosome, detection of foreign gene quantity by PCR and Dot blot and Southern blot and Northern blot for expression protein, investigated the genetic regulation of transgenic swine.

Abstract:家族性心肌肥大症是发生频率很高的遗传病,其发生率在0.2%~0.5%之间,中国有多达600万的突变基因携带者。家族性心肌肥大症与两类基因的突变有关,一是心肌蛋白相关基因,目前已发现10种,其中主要是β-肌球蛋白基因、肌球蛋白结合蛋白-C基因和肌钙蛋白结合基因,占患者总数的75%以上。二是非心肌蛋白基因,包括线粒体基因组DNA、线粒体蛋白基因。本文对引发家族性心肌肥大症的分子遗传机理进行了总结。

Abstract:Degenerative retinal disease in laboratory and domesticated animals represents a wealth of different pathological manifestations which are important for understanding the basis for human disease. Animals with degenerative retinal disease, homologous to that found in humans, make useful models for investigation of treatment by retinal transplantation, pharmaceutical and gene therapy.

Abstract:Parkinson disease (PD) is a neurodegenerative disease characterized with bradykinesia, tremor, rigidity, and abnormal postural reflexes due to lack of dopamine in the substantia nigra-striatum system resulting from the degeneration and necrosis of dopaminergic neurons. Experimental research is required in order to increase our knowledge on the cellular mechanism and functional consequences of this degenerative process. These models allow investigations of new therapeutics in order to improve the treatment of patients or to test new drugs for protection of any remaining dopaminergic neurons. The present article looks at the relative merits and limitation of each model, and the neuronal impairment it induces. It is becoming more and more clear that there is an urgent need for a chronic model integrating all the clinical features of PD including the gradual but continuous nigral degeneration observed in the human pathology.
