    • Apoptosis and Significance of Kupffer Cells in the Liver and Hepatocarcinoma Tissues after Ischemia and Reperfusion

      Abstract:目的研究肝癌组织缺血再灌注后一氧化氮(NO)和一氧化氮合成酶(iNOS)的改变、枯否细胞(Kupffer cell)的凋亡及其关系。方法通过超声引导将VX2肿瘤组织混悬液穿刺注射到新西兰兔肝脏左中叶,建立肝脏肿瘤模型,用无损伤血管钳阻断肿瘤所在肝叶的肝动脉分支60 min后去除血管阻断恢复血流,随机将模型动物分为缺血再灌注前(对照)、缺血再灌注后0 min1、h1、d3、d、1周6个时间组,取肝脏组织和肿瘤组织,分别测定NO和NOS的含量。用HE染色法观察肝组织和肝癌组织中的枯否细胞凋亡情况。结果肝组织再灌注后0min开始NO和iNOS浓度均明显下降,至再灌注7 d仍处于较低水平(44.41±2.0,3.70±0.2)。在肝癌组织,除NO浓度下降外,iNOS浓度从缺血再灌注0 min至7 d则明显升高(18.45±1.9)。肝组织和肝癌组织的枯否细胞凋亡显著增加,于再灌注1 d和1 h分别达最高峰(13.69±3.1,13.66±1.8)。结论枯否细胞对肝癌组织缺血再灌注过程中的iNOS及NO的产生、细胞损伤与凋亡起重要作用。

    • Comparison of Biological Characteristics of Bone Marrow Stromal Cells in Fetal and Adult SD Rats

      Abstract:Objective To compare the biological properties in vitro of bone marrow stromal cells(BMSC) derived from fetal and adult rats.Methods\ The second passage cells primarily cultured from 3 weeks and 3 months old rat femoral marrow were induced by osteogenesis medium and compared with respects to cellular growth observed by phase-contrast microscopy.The proliferation capacity was determined by MTT method.Mineralization in vitro was assessed by Von Kossa and Alizarin red staining.Total protein level,expression of mRNA of collagen I was detected by RT-PCR and ultrastructure was examined by transmission electron microscopy.Results\ Cells from fetal rats grew faster than those from adults,their doubling times were 4.8 d and 5.4 d,respectively.After seven days many colonies were formed among fetal cells,which could not be observed among adult cells.Strong positive cells stained by von Kossa and Alizarin red and total protein level of the former were more frequently observed than those of the latter.Expressions of mRNA of Collagen I were demonstrated by agarose electrophoresis by Day7 and many intra-cellular reticula and precollagen extracellular matrices were showed by electron microscopic examination.Conclusion\ Both cells can be transformed into osteoblasts in vitro,but considering the proliferation capacity and osteogenic potency,BMSCs derived from fetal rat are most suitable for studies on cell /biomaterial interaction.

      Abstract:目的构建中枢神经系统特异表达的野生型及突变型α-synuclein转基因表达载体.方法通过PCR方法扩增α-synuclein基因,产物亚克隆到pGEM-T载体,经测序无误后克隆到pCEP4载体,并用神经特异的PDGF启动子替代pCEP4载体的CMV启动子构建野生型转基因表达载体.利用大引物突变法构建A53T和A30P的转基因表达载体.结果通过限制性内切酶及DNA测序证实野生型及突变型α-synuclein均成功的克隆到所需要的载体中.结论该转基因表达载体的成功构建为帕金森氏病转基因动物的构建及帕金森氏病发病机制的研究奠定基础.

      Abstract:目的研究健乳灵对乳腺增生大鼠乳腺组织中抑癌基因p16、p53表达的干预作用.方法用苯甲酸雌二醇和黄体酮制作乳腺增生病大鼠模型,分别用不同剂量的健乳灵治疗60 d,用免疫组化法测定乳腺组织的p16、p53表达水平.结果健乳灵能抑制乳腺上皮增生,降低大鼠乳腺增生组织的p16、p53蛋白表达水平,与模型组比较有显著性差异(p<0.01).结论健乳灵对大鼠的乳腺增生病有调节和治疗作用.

      Abstract:目的通过定量分析雌激素受体在动脉粥样硬化兔心脏和肝脏内的相对基因表达,研究雌激素受体基因mRNA的表达水平与动脉粥样硬化(AS)发生的关系。方法手术法去势新西兰雌兔,高胆固醇饮食法复制AS模型,以管家基因-βactin作为内源性校准基因,用SYBR Green I作为荧光染料,采用实时荧光定量逆转录聚合酶链式反应(real-time RT-PCR)对ERαmRNA的表达进行了分析。结果对照组(N)、模型组(C)和假手术组(SC)主动脉硬化斑块面积分别为0、(0.75±0.24)和(0.56±0.27),饮食法复制AS模型成功;ER在对照组兔心脏和肝脏内的相对基因拷贝数分别为0.46、0.49,在模型组的去势雌兔心脏和肝脏内的相对基因拷贝数分别为0.29、0.32,在假手术组兔心脏和肝脏内的相对表达量分别为0.53、0.51。结论兔心脏、肝脏内都有雌激素受体α分布;去势AS雌兔组织内雌激素受体表达量的减少可能与动脉硬化的形成有关。

