    • Protective Effect of Anti-SARS-CoV Antibody on SARS-CoV Infection in Macaca Mulatta

      Abstract:Objective The SARS is an infectious disease which was newly conformed in China. The effective clinical treatment for this disease is still being under studying, The aim of the present study was to investigate the protective effect of antiSARS-CoV antibody on SARS-CoV infection in Macaca mulatta monkey model. Methods To analyze changes of anti-SARS-CoV antibody treated SARS-monkeys. Results The aquired neutralization antibody for SARS-CoV inhibited the replication of the virus to a certain extent. The pathological changes of the antibody treatment group were lighter than those in the SARS-monkey group. Conclusions Our findings suggested that anti-SARA-CoV neutralization antibody therapy is a potentially useful clinical treatment for SARS.

      Abstract:目的血管紧张素转换酶是肾素-血管紧张素系统的重要组成部分。血管紧张素转换酶主要功能是通过把血管紧张素Ⅰ转换成血管紧张素Ⅱ来调节血压,最新的研究显示它也是SARS冠状病毒的一种受体。建立血管紧张素转换酶2基因剔除小鼠,为进一步的SARS机理研究提供工具。方法通过将D3小鼠胚胎干细胞DNA进行同源重组,用反向表达的新霉素基因替代ACE2基因-2000bp至+489bp的DNA片段,并用Western blot检测纯合体小鼠ACE2基因的表达。结果血管紧张素转换酶2基因剔除小鼠ACE2基因的表达被完全阻遏。结论建立了ACE2基因剔除小鼠。

      Abstract:Objective ACE2 is a receptor of SARS-CoV. In this paper, the replication of SARS-CoV in murine fibroblasts expressing human ACE2 and ACE2 knockout mice was studied. Methods The murine fibroblasts expressing human ACE2 and ACE2 knockout mice were used to investigate the replication of SARS-CoV and protective effect of ACE2 knockout on SARS infection in mice. Results The SARS-CoV infected and replicated in the murine fibroblast cells expressing human ACE2. The Block of ACE2 expression inhibited the infection and replication of SARS-CoV in vivo. The ACE2 gene knockout showed a protective effect on SARS-CoV infection in mice. Conclusion Our results suggested that ACE2 is a functional receptor for SARS- CoV and a possible target gene for SARS therapy.

    • Comparative Study on Pathogenesis of SARS Pulmonary Fibrosis

      Abstract:Objective To explore the possibility of applying SARS-CoV infected animal model to clarify the pathogenesis of pulmonary fibrosis(PF) in SARS.Methods We compared the difference and similarity of SARS features and pathogenesis between SARS patients and SARS animal model emphasized on PF.Results Beginning from alveolitis,PF in different diseases has similar pathological changes in a progressive and reparative process.About 7-8% of SARS convalescent patients showed apparent PF,which affected their life quality and increased medical treatment costs.PF in SARS patients appears in the early stage and develops very fast.These patients died within a few days with severe pathological changes in many organs,especially in the lungs.SARS-CoV infected animal model had been established,which showed similar clinical,serological and pathological changes to those in SARS patients.In these animals,PF is also apparent with increased amount of fibroblasts and collagen.Conclusions SARS-CoV infected animal models are useful in studies on pathogenesis of pulmonary fibrosis in SARS patients.

      Abstract:Objective To establish the SH-SY5Y cell line stably expressing vesicular monoamine transporter 2(VMAT2).Methods An eukaryotic expression vector of VMAT2 was constructed and then transfected into SH-SY5Y cells by Lipofectamine.The expression of VMAT2 in SH-SY5Y cell line was detected by Western-blot and immunofluorescence technique.Double immunofluorescence technique was used to determine the localization of VMAT2 in the cells.Results VMAT2 was highly expressed in SH-SY5Y cell line revealed by Western-blot and immunofluorescence and was found to locate on LDCV shown by double immunofluorescence technique.Conclusion This work has laid a good foundation for the further studies on the role of VMAT2 in Parkinson's Disease(PD).

