    • The Influence of Nilestriol on Endometrium in Ovariectomized Rats

      Abstract:Objective To investigate the influence of nilestriol on endometria of ovariectomized rats. Methods Thirty-six female 7-month old Sprague-Dawley rats were divided randomly into sham-operation group(12 rats) and model group(24 rats).Bilateral ovariectomy was performed on the rats of model group.The rats of sham-operation group were only sham operated.Two months after operation the model group was divided randomly into two subgroups: model group and nilestriol group.Nilestriol group was treated with nilestriol;model group and sham-operation group received saline.After two months of the treatment all rats were killed and the endometria were removed and processed for histopathological examination.Result In model group(8 rats) the endometria were of proliferative phase or atrophy phase.In sham-operation group the ecndometria were of proliferative phase(5 rats) and secretory phase(3 rats).In nilestriol group all of the endometria were of secretory phase(8 rats).Conclusion Nilestriol can induce a secretory phase endometria in ovariectomized rats.Because those results are in apparent contrary to the current endocrinology theory and are different from the reports of previous animal experiments,we decided to make this report and believe it deserves further studies.

      Abstract:Objective To investigate the histocompatibility of materials for dermis and to provide data for tissue engineering skin construction.Methods Five scaffolds,collagen sponge,gelatin sponge,Poly(lactic acid-co-glycolic acid)(PLGA),poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid)(PHBV) and composite collagen gel were subcutaneously implanted into rats,respectively.The morphological changes and degradation process were observed.Results The tissue reactions induced by composite collagen gel and collagen sponge were slightly with reparative connective tissue proliferation,by PLGA and PHBV were moderately with lymphocyte and macrophage infiltration,and by gelatin sponge was similar to that of PLGA and PHBV.Conclusion\ These 5 scaffolds were histocompatible and degradable material to promote fibroblast ingrowth and vascularization,and were suitable for construction of tissues engineering skin.

    • Comparison of Polycystic Ovary-Like Syndrome Models in Rats Established by Subcutaneous Injection of Testosterone Propionate or HCG

      Abstract:Objective Two polycystic-like ovary syndrome models in rats induced by testosterone or HCG were compared.Methods In the model 1,Female Sprague-Dawley rats at age of 9 days were given subcutaneous injection of testosterone propionate.In the model 2,Female Sprague-Dawley rats at age of 24 days were given subcutaneous implant of levonorgestrel.3 days later HCG was subcutaneously injected to these rats for 9 days.Then the ovulation,morphology of ovary,blood serum levels of testosterone(T),estradiol(E-2),follicular stimulating hormone(FSH),luteinzing hormone(LH),fasting plasma glucose(FPG),fasting insulin(FINS) in the two models were assyed.Results Rats of both two models showed polycystic ovary-like syndrome manifestation.But the ovary of rats in model 1 was smaller than that of controls,and bigger in model 2.Serum levels of T,FPG,FINS increased in two models.Serum levels of E-2 increased,FSH and LH decreased in model 1.Serum levels of FSH and E-2 decreased,LH increased in model 2.Conclusion The two polycystic ovary-like syndrome rat models manifested with PCO,hypertestosteronemia and hyperinsulinemia.Those rats showed similar changes in human PCOS,and can be used as model animals for certain rat experimental research.

    • Inhibitory Effect of Honeybee Pollen Polysaccharide on S180 Sarcoma Growth in Mice

      Abstract:目的通过对小鼠腋下移植肿瘤S180后给荷瘤小鼠灌胃花粉多糖液,以观察花粉多糖液对肿瘤的抑制作用。方法将60只荷瘤小鼠随机分为环磷酰胺组(50mg/kg)、阴性对照组、花粉多糖实验组(200、400、600、800mg/kg4个剂量),连续灌胃给花粉多糖液10d。观察:①小鼠一般状况②抑瘤率③吞噬指数。结果800mg/kg花粉多糖组可明显降低瘤体重量,抑瘤率明显提高,达43.6%,与对照组比较差异有显著性(P〈0.05),并呈剂量一效应关系。同时,吞噬指数与肿瘤对照组比较具有极显著性(P〈0.01)。结论花粉多糖液达到一定浓度时可抑制肿瘤和提高小鼠腹腔巨噬细胞的吞噬作用。

