Abstract:目的观察大鼠重复给与携带肝细胞生长因子基因的重组腺病毒(recombinant adenovirus carrying hepatocyte growth factor gene,Ad-HGF)后,血清中抗腺病毒抗体的出现时间及消失规律。方法大鼠给与不同剂量的Ad-HGF共14次,利用病毒中和反应(细胞病变效应及绿色荧光方法)观察血清中抗腺病毒抗体。结果手术及正常对照组和实验组动物给药7次前血清处理的细胞均出现明显细胞病变效应及强荧光。之后各剂量组动物血清均对腺病毒有不同程度的中和作用,停药后13周到14周抗体水平很低或已不存在。同时观察到动物性别及给药剂量大小无差异。结论大鼠肌注Ad-HGF7次后,动物血清中可检测到抗腺病毒抗体,停药后13周或14周抗体基本消失,此结果对Ad-HGF的实际应用具有指导意义。

Abstract:目的观察重组人组织型纤溶酶原激活剂缺失变体(reteplase)对犬冠状动脉血栓的溶栓作用。方法实验用杂种犬,设溶剂对照组r、eteplase 100×104、50×104和25×104IU/kg 4组。手术分离冠状动脉左旋支(LCX),以血管内膜粗糙 电刺激法形成冠状动脉左旋支血栓,检测静脉注射不同剂量reteplase后对冠状动脉左旋支血流量、栓塞再通率、心肌梗死范围、伤口失血量和末梢出血时间的影响。结果静脉注射reteplase可使栓塞的冠状动脉血管很快出现再通,血流量显著增加,心肌梗塞范围明显缩小,明显增加伤口出血量,延长末梢出血时间,rete-plase各剂量组与溶剂对照组比较均有显著差别(P<0.001)。结论reteplase对犬冠状动脉血栓具有显著的溶栓作用。

Abstract:Objective To investigate the expression of GLAST and GLT-1 in rats with epilepsy.Methods Glutamate transporter GLAST and GLT-1 genes expression were detected by real-time fluorescent quantitative PCR assay with TaqMan probe in different time after epileptic seizure.Results The expression amount of GLAST and GLT-1 in cerebrum were obviously lower than that of the control in different time after epileptic seizure(P < 0.01).Furthermore,the amount of expression had evidently difference in cerebrum in different time after epileptic seizure(P < 0.01).The expression amount of GLAST in cerebellum was not significantly different in different time after epileptic seizure(P> 0.05),but they were obviously lower than that in the control(P<0.01).However,GLT-1 was not expressed in cerebellum.Conclusion The expression amount decrease of GLAST and GLT-1 may be one of the factors of epileptic seizure.

Abstract:目的观察运行中的独立通气笼盒(Individually Ventilated Cages,即IVC)在大鼠饲养过程中的温度、湿度变化及饲养只数与氨浓度的关系,指导科学饲养与管理。方法每天记录运行中的IVC的温度、湿度变化,用大气采样机和气体吸收管进行采样,按GB14925-2001方法测定。结果全年温度在(22.3±1.9)℃,相对湿度为(70.1±14.9)%,氨浓度随动物只数增加饲养天数增多而升高,有的超标严重,未达屏障环境国家标准。结论大鼠体重在(290±6)g时,每笼饲养不超过3只,且2 d更换一次垫料为宜。