    • Histopathological Observation on Early Injuries of Multiple Organs in Severely Burned Rats at Plateau

      Abstract:Objective To investigate the severity of early injuries and pathological changes of liver, kidney and small intestines in rats with severe burn at plateau.Methods Forty eight Wistar rats with 30% full thickness burn were randomly divided into 6 groups(n=8) and another 8 healthy Wistar rats as control group.The liver,kidney and small intestine were harvested for histopathological examination at 3,6,12,24,48 and 72 hours after burning.Resulte The injury of multiple organs in rats was obvious at 3 h post-burn and became severe at 48 h-72 h post-burn.Conclusion The injuries of multiple organs,especially the liver,kidney and small intestine in rat with severe burn at plateau occur early and last for a long time.This condition of injuries are of a direct correlation with the influence of hypoxic environment at plateau.

      Abstract:目的探讨不同山羊个体的精子结合和内化外源DNA能力差异性,以及精子内化能力对制备转基因山羊胚胎阳性率的影响。方法用地高辛标记与检测试剂盒检测12只山羊的精子分别结合、内化质粒DNApEGFP-N1的效率;体外受精实验检测胚胎阳性率与精子内化pEGFP-N1能力的关系。结果不同山羊个体的精子结合和内化pEGFP-N1的效率存在差异,内化效率最高为53.7%,最低为3.1%;3号公羊的精子体外转染外源基因后行体外受精,所得的2~8细胞胚胎表达外源基因pEGFP-N1的阳性率最高,7、8、10、11号公羊的精子体外转染外源基因后行体外受精,所得的2~8细胞胚胎均未表达GFP蛋白。结论不同山羊个体的精子自发结合、内化外源DNA的能力存在差异;不同山羊个体的精子体外转染外源基因后行体外受精,所得的2~8细胞胚胎表达外源基因的比例不同。

      Abstract:目的为犬线粒体DNA应用于系统进化和法医学物证鉴定提供基础。方法对106只无亲缘关系的犬线粒体DNA高变区Ⅰ(1041—1318)进行测序。结果测序得到了23个单倍型,它们的差异由24个突变点产生,其中包括碱基转换、颠换及插入。大多数突变点是碱基替代,转换为79.2%,颠换为16.7%,碱基插入为4.1%。结论犬线粒体DNA高变区Ⅰ序列,遗传差异度(相当于杂合度)为0.89,两个无关个体的偶合概率为0.1213,具有高度序列多态性。

      Abstract:Objective The distribution and morphology of 5-HT immunoreactive endocrine cells in the digestive tract of Lacerta vivipara Jacquin were investigated.Methods ABC immunohistochemical technique was applied.Results The 5-HT cells distributed throughout the digestive tract,from esophagus to rectum.The distribution density curve appeared waveform and there were three density peaks: cardiac part of the stomach,pylorus of the stomach and ileum.The 5-HT cells were round, ellipse,shuttle-like or tapering in shape,mainly round and ellipse,and distributed widely at the basal region of epithelium,between glandular epithelial cells and between acini.Conclusion In view of their morphology we would suggest that 5-HT cells have both endocrine and exocrine functions. A comparison of the distribution density of 5-HT cells in the digestive tract between sevtral few reptiles suggest that the difference of their distribution density might be related to feeding habit and surroundings as well.

    • Establishment of an Experimental Canine Model of Right Ventricle and Pulmonary Artery Reconstruction

      Abstract:Objective To investigate the method to establish laboratory animal model of right ventricular outflow tract and pulmonary artery reconstruction(RV-PA)in dogs.Methods The bovine jugular vein conduit(BJVC) that was fixed with polyexpoxy compound(PC) was implanted between the right ventricle and pulmonary artery in ten young dogs.Right ventricular and pulmonary artery pressures were measured directly before and after implantation.Results Seven dogs survived one month after implantation.The systolic pressure, diastolic pressure and mean pressure of the right ventricle and pulmonary artery were not significantly changed after reconstruction with the conduits.Conclusion An animal model of RV-PA reconstruction could be successfully established with PC-fixed BJVC in dogs.