    • Establishment of a Model of Simultaneous Pancreas-kidney Transplantation with Portal-enteric Drainage in Miniature Pigs

      Abstract:目的建立符合正常生理特点的胰岛素门静脉回流胰液内引流式胰肾联合移植(SPK)动物模型。方法选用20头近交系甘南小型猪作为SPK的供受体。供胰肾原位灌注后整块切取。修整时行供体左肾静脉与脾静脉端端吻合。受体血管吻合采用供体门静脉与受体脾静脉端侧吻合,供体腹主动脉的远端与受体的腹主动脉斜侧吻合;供体十二指肠与受体空肠侧侧吻合,使胰液经节段十二指肠从肠道引流;供肾输尿管皮肤造瘘并置管体外引流用于监测尿量。全组均未作抗排斥处理。结果10头受体猪中除1头因严重的腹胀于术后1 d死亡外,其余9头平均存活(4.5±1.5)d,术后移植胰、肾均即可发挥功能。结论在小型猪建立门静脉回流胰液内引流式SPK模型是可行的,且该术式更符合生理特点,可望用于临床。

    • Antilipemic Effect of Extracts of Ampelopis Grossedentata and Lotus Leaf (EAGLL)on Rats with Hypercholesterolemia

      Abstract:Objective To establish a rat model of hypercholesterolemia,and examine whether oral administration of extracts of Ampelopis grossedentata and Lotus leaf(EAGLL) can reduce level of TC of rat.Methods\ 50 rats of hypercholesterolemia were divided randomly into five groups: low dose group of EAGLL 5 mg/kg,moderate dose group of EAGLL 10 mg/kg,high dose group of EAGLL 20 mg/kg,model control group fed with a high cholesterol diet,medicine group of oral cholesterol-reducing medicine 180 mg/kg.All test groups of were administrated continuously for 28 days.TG,TC,HDL-C,weight,fat and Lee's index were measured.Results\ Blood biochemical analysis showed TC value was lower after EAGLL administration than before in the moderate and high dose groups(P<0.05).Lee's index was lower in high dose group than in model control group(P<0.05).Conclusion\ EAGLL decreased value of cholesterol and Lee's index of rat model of hypercholesterolemia.

    • Effect of Dibutyltin Dilaurate on Activity of Enzyme in Testis Tissue of Male Rats

      Abstract:Objective To study the effect of dibutyltin dilaurate(DBTD) on reproductive system of male rats.Methods 28 healthy male rats were divided into 4 groups,7 each.The rats were administrated orally with DBTD at the dose of 0,5,10 and 20 mg/kg,respectively,and killed after 5 weeks for measurement of ACP,LDH,NO,NOS and tin content in testis tissue.Results There was no difference in different dose groups of tin content in testis tissue compared with the control group,but the activity of LDH,NO in dose groups was different from control group.At the same time,the content of NOS in 10,20 mg/kg groups and the activity of ACP in 20 mg/kg group were also different from the control group.Conclusion The study shows that DBTD has testis toxicity,and it can disturb the activity of enzyme in testis tissue.

    • Red Cell Immune Adherence Function, IL-6 and sIL-2R Levels in Experimental Epileptic Rats

      Abstract:Objective To investigate the changes of red cell immune adherence function,IL-6 and sIL-2R levels in experimental epileptic rats.Methods Healthy Wistar rats(n=40) were divided into control group(n= 10,treated with saline) and model group(litium-pilocarpine-treated group,n =30).The red cell immune adherence function according to RBC-C3bR rosette-forming ability was measured by red cell C_(3b) receptor rosette test.IL-6 expression was determined by radioimmunoassay and sIL-2R measured with sandwich enzyme linked immunosorbent assay(ELISA).Results There was a statistically significant difference in RBC-C3bR and RBC-IC between the two groups(p< 0.05),and the differences of IL-6 and sIL-2R were very significant(p<0.01).Conclusion Our findings indicate that immune factors probably participate in the epilepsy pathogenesis and epilepsy may lead to immunological alterations.

      Abstract:Objective To obtain the basic data of biological parameters of cleaning inbred strain BALB/c mice.Methods\ Routine methods and instruments were used to determine the biological parameters of 2 months old cleaning inbred strain BALB/c mice and immunological parameters of conventional inbred strain BALB/c mice.Results\ Similarities are shown in hematophysiological indices,biochemical indices,avoirdupois,weight of viscera,and index of viscera.Between cleaning BALB/c mice and conventional BALB/c mice,there are significant differences in hematophysiological indices and biochemical indices.On CD4~ /CD8~ indices,there are significant differences between male and female in conventional BALB/c mice,also between cleaning BALB/c mice and conventional BALB/c mice.Conclusion Biological parameters of cleaning inbred strain BALB/c mice are stable,owing to strictly controlled microorganisms and parasites;and some resulting influences on immunological function of mice.