    • Microsatellite DNA Monitoring Cultivation of Recombinant Inbred Strain of Mice

      Abstract:Objective In order to promote the cultivation and establishment of new inbred mice, the heredity genes of baby mice were monitored by microsatellites DNA and selectively breeding in this experiment. Methods DNA polymorphism in baby mice between inbred C57 and BALB/c mice was assessed by microsatellites DNA with PCR amplification. The inbred mice were bred using high and moderate similarity coefficient of female C57 mice of Fo . Results There were 14 polymorphism loci for F2 mice, and rate of homo-gene was 53.3 % . There were 29 homo-gene loci in F,o mice, and rate of homo-gene loci was 96.7 % . The similarity coefficient of mice in the sequential generations was gradually increasing by 3% - 10% from generation to generation. The fact that the mice of F,o generation are inbred mice, was verified and proved by skin transplantation. Conclusion A new method of fast cultivation of inbred mice has been established, therefore, much improved the traditional inbred cultivation techniques. It may considerably facilitate researches on blood relationship and genetic distance in animals in future.

    • Anesthetic Effects of Sumianxin in Combination with Pentobarbital in Dog Experiments

      Abstract:Objective To investigate the anesthetic effect of sumianxin in combination with pentobarbital in dog experiments. Methods Sixty healthy dogs were included in the present study, mules and females together. The anesthetics were administered intramuscularly. Results The anethesia induction time was 6 minutes in average. The anesthesia time lasted at least 270 minutes. Although sumianxin anethesia revealed well response for sedation, analgesia, as well as muscular relaxation, the combined use of pentobarbital in a half of its recommended dosage made a better anesthetization showing prolonged and stable anesthesia, with quick and smooth resuscitation. Conclusion Sumianxin combined with pentobarbital anethesia appears to be a satisfactory method for dogs during prolonged operation.

      Abstract:目的观察丙泊酚麻醉对家兔血液学、血液生化、血气及血流动力学指标的影响。方法1%丙泊酚注射液与5%葡萄糖溶液以1:4比例稀释配比,6号头皮针兔耳缘静脉穿刺,以20滴/min的速度维持静滴,15min时,抽取兔耳缘静脉血测血液学指标14项、血生化学指标22项,抽取兔耳中动脉血测血气指标14项。结果(1)血液学指标:血红蛋白(Hb)、红细胞计数(RBC)较麻醉前降低(P〈0.01),红细胞比积(HCT),平均血红蛋白量(MCH)、平均血红蛋白浓度(MCHC)较麻醉前有所上升(P〈0.01),其余指标差异不显著(P〉0.05);(2)血生化指标:天冬氨酸转氨酶(AST)、总蛋白(11P)、白蛋白(Alb)、球蛋白(Glb)、谷酰转肽酶(ALP)、胆碱酯酶(CHE)、肌酸激酶(CK)、肌酸激酶同工酶(CK—MB)、葡萄糖(GLU)、钙(Ca)、钠(Na)均较麻醉前降低(P〈0.01,P〈0.05);(3)血气指标:PCO:较麻醉前明显升高(P〈0.01),pH、钾(K)、HCT、碳酸氢根(HCO3-)、氧合血红蛋白(THb)均较麻醉前降低(P〈0.01)。结论丙泊酚麻醉家兔对其血液学、血生化学、血气指标、血流动力学均有不同程度的影响。

    • Measurement and Analysis of Hematological and Blood Biochemical Indices in Bred Rhesus Monkeys

      Abstract:Objective To investigate the hematological and blood biochemical parameters of rhesus monkeys raised in different geographic regions. Methods Twenty-two hematological and nineteen blood biochemical indices of rhesus monkey bred in Hainan and Guangxi were measured using an automatic blood cell analyzer and automatic blood biochemical analyzer. Statistical comparisons of the results were made between these two breeds of monkeys of the same gender and age. Results The reference range values of Cr, ALB, IBIL were higher in adult Guangxi monkeys compared to those in Hainan monkeys. EOS, EOS%, MCV, MONO% , MCH were higher in female Hainan monkeys compared to those of Guangxi monkeys. MCV and NEU were higher in juvenile Guangxi monkeys than those in Hainan monkeys. MONO was higher, MCV, NEU were lower in adult Guangxi monkeys compared to those in Hainan monkeys. Conclusion Except the age and gender, the conditions in different geographic regions significantly affect blood parameter value in rhesus monkeys.

