Abstract:目的通过测定不同年龄阶段实验恒河猴的免疫球蛋白IgGI、gMI、gA和补体C3、C4在血清中的含量,建立部分免疫学正常参考值,并进行年龄组间的比较分析。方法人工饲养健康恒河猴分三个年龄组,每组20只,共60只,经后肢静脉采血,制备血清,检测IgGI、gMI、gA、C3和C4。结果青年组、成年组和老年组免疫球蛋白IgG测定结果分别为9.12±1.89 g/L1、2.36±1.57 g/L和13.10±1.82 g/L,IgM分别为1.01±0.24 g/L、1.46±0.44 g/L和1.25±0.34 g/L,IgA分别为0.26±0.10 g/L、0.39±0.12 g/L和0.34±0.10 g/L,C3分别为1.20±0.21 g/L0、.84±0.10 g/L和0.83±0.27 g/L,C4分别为0.22±0.05 g/L、0.17±0.03 g/L和0.16±0.06 g/L。结论青年组IgGI、gM和IgA含量极显著低于成年组和老年组(P<0.01),而青年组补体C3、C4含量极显著高于成年组和老年组(P<0.01),且各项测定结果在人类正常参考值范围内,IgGI、gM和IgA随年龄的变化情况与人类极为相似。

Abstract:Objective To establish an optimal model of medulla-suppressed anemia mice for curative observation. Methods 80 normal BALB/c mice were distributed randomly into 10 groups. The medulla haematogenous effect was compared among the normal group, the irradiation group, the injection cyclophosphamide group, the irradiation and injection cyclophosphamide combation group and the irradiation, cyclophosphamide and chloramphenicol combation group. Results Compared with normal group, the hemogram and bone marrow cell in other groups all decreased singificantly. Through analyzing death rate and amount of whole blood cell, effect of the irradiation and cyclophosphamide combation group was the most remarkable. Conclusion The irradiation and cyclophosphamide combation group is the best model for suppressing hematopoiesis of bone marrow in anemia mice.

Abstract:目的观察灵芝合剂(GJLM)对脂多糖(LPS)诱导大鼠星形胶质细胞表达组织因子(TF)的影响。方法培养Wistar乳鼠星形胶质细胞,用抗神经胶质纤维酸性蛋白单克隆抗体确认星形胶质细胞。细胞冻融液TF活性检测采用一期血浆复钙时间法。结果与对照组相比,LPS使星形胶质细胞表达TF明显增加(591±33 mU/mL vs382±25 mU/mL,n=12,P<0.01),并呈剂量依赖性(r=0.993);GJLM能明显地抑制LPS诱导星形胶质细胞表达TF(108±19 mU/mL vs 591±33 mU/mL,n=12,P<0.01)。结论LPS促进星形胶质细胞表达TF,而GJLM能抑制LPS诱导星形胶质细胞表达TF。

Abstract:目的探讨奶牛附红细胞体对兔的感染情况。方法用奶牛附红细胞体及被鼠转接的奶牛附红细胞体,腹腔接种连续7d静注免疫抑制剂(地塞米松)的新西兰兔,采用血液涂片染色镜检的方式,观察附红细胞体在兔体内的出现及消长规律。结果在使用免疫抑制剂的情况下,奶牛附红细胞体经小鼠转接后,可以造成兔感染,并呈现一定的规律性。附红细胞体在兔体内的首现期、高峰期、消失期分别为接种后的第5天,第6—7天和第9天。结论附红细胞体的宿主特异性可被打破。

Abstract:Objective To investigate the cell compatibility of materials for dermis and to provide data for tissue engineering skin.Methods Four scaffolds which including collagen sponge,gelatin sponge, Poly lactic acid_co_glycolic acid(PLGA) and poly 3_hydroxybutyric acid_co_3_hydroxyvaleric acid(PHBV) were prepared and cocultured with fibroblast or ES_derived epidermal stem cells in vitro.The cell compatibility was estimated through the adhesion rate and proliferation rate to fibroblast and the adhesion rate to ES_derived epidermal stem cells.Result Fibroblast and ES_derived epidermal stem cells all could adhere and survive in the four porous scaffolds and the fibroblast could proliferate,and the proliferation rate were higher in collagen sponge,PHBV and PLGA than in gelatin sponge.Conclusion The cell compatiblility is good in the four dermal scaffolds and suitable for constructure the new tissue engineering skin.

Abstract:目的建立一种与引起人体酒精性肝损伤发生机制类似的小鼠动物模型,并研究五味子酒精提取物对其影响及作用。方法ICR小鼠随机分为正常对照组,实验Ⅰ组,实验Ⅱ组,实验Ⅲ组和实验Ⅳ组,用含酒精的复合液体食物和细菌酯多糖(LPS)制备小鼠酒精性肝损伤模型。实验Ⅲ组和实验Ⅳ组分别在饲料中添加0.5%和1.5%五味子酒精提取物,饲喂8周后,实验Ⅱ、Ⅲ、Ⅳ组于尾静脉注射1 mg/Kg的LPS,6 h后测定血清ALT,AST,GGT含量,并取肝脏组织做病理学检查。结果混合酒精液体饲料联合LPS成功制作了小鼠酒精性肝损伤模型,ALT显著升高(16.3±5.2 U/L vs 52.6±18.7 U/L);五味子酒精提取物能显著地抑制血清ALT的升高(P<0.05);肝组织病理切片显示局部肝细胞变性,注射LPS后细胞核增大、核浓染,其余未见明显改变。结论酒精联合LPS建立了小鼠酒精性肝损伤模型,适用于酒精性肝损伤防治药物的研究;五味子酒精提取物具有降低酒精性肝损伤作用。

Abstract:Objective To analyse SNP(single nucleotide polymorphism) loci of the domestic inbred mouse gene by the lately established method(single_tube bi_directional allele specific amplification(SB_ASA)).Method Four domestic inbred mouse strains were detected by 16 SNP loci with the method of SB_ASA,and sequencing was designed to validate the reliability of the method.Results SB_ASA was successfully used to type 4 inbred mouse strains for 16 SNP loci,and the results were completely consistent with sequencing.Conclusion SB_ASA could be used to exactly inspect genotype of the domestic inbred mouse.