      Abstract:目的探讨磷烧伤与肝细胞及其线粒体损伤之间的关系.方法健康wistar大鼠随机分为磷烧伤组、烫伤组、正常对照组.每致伤组每时相点6只,分别于伤后5、10和24 h放血活杀,取血测定丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH).同时取肝组织,检测肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)和谷胱甘肽(GSH)活性以及线粒体呼吸控制率(RCR)、线粒体氧化磷酸化效率(pi/O)、线粒体琥珀酸脱氢酶(SDH)活性变化.结果磷伤后血ALT、GOT、LDH不仅显著高于对照组,也显著高于烫烧伤组(P<0.05);磷烧伤组RCR、Pi/O及SDH活性显著低于对照组与烫伤组(P<0.05);磷烧伤组肝组织SOD,GSH低于烫伤组但显著高于对照组(P<0.05);MDA显著高于对照组但低于烫伤组(P<0.05).结论肝细胞及线粒体的损伤不仅有过氧化损伤因素起作用,还有磷转化为磷酸后对肝细胞及其亚结构的毒性作用.

      Abstract:Objective To set up the normal value of cardiac hemodynamic variables and left ventricular systolic function for healthy Macaca mulatta.Method\ Cardiac hemodynamic variables and left ventricular systolic function indexes of 10 healthy adult Macaca mulatta(4 female,6 male) were measured separately by pulsed Doppler and M-mode echocardiography.All indexes were compared between both sexes.Results\ All indexes of Macaca mulatta were similar to that of human newborns.And all indexes between the males and females were not statistically different.Conclusion\ The cardiac hemodynamic variables and left ventricular systolic function indexes of Macaca mulatta are considered to be normal physiologic value.

    • A Comparison Study between the SIVmac251 Titer in Cultured PBMC and the Plasma Viral Load after Infection

      Abstract:Objective To explore a method which is being used to screen applicable monkeys for the SIV modeling.Method The pre-infection monkey's PBMCs were cultured and infected with SIVmac251,then,the viral level was tittered and compared with the plasma viral load from the same monkey which infected with the same SIVmac251.The results were analyzed and evaluated whether it may be used to screen the applicable monkeys for SIV model.Result According to the data of 14 monkeys both in vitro and in vivo,the coincidence rate was 71.43%.There were 6 monkeys(42.9 %) with lower viral load in vivo and 4 monkeys with lower viral titer in vitro.If these 4 monkeys with lower viral titer were ruled out,there remained 10 monkeys,and there were only 3 monkeys(30%) with lower viral load among them.Conclusion By using of the method to detect PBMC viral titer in vitro,it might be helpful to choose the higher titer monkeys for modelling SIV infection efficiently.However,it isn't a perfect method.Establishing the clear genetic file of SPF monkey colony may be a reasonable way to get satisfactory monkey model for research use.

      Abstract:Objective To establish an animal model with a close similarity of the clinical pelvic inflammation.Mathods Healthy adult New Zealand rabbits were used in this experiment.Escherichia coli was inoculated on the surface of the mesosalpinx and injected into fallopian tubes.7 days later,the degree of pelvic inflammation was measured by gross observation,light microscopy,and electron microscopy.Results Pelvic marked hyperaemia,oedema,purulent discharge and adhesion were observed in peritoneum and pelvic organs.Histologic evaluation revealed significant peritoneal inflammation.Conclusion A reproducible animal model of pelvic inflammation could be established and used in experiment reseach.

      Abstract:Objective To develop a method with high sensitivity,specificity and rapidity to detect pathogenic dermatophytes in laboratory animals.Methods One fragment of CHS1 gene was selected as universal primer of dermatophytes in this PCR amplification.Otherwise,a random primer FM1 was also used to amplify three dermatophytes species standard strain: Trichophyton mentagrophyton,Microsporum gypseum,Microsporum canis.Results There were distinguishable fingerprinting patterns among three dermatophytes species.Conclusion The test could detect some common dermatophytes species of laboratory animal rapidly and specifically.

      Abstract:The mitochondrion of animals is a organelle appearing in cytoplam of eukaryotes. According to the complete sequences that have been sequenced,it contains genes encoding 13 proteins,2 rRNAs,22 tRNAs and one major non-coding region(D-Loop).It is reported that a R-Loop structure has been found,with the characteristics of maternal inheritance,and the average evolution rate is 10 times faster than that of nuclear genome,especially D-Loop.The existence of tandemly repeat sequences in control region of mitochondria would be regarded as a special molecular marker in animals researches because of the interspecies and intraspecies differences.According to the structural and genetic characteristics of mtDNA,by means of advanced bioinformatics and rich databases,the researchers have made great progress in the field of molecular phylogenetics of animal populations,and make certain taxonomic status,interspecies and intraspecies relationship.