      Abstract:目的研究补阳还五汤对脑缺血再灌注大鼠皮质神经细胞凋亡的影响。方法36只SD大鼠随机分为3组,每组12只:①假手术组;②模型对照组;③补阳还五汤组。线栓法制作大鼠脑缺血再灌注模型,脑缺血-再灌注48h后TIC染色计算脑梗塞体积,TUNEL染色检测神经细胞凋亡,免疫组化方法检测缺血侧皮质Bcl-2/Bax的表达,电镜观察线粒体的变化。结果脑缺血再灌注后缺血侧皮质神经细胞凋亡增多,同时Bcl-2表达减少,Bax表达增多,Bcl-2/Bax比值下降,线粒体肿胀且大多破裂。补阳还五汤干预后,细胞凋亡减少,脑梗塞体积减小,Bcl-2/Bax比值上升,线粒体肿胀减轻。结论补阳还五汤可能通过抑制神经细胞凋亡而减小脑梗塞体积,其机制可能是通过上调Bcl-2/Bax比值,保护线粒体,防止发生线粒体通透性转变。

    • Development of Combined Hypercholesterolemic/ Hypertriglyceridemic Mouse Model

      Abstract:Objective To establish a combined hypercholesterolemic/hypertriglyceridemic mouse model by cross-breeding of LPL and ApoE knockout mice. Methods The model was developed by crossbred, backcross and intercross of the 2 lines of knockout mice, and rescued by muscular expression of a LPL beneficial mutant in homozygous neonates. The genotype was determined by PCR and confirmed by blood lipid assay. Results The LPL and ApoE double knockout mice could be rescued by muscular expression of LPL. Plasma TG and TC levels in adult LPLO/ApoEO mice were increased 76 and 14 fold, respectively, compared with wild-type littermates. Conclusion hypercholesterolemic/hypertriglyceridemic mouse model has been developed successfully.

    • Establishment of a Rabbit Model of Cardiopulmonary Bypass Flushed without Blood

      Abstract:Objective To establish a rabbit model of the cardiopulmonary bypass(CPB) flushed without blood.Method Thirty New Zealand rabbits were included in this experiment.The CPB procedure was established via right subclavian artery and right atrium cannulation.The blood-gas parameters were examined before and after CPB.Results The rabbit cardiopulmonary bypass model was established successfully.The results of arterial blood gas analysis were satisfactory.No hemorrhage and operation death occurred in this experiment.Conclusion A rabbit model of cardiopulmonary bypass flushed without blood has been established and showed to be feasible for related pathophysiological research work.

    • Use of 3R in the Anti-tumor Drug Screening

      Abstract:Objective To investigate the possibility to use 3R(replacement,reduction,refinement) method in anti-tumor drug screening.Methods(1) The KM mice were inoculated with S-(180),H-(22) and EAC tumors to establish "three-tumors in one" model;(2) 1/3 KM mice were inoculated with S-(180),1/3 KM mice were inoculated with H-(22),the others were inoculated with EAC,to establish "one-tumor in one" model.The "three-tumors in one" model and the "one-tumor in one" model mice were infused with the anti-tumor drug per os,to observe the effect on the ratio of tumor inhibition and immune function.Results(1)The tumor growed slower in the "three-tumors in one" model mice than in "one-tumor in one" model,but there was no influence on the judgment of the drug's tumor-inhibition function.(2) The "three-tumors in one" model was very close to the "one-tumor in one "model in immunological parameter such as thymus,spleen,DTH,phagocytic and clearing indices.Conclusion The "three-tumors in one" model can be used to tumor inhibition and immune function experiments of the anti-tumor drug screening.