    • Ultrastructural Changes of the Lung in Rats Suffering from Acute Respiratory Distress Syndrome (ARDS)

      Abstract:Objective An electron microscopic study was carried out to examine the pulmonary changes of rats suffering from acute respiratory distress syndrome(ARDS).Methods ARDS animal disease model was developed by means of paraquat(PQ) toxicant exposure.The pulmonary tissue was examined by electron microscopy.Results The pulmonary alveolar wall of normal rats was thin and intact.Lamellar bodies in the type II pneumocytes did not show any sign of evacuation.Pathological changes caused by pneumonic damage of pulmonary tissue in ARDS rats were observed.Conclusion Various pathological changes caused by ARDS pulmonary damage can be observed under the transmission electron microscope.The evacuation of substance from lamellar bodies in the type II pneumocytes can be seen as a characteristic pathological change caused by ARDS.Such lamellar body alteration in type II pneumocytes perhaps provides an important basis for the judgement of ARDS attack.

    • Comparison of RAPD Maps of Gerbil (Meriones unguiculatus) and Laboratory Rats and Mice

      Abstract:Objective A comparison of RAPD maps of gerbils and laboratory rats and mice was made in order to explore a new method for distinguishing different strains of laboratory animals.Methods DNA was extracted from the tail tissues from different strains of animals(gerbil,BALB/c,C57BL/6,KM,F344,BN,SD).Six pairs of primers were used to amplify their DNA.Results The amplified band patterns were apparently different in the products of 5 primers(p1,p2,p3,p4 and p6).Conclusion Our results indicate that it is feasible to distinguish different strains of laboratory animals by RAPD method.

    • Pharmacological Effects of 5 Chinese Herbal Medicines and Their Composite on Canine Parvovirus

      Abstract:目的对山茶叶、轮叶党参、卵叶芍药、苦味西葫芦、狼毒大戟5种中草药的提取物进行了体外抗犬细小病毒(CPV)筛选,筛选出抗病毒效果较好的山茶叶,并由山茶叶、黄芪等组成复方,并对该复方进行了抗病毒效果评价与药理作用研究。方法采用体外细胞培养和整体实验。结果研究表明山茶叶抗CPV作用较强,其对CPV的抑制率为55.24%,由山茶叶组成复方对CPV的抑制率为63.04%,抗病毒效果明显优于单方的山茶叶和药物对照黄连,对CPV50%抑制浓度(IC50)为1:587.5。并对大肠杆菌、沙门氏菌、金黄色葡萄球菌有抑制作用,可增强机体免疫力。结论提示本复方有较强抗病毒作用。

    • Effects of Cardioplegic Preconditioning on Ischemia/Reperfusion Injury of Isolated Rat Heart

      Abstract:Objective To study the influence of cardioplegic preconditioning on the contractile function of isolated inchemic/repperfused rat heart.Methods The Langendorff model of isolated perfused rat heart was used to study the effects of cardioplegic preconditioning on LVEDP,DP, dp/dt,-dp/dt,CF,protein contents and LDH activity before and after ischemia/reperfusion.Results Compared with the control group,cardioplegic preconditioning benefited the recovery of cardiac heamodynamics.It significantly delayed the onset of ischemic contraction.During the reperfusion period,it improved the recovery of contractile function and minimized the leakage of intracellular protein and lactate dehydrogenase.Conclusion cardioplegic pre-conditioning has protective effects against ischemia/reperfusion injury,and hence,might be a promising measure in clinical use.

      Abstract:Objective Through analyzing the monitoring results of several laboratory animal facilities to discuss more appropriate parameters of scientific and actual environmental indicators.Methods\ The relative environmental indicators were tested according to the National Standards for Laboratory Animal Environment and Facility(GB 14925-2001).Results\ The results showed the air exchange rate,the air borne bacteria,temperature and relative humidity,noise,pressure gradient,work-illumination were consistent with the National Standards.However, regards some parameters,there are significant differences among different places within certain same facilities.The air cleanliness and pressure gradient were often key obstacles to meet the requirement of the national standard for barrier system.Conclusion\ To promote standardization of laboratory animal environment and facility in China,it suggests that standards of design and construction for laboratory animal facilities should be established as soon as possible,to reduce the difference between facilities or at different places in the same facility.The objective value,most suitable value and permit deviation should be taken into account when the national standard is revised in the future.