Abstract:Objective To investigate the influence of burn injury with artificial seawater immersion on kidney pathological changes in experimental rabbits.Methods The healthy rabbits were randomly divided into control group(n = 4),simple burn injury group(n = 10,immediately studied after burn injury) and seawater immersion group(n=10,the rabbits were immersed in the seawater for 3 hours after burn injury.) Light and electron microscopes were employed to examine the pathological changes of kidney.Immunohistochemistry was also performed.Results By light microscopy,we found the dilation and hyperemia of glomerular capillaries,or swelling of endothelial cells.Glomerular cells increased and capillaries condensed.Hydrolic swelling in the epithelial cells of proximal convoluted tubules,hyaline degeneration and vacuolation were observed.The epithelial cells of some distal convoluted tubules disappeared and protein casts were found in the cavities.By electronmicroscopy,the swelling of endothelial cells,hypertrophy of mesangial cells and enlargement of mitochondria were found.Deformation and confluence of foot processes were also observed.Some mitochondria of the epithelial cells of renal tubules demonstrated vacuolation.Cytoplasm of some cells condensed.Hyperemia was found in the kidney mesenchyme.Conclusion The pathological changes of kidney appear to be associated with burn injury with artificial seawater immersion in experimental rabbits.

Abstract:目的本研究通过给与五指山小型猪高脂饲料,旨在诱发小型猪的高脂血症。方法将30头小型猪随机分成3组:①对照组(n=6),喂正常猪饲料;②1.5%胆固醇组(n=12),饲喂含1.5%胆固醇、10.0%的牛油、6.0%花生油和0.5%胆盐的饲料;③3%胆固醇组(n=12),饲喂含3.0%胆固醇、10.0%的牛油、6.0%花生油和0.5%胆盐饲料。高脂饮食6个月。每月测定体重和血脂(TG、TC、HDL-C、LDL-C)。结果和结论五指山小型猪对高脂饮食反应敏感,高脂饮食一个月内,高脂饮食组TC、HDL-C和LDL-C从极低水平迅速升高,随后逐月缓慢升高。至高脂饮食4个月时,TC达到15.5~17.7mmol/L,LDL-C达到12~13mmol/L,HDL-C达到4.45~5.76mmol/L,与基线相比,TC升高约10倍,LDL-C升高约15倍,HDL-C升高约7倍,4个月后变化趋于稳定,而对照组没有变化,TG没有明显变化趋势,形成了典型的高胆固醇血症。1.5%胆固醇组和3%胆固醇组血脂指标差异不明显(P〉0.05),提示1.5%的胆固醇含量就足以获得高脂血症模型。

Abstract:目的为了便于观察荷瘤实验动物血清唾液酸的变化情况,也鉴于国内文献尚无大鼠和小鼠血清正常参考值。方法采用测定血清唾液酸的定量方法。结果和结论188只大鼠血清平均唾液酸为(3.45±0.41)mmol/L,雌、雄大鼠平均唾液酸间无统计学差异(P>0.05),大鼠总体血清唾液酸频率服从正态分布,正常参考值范围为(2.65~4.25)mmol/L。100只小鼠平均血清唾液酸为(3.04±0.30)mmol/L,雌、雄小鼠平均唾液酸间有非常显著的统计学差异(P<0.001),小鼠总体血清唾液酸频率服从对数正态分布,其正常参考值范围为(2.44~3.64)mmol/L。

Abstract:Objective To observe the apoptosis in the livers of p21~(HBsAg/HBsAg) transgenic mice.Methods The liver of p21~(HBsAg/HBsAg) transgenic mice(6,12, 18,24 months) and corresponding p21~( / ) mice were assessed by flow cytometry.Result\ The apoptosis rate of the hepatocytes in p21~(HBsAg/HBsAg) transgenic mice was lower than that of wild mice of the same months.Conclusion\ HBsAg can inhibit apoptosis in liver cells of p21~(HBsAg/HBsAg) transgenic mice.

Abstract:Objective To observe the changes on content of nitric oxide in serum and expression on epidermal growth factor receptor of gastric mucosa in cold stimulated rats. Methods Twenty SD rats were divided into control, model group at random, NO in serum and EGFR in gastric mucosa of rats were tested. Results The gastric mucosa were differently damaged in model group, and there were significant difference on content of nitric oxide in serum and expression of epidermal growth factor receptor in gastric mucosa between two groups. Conclusion Cold simulation may result in the gastric mucosa damage in rats, which also affect the content of nitric oxide in serum and expression of epidermal growth factor receptor in gastric mucosa of rats.