      Abstract:Objective To set up a type 2 diabetes mellitus SD rat model.Methods SD rats were fed on a diet enriched in fat and fructose for 4 weeks to induce insulin resistance.The rats were injected intrapertoneally with a low-dose streptozocin(STZ) once(30 mg/kg) to induce hyperglycemia after 2 weeks.Results Insulin sensitivity index was decreased significantly in control group and hypertension was seen after rats were fed on a diet enriched in fat and fructose.There were hyperglycemia,fast blood insulin increased and lower insulin sensitivity in rat models after STZ injection.Conclusion The experiment test provided an ideal type 2 diabetes mellitus SD rats model.

      Abstract:Objective To observe the effects of a Chinese medicine "pneumonia mixture"on immunological,antibiotic and anti-inflammatory function in mice.Methods Carbon particles clearance test and serum assessment in vivo of mice were used to observe the effects of this product on the specific and nonspecific immunologic function in mice.The tube dilution method was used to test the MBC(minimum bactericidal concentration) to common pathogens in mice.Capillary permeability and granuloma formation were used to assess acute and chronic inflammatory responses.Results Pneumonia mixture can strengthen phagocytic function in mice.It can improve the serum half-hemolytic content(HC-(50)) and leukocyte count in immunologically incompetent mice.It can also raise T lymphocyte transformation rate.This product has antibacterial effect to pathogens such as Staphylococcus aureus.It can reduce capillary permeability and granuloma formation in mice.Conclusion This product has antibacterial and anti-inflammatory effects and can improve immunologic function of organisms.

      Abstract:目的观察不同浓度肿瘤腹水接种后,Wistar大鼠腹水产生的情况与其本身免疫功能的关系。方法24只大鼠被接种不同浓度的Walker-256肿瘤腹水0.3ml,另外12只大鼠腹腔注入生理盐水0.3ml作为对照组,8d后观察各组大鼠产生腹水的数量及细胞免疫及体液免疫功能各指标。结果高浓度腹水接种后产生腹水的大鼠只数少于稀释后接种的大鼠只数,各组免疫指标均具有统计学意义,P〈0.05,其中高浓度腹水接种组免疫功能最好,稀释后接种的大鼠免疫功能最差。结论不适当地增加含肿瘤腹水的浓度并不能增加产生腹水的可能性,相反确有可能激活大鼠本身的细胞和免疫功能消灭腹水内肿瘤细胞,使得产生腹水的可能性降低。

      Abstract:Objective To provide useful reference data on the standardization of utility and control procedure of plastic film isolators. Methods To explore a useful and appropriate procedure of assessment of internal environment in isolators according to the national standards. Results The measurement results demonstrated that those parameters can well satisfy the demands of evaluation of the function and feasibility of the plastic film isolators in accordance with the national standards. Conclusion The measurement procedure described in this study may well be used in evaluation of plastic film isolators.

    • Experience of Anesthesia Management on Auxiliary Liver Transplantation in Dogs

      Abstract:Objective To study the characteristic issues of anesthesia management on auxiliary liver transplantation. Methods 26 healthy dogs were operated to establish an auxiliary liver transplantation model under general anesthesia and trachea intubation. Mean arterial pressure and heart rate were monitored by external jugular vein and common carotid artery cannulation. Arterial blood samples were taken, before and 15 minutes after aorta blocking and 15 minutes after aorta opening, for blood gas analysis. Results No anesthetic death occurred among all the 13 cases during operation period. The apparent hemodynamic fluctuation of recipient dogs was observed during the period of abdominal aorta blocking and opening. Metabolic acidosis also took place during this period. Apparent rise of blood potassium was detected during reperfusion period. Conclusion In order to improve the survival rate, it is important to actively maintain the stability of hemodynamics, acid-base balance and correct electrolyte disturbance.

      Abstract:Magnesium is involved in antioxidant defense mechanisms in the body.Magnesium-deficient animals are more susceptible to free radical injury,supporting the hypothesis that magnesium deficiency reduces the threshold of antioxidant capacity.Inadequate intake of this nutrient has been associated with some diseases,where pathogenic role of free radicals is suggested.This article details the progress in research on influences of magnesium in the body's antioxidant system.