    • PTCA Wire-Assisted Retrograde Endotracheal Intubation in Mice

      Abstract:Objective To establish a new method of endotracheal intubation in small animals.Methods Fourty C57BL/6 mice were included in this experiment.A wire of percutaneous transluminal coronary angioplasty(PTCA) was retrogradely inserted into the trachea and pushed into the oral cavity.Guided by the PTCA wire,a tracheal cannula was advanced into the trachea.Results Endotracheal intubation succeeded in all mice without severe complications.Conclusion APTCA wire-guided endotracheal intubation technique in mice has been established.This method is simple,safe,effective and with only mild damages in comparison with other existing tracheal intubation procedures.

    • Surgical Dissection of Dorsal Root Ganglions and Sympathetic Ganglions in Living Rats

      Abstract:Objective To describe a practical and reliable surgical procedure to dissect dorsal root ganglions and sympathetic ganglions from living rats. Methods Sixty two male Wistar rats were anesthetized by intraperitoneal injection of pentobarbital. When stripping dorsal root ganglions, the rat was in pronation. The paravertebral muscles were detached, and after careful laminectomy the intervertebral foramen was opened, thus the dorsal root ganglion can be exposed and dissected. When stripping sympathetic ganglions, the rat was in supine. After opening the right thoracic cavity, the sympathetic trunk should be found, then to expose and dissect the sympathetic ganglions. Results Operation to dissect dorsal root ganglions was performed on 41 rats. The cell culture of ganglion cells succeeded in all 41 cases. Operation to dissect sympathetic ganglions was carried out on 21 rats. The cell culture of ganglion ceils was successful in 19 cases and failed in 2 cases because of contaimination during cell culture. Conclusion The above described surgical procedures are useful and reliable to obtain dorsal root ganglions and sympathetic ganglions from living rats, with well kept viability and activity of the ganglion ceils.

      Abstract:The family of ErbB tyrosine kinase receptors can transmit signals derived from growth factor of epidermal growth factor family.There are four members in the ErbB tyrosine kinase receptor family: ErbB1,ErbB2/HER2/neu,ErbB3 and ErbB4.ErbBs play an essential role in the development of embryonic heart and maintaining cardiac function of adult heart.Conditional ErbB2 mutant mice provide an animal model for the major side effect,cardiomyopathy and heart failure, which can be observed in some patients undergoing chemotherapy with Trastuzumab and has revealed the important role of ErbB2 in normal heart function and ErbB2 function as an essential co-receptor of ErbB3 and ErbB4.Furthermore,ErbB2/4 heteromers transmit neuregulin-1 signals in the developing and adult heart.ErbB receptors are known regulators of many signaling cascades,such as those involving ras/mitogen-activated protein kinase,phosphatidylinositol-3 kinase /Akt,phospholipase C and calcium-dependent kinase.

      Abstract:Along with the conversion of medicine research pattern, the model research for replacing animals experiment has become an important aspect of toxicology development. Presently, the substitute methods for cosmetics toxicity test were investigated from monolayer cell culture, organ culture and chicken embryo culture run into artificial organ reconstruct in 3- dimensions. A good few of products have been provided merchandizing. Numbers of methods has been validating by relevant institution and application by EU and USA. Therefore, exploiting new replacement products and methods for cosmetics safety evaluation is not only the bulwark technique problem that inspection and quarantine must be replying but also the requirement of development of laboratory animal sciences.

    • Analysis of Correlation between Hyperlipidemia and Atherogenesis in the Gerbil

      Abstract:Objective To analyze the correlation between hyperlipidemia and atherogenesis in gerbils. Methods Fourteen gerbils in the experimental group were fed with a diet containing 1% cholesterol, 10% lard and 10% yolk powder. Twelve gerbils in the control group were fed with normal chow. Serum total cholesterol, triglyeerides, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol of gerbils in the two groups were measured at the 0, 7th, 30 th, 60th, 90th and 120th day. Finally all the animals were sacrificed and examined by histopathology. Results Serum TC, LDL-c and HDL-c of gerbils in experimental group were much higher than those in the control group during the 120 days, with increased serum LDL-e concentration in particular. Fatty liver, hepatoeirrhosis, splenomegalia and aortic atherosclerosis were observed in the experimental group. Conclusion Aortic atherogenesis in gerbils can be promoted by hyperlipidemia. The gerbil might be used as a suitable laboratory animal for fatty liver research.