Abstract:Objective To study the effect of high voltage electric field(HVEF)on growth of Wistar rat and to discuss its biological mechanism.Methods 36 adult Wistar rats of 8_weeks_old age were randomly divided into three groups: control group A,treated group B and treated group C.Each group has 6 males and 6 females.The two treated groups were exposed to different kinds of HVEF.After 12 weeks'experiment,the growth indexes(such as the curve of weight changes,measurements of body conformation,the coefficients of main visceras)of rat were measured.Results The body mass of rat in treated group B was heavier than that of control group A(P < 0.05).The coefficients of liver,kidney and brain of rat in treated group B were lower than that in control group A(P < 0.05).The coefficient of liver of treated group C was lower than control group A(P < 0.05)Conclusion Steady HVEF may prominently increase Wistar rats' body weight and improve their growth rate.Pulsed HVEF of ultra_low frequency has little effect on rat.

Abstract:Objective To measure the indexes of biological characteristics of organ weight and organ coefficient of SPF BALB/c nude mice.Methods 38 SPF BALB/c nude mice(19 males and 19 females) were chosen,and the body weight and 12 organs' weight were determined,then the organ coefficients were counted.Results Between female and male nude mice,the kidney weight and brain weight showed significant difference(P<0.01) and the difference of weight,heart,liver,adrenal gland weight was obvious(P<0.05),the coefficient of brain,adrenal gland had remarkably significant difference(P<0.01) and the coefficient of kidney was obvious(P<0.05).Linear analysis showed that there were obvious linear relations between body mass with heart,liver,kidney,brain,adrenal gland and testicle in male mice;there were linear relations between body weight and liver,kidney,and stomach in female mice.Conclusion Differences existed in weight,heart weight,liver weight,kidney weight,brain weight,adrenal gland weight,coefficients of kidney,brain,and adrenal gland between female and male nude mice,and there had linear relationships between body weight and organs.

Abstract:Objective To compare the skin of BAMA miniature pig with human's.Methods\ Skin samples from BAMA miniature pigs and human were excised and the morphology of the skin was examined by light microscope and immunohistochemical method.Resemblance and disparity between BAMA miniature pig and human was found and described.Results\ The layer,the layer thickness,the dermal-epidermal interface,the cyto-distribution,the structure of extracellular matrix were similar between BAMA miniature pig skin and those of human.Conclusion\ BAMA miniature pigs can be used as models of wound repair and drug permeation,it also can be used as materials to prepare dermal substitute for wound repair.

Abstract:目的建立和优化重组人淋巴毒素α(rhLT-α)工程菌高密度发酵方法以及目的蛋白的复性和纯化方法,得到高纯度的重组人淋巴毒素α。方法高密度发酵rhLT-α工程菌,超声破碎细胞提取包涵体,经过复性后,用DEAE Sepharose FF,Phenyl Sepharose FF(LS)进行分离纯化。结果重组人淋巴毒素α以包涵体形式表达,经过破碎离心收集包涵体,洗涤后包涵体纯度达到85%以上。复性后,经两步纯化,最终目的蛋白纯度达98%以上。活性达6×108U/mg以上。结论建立了高效简便的淋巴毒素生产工艺,为淋巴毒素的规模生产打下了基础。

Abstract:Objective To found one model of chronic myocardial infarction in rats. Methods The Wistar rats were anaesthetized by 10% hydro-chloro-aldehyde, then intubated through oral tracheal and performed left anterior thoracotomy between the region of third and fourth intercostal, finally ligated the left anterior descending coronary artery. Results The survival rate was 80.8% beyond 3 weeks. Conclusion This operation is easy to perform and has high success, and also has little damage to animals.

Abstract:Objective To compare the efficacy and usefulness of domestic and imported biochemical marker kits and to determine if the domestic product deserves popularization for genetic monitoring.Method\ 13 biochemical markers(ADH,etc) were assessed using the GB/T 14927.1-2001 biochemical genetic monitoring methods.Result\ All tests demonstrated apparent simplicity,stability and accuracy of the domestic test kits.Conclusion\ The domestic biochemical markers kit is easy to operate, stable in use and convenient for transport,can well fulfill the demands of genetic quality monitoring,and deserves practical application.

Abstract:Objective To compare three methods for inspection of ectoparasites on laboratory animals.Methods\ Samples were taken by animal sacrifice,pulling up the animal's hair and scraping the animal's skin.Results\ The infectious rate of mite in mice from four laboratory animal center was 75.5%,90.0%,73.3% and 85%,respectively,which was higher than those obtained by the methods of pulling up the animals' hair(19.8%,27.3%,10.0%,75.0%) or scraping the animals' skin(19.8%,60.6%,40.0%,65.0%).The infectious rate of louse in Guinea pigs and mite in New Zealand rabbits was all 100% revealed by sacrifice method.Conclusion\ Sacrifice method is better than pulling up hair and scraping skin methods,but combination of those two or three methods is the best way to monitor ectoparasites on laboratory animals.

Abstract:Mongolian gerbils is widely used as lab animal. It has many advantages, such as strong anti-drought ability, unique biological properties and so on.
